Peripheral blood mononuclear cells from patients with multiple myeloma, gastrointestinal tumors, and inflammatory bowel disease were analyzed for binding of various lectins. The results demonstrated that in most of the patients with multiple myeloma a significantly increased percentage of cells positive for Lotus tetragonolobus agglutinin (LTA), peanut agglutinin (PNA), soybean agglutinin (SBA), and wheat germ agglutinin (WGA), and a decreased number of Agaricus bisporus agglutinin (ABA) positive cells were present as compared to a normal control group. This could not be shown in malignant or inflammatory disorders of the gastrointestinal tract where only some patients exhibited an increased PNA and LTA binding, respectively. Patients with the systemic malignant disease differed from patients with solid localized tumors by a significantly altered number of ABA, LTA and SBA-positive peripheral blood mononuclear cells. Double fluorescence studies using monoclonal antibodies and lectins revealed that most of the cells expressing receptors for ABA had also receptors for OKT3, whereas most of the cells with receptors for LTA, PNA SBA, and WGA were found to be positive for OKM.  相似文献

目的建立高效液相色谱法测定荷叶与莲房中荷叶碱含量的方法。方法采用Agilent Eclipse XDBC18(4.6 mm×250 mm,5μm),以乙腈-水-三乙胺-冰醋酸(27∶70.6∶1.6∶0.8)为流动相;流速:1.0 mL·min-1;柱温:25℃;检测波长:270 nm。结果荷叶碱在3.0148.16μg·mL-1范围内线性良好(r=0.999 0),平均回收率为96.97%,RSD=0.99%(n=6)。结论该方法准确、简便、灵敏度高、重现性好,可用于荷叶与莲房中荷叶碱含量的测定。  相似文献

IntroductionVulvar cancer has a lower incidence in high income countries, but is rising, in part, due to the high life expectancy in these societies. Radical vulvectomy is still the standard treatment in initial stages. Wound dehiscence contitututes one of the most common postoperative complications.Presentation of caseA 76 year old patient with a squamous cell carcinoma of the vulva, FIGO staged, IIIb is presented. Radical vulvectomy and bilateral inguinal lymph node dissection with lotus petal flaps reconstruction are performed as the first treatment. Wound infection and dehiscence of lotus petal flaps was seen postoperatively. Initial management consisted in antibiotics administration and removing necrotic tissue from surgical wound. After this initial treatment, negative wound pressure therapy was applied for 37 days with good results.DiscussionWound dehiscence in radical vulvectomy remains the most frequent complication in the treatment of vulvar cancer. The treatment of this complications is still challenging for most gynecologic oncologist surgeons.ConclusionThe utilization of the negative wound pressure therapy could contribute to reduce hospitalization and the direct and indirect costs of these complications.  相似文献

目的研究莲房原花青素(Proanthocyanidins from Lotus Seedpod,LSPC)对食饵性高脂血症模型大鼠脂质过氧化和内皮素1(ET-1)的调节作用。方法Wistar大鼠随机分成4组,即对照组(C组)、高脂血症模型组(H组)、LSPC高剂量组(HD组)、LSPC低剂量组(LD组),除对照组饲普通饲料外,其余各组喂饲高脂饲料并进行干预实验比较。4周后检测大鼠血脂及血浆超歧物过氧化物酶(SOD)、丙二醛(MDA)、一氧化氮(N0)、ET-1的变化。结果与对照组相比较,H组血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL-C)水平显著升高;血浆中MDA和ET-1含量明显增高,而SOD活性和NO含量明显降低。HD组和LD组血浆SOD活性和NO含量显著升高,MDA和ET-1含量明显降低,但对血脂水平无显著性影响。结论LSPC具有心血管保护作用,其作用机制与抗氧化作用和改善血管内皮功能有关。  相似文献

目的探讨莲房原花青素(lotus seedpod procyanidins,LSPC)对D-半乳糖衰老小鼠脑组织抗氧化系统的影响。方法60只昆明小鼠随机均分为5组:对照组(A组)、衰老模型组(B组)、LSPC低剂量组(C组,15mg/ml)、LSPC中剂量组(D组,30mg/ml)和LSPC高剂量组(E组,60mg/ml)。采用D-半乳糖建立小鼠衰老模型,化学比色法检测脑组织中总一氧化氮合酶(NOS)和神经元型一氧化氮合酶(neuronal nitric oxide synthase,nNOS)活性,一氧化氮(nitric oxide,NO)含量。用免疫组化法检测nNOS在脑组织中的表达。结果与B组相比较,C、D、E组LSPC能明显减少NO含量(P〈0.05);D、E组LSPC不仅能明显降低总NOS和nNOS活性(P〈0.01,P〈0.05),且能减少nNOS在脑组织中的表达。结论LSPC对D-半乳糖所致脑衰老具有明显的保护作用。  相似文献

Fucose binding protein (FBP) from Lotus tetragonolobus seed was studied by fluorescence microscopy for its binding characteristics to various guinea pig peritoneal macrophage populations. Fluoresceinated FBP (FITC-FBP) was bound optimally at 22 degrees C in a punctate distribution and was internalized at 37 degrees C. Binding of FBP to macrophages was reversed specifically by the competitive sugar L-fucose, and not by D-fucose, L-rhamnose, or D-galactose. FBP was bound with greater frequency and intensity to 3-day oil-elicited peritoneal macrophages which are responsive to migration inhibition by FBP and migration inhibitory factor (MIF) than to resident or 7-day inflammatory macrophages which are unresponsive to activation by the same effectors. Competition for visual binding of FITC-FBP to macrophages was demonstrated by preincubation of cells with unlabeled FBP or MIF. Competition of FITC-FBP binding by MIF was reversed by L-fucose. These results indicate that FBP binds preferentially, with greater frequency and intensity, to macrophage subpopulations which are responsive to MIF than to MIF-refractory macrophages. The data further supports the existence of a common receptor site for MIF and FBP on the macrophage membrane which involves fucosyl determinants.  相似文献

Herbicide phosphinothricin (PPT) inhibits glutamine synthetase (GS), a key enzyme in nitrogen assimilation, thus causing ammonia accumulation, glutamine depletion and eventually plant death. However, the growth response of Lotus corniculatus L. plants immersed in solutions with a broad range of PPT concentrations is biphasic, with pronounced stimulating effect on biomass production at concentrations ≤ 50 μM and growth inhibition at higher concentrations. The growth stimulation at low PPT concentrations is a result of activation of chloroplastic isoform GS2, while the growth suppression is caused by inhibition of both cytosolic GS1 and GS2 at higher PPT concentrations. Since the results are obtained in cell-free system (e.g. protein extracts), to which the principles of homeostasis are not applicable, this PPT effect is an unambiguous example of direct stimulation hormesis. A detailed molecular mechanism of concentration-dependent interaction of both PPT and a related GS inhibitor, methionine sulfoximine, with GS holoenzymes is proposed. The mechanism is in concurrence with all experimental and literature data.  相似文献

《金瓶梅》是中国古代的著名小说,作者兰陵笑笑生于此首次提到了缅铃一词,不过自此以后,明清两代文人笔下的缅铃又都莫衷一是,极具臆想、讹传色彩。剖析《金瓶梅》内缅铃的来历和用处,却可以发现其背后隐藏着的宦官性事的内容。宦官,作为腐刑之下的刑余之人,自东汉以后就有婚娶之事,然其性事充满了非正常性。李瓶儿作为《金瓶梅》中的重要角色,其死亡背后亦有宦官性事的影响。  相似文献
