目的研究维生素K联合骨化三醇、钙尔奇D对男性骨质疏松的治疗效果及对骨代谢的影响。方法采用美国Hologic公司生产的Discovery WA型双能X线骨密度仪检测患者腰椎L_(1~4)骨密度(bone mineral density,BMD)与股骨颈骨密度。对男性骨质疏松患者采用固力康联合骨化三醇、碳酸钙D3进行治疗,连续治疗1年,治疗期间监测患者骨代谢变化。采用美国Thermo公司全自动酶标免疫分析仪检测患者血清骨代谢指标BGP、CTX-1、细胞因子IL-6、IL-10水平。结果治疗6个月后患者腰椎(L_(1~4)) BMD、股骨颈BMD开始增加,治疗12个月后患者腰椎(L_(1~4)) BMD、股骨颈BMD较治疗前增加明显。治疗后,血清骨形成指标BGP水平升高、细胞因子IL-10水平升高;血清骨吸收指标CTX-1水平、细胞因子IL-6水平降低。结论维生素K可提高患者腰椎与股骨颈骨密度,升高血清骨形成指标BGP、细胞因子IL-10水平,降低血清骨吸收指标CTX-1、细胞因子IL-6水平,维生素K治疗男性骨质疏松效果显著,是临床上治疗骨质疏松的重要策略之一。  相似文献

目的:通过观察傅青主火丹神方对带状疱疹患者T细胞亚群及相关细胞因子的影响,探讨其临床疗效及机制。方法:选择136例带状疱疹患者随机分为观察组和对照组,每组68例。对照组予常规西药治疗,观察组加用傅青主火丹神方治疗,观察两组患者临床疗效,并测定比较两组患者治疗前后T细胞亚群,白细胞介素-4(IL-4),白细胞介素-6(IL-6),肿瘤坏死因子-α(TNF-α)水平,实时荧光定量PCR测定血清信号转导和转录激活因子3(STAT3),磷酸化-STAT3(p-STAT3)mRNA水平。结果:观察组的总有效率为97.1%,明显高于对照组的83.8%(χ2=5.146,P0.05)。治疗后,两组患者的疼痛视觉(VAS)评分均较治疗前明显降低(P0.05),观察组较对照组下降更为明显(P0.05)。随访6个月,观察组的后遗神经痛发生率5.9%,明显低于对照组的26.5%(P0.05)。治疗后,两组患者CD3~+,CD4~+细胞比例均较治疗前明显升高(P0.05),观察组CD8+细胞明显降低(P0.05),CD4~+/CD8+明显升高(P0.05),且高于对照组(P0.05)。治疗后,两组患者IL-4,IL-6均较治疗前明显降低,TNF-α明显升高(P0.05),观察组变化更加明显(P0.05)。治疗前,两组患者的STAT3,pSTAT3在同一水平。治疗后,两组患者的STAT3,p-STAT3均较治疗前明显降低(P0.05),观察组降低更加明显(P0.05)。结论:在常规西药基础上,加用傅青主火丹神方治疗能更好的减轻临床症状,提高临床疗效,其机制可能通过调节机体免疫功能,减轻两面神激酶(Janus kinase,JAK)/STAT通路激活,减少炎症因子释放,并进一步减轻炎症反应有关。  相似文献

朱氏盆炎汤治疗湿热瘀结型盆腔炎性疾病后遗症疗效观察   总被引:1,自引:0,他引:1  
目的:观察朱氏盆炎汤治疗湿热瘀结型盆腔炎性疾病后遗症的临床疗效。方法:选取2013年1月—2014年12月就诊于本院门诊的湿热瘀结型慢性盆腔炎患者112例,随机分成治疗组60例和对照组52例。治疗组给予朱氏盆炎汤颗粒剂治疗,对照组给予妇科千金胶囊治疗。结果:两组证候疗效比较,治疗组有效率98.3%,对照组有效率57.7%,治疗组优于对照组(P0.05);两组体征疗效比较,治疗组有效率71.7%,对照组有效率40.4%,治疗组优于对照组(P0.05);治疗组治疗后体征评分、证候评分优于对照组(P0.05);治疗组治疗后血清TNF-α水平优于对照组(P0.05)。结论:朱氏盆炎汤治疗湿热瘀结型盆腔炎性疾病后遗症具有显著疗效。  相似文献

Strategies aiming at controlling and modulating inflammatory cues may offer therapeutic solutions for improving tendon regeneration. This study aims to investigate the modulatory effect of pulsed electromagnetic field (PEMF) on the inflammatory profile of human tendon-derived cells (hTDCs) after supplementation with interleukin-1β (IL-1β). IL-1β was used to artificially induce inflammatory cues associated with injured tendon environments. The PEMF effect was investigated varying the frequency (5 or 17 Hz), intensity (1.5, 4, or 5 mT), and duty-cycle (10% or 50%) parameters to which IL-1β-treated hTDCs were exposed to. A PEMF actuation with 4 mT, 5 Hz and a 50% duty cycle decreased the production of IL-6 and tumor necrosis factor-α (TNF-α), as well as the expression of TNFα, IL-6, IL-8, COX-2, MMP-1, MMP-2, and MMP-3, while IL-4, IL-10, and TIMP-1 expression increased. These results suggest that PEMF stimulation can modulate hTDCs response in an inflammatory environment holding therapeutic potential for tendon regenerative strategies. © 2019 Orthopaedic Research Society. Fracture repair is a complex process requiring heterotypic interactions between osteogenic cells and immune cells. Recent evidence indicates that macrophages are critically involved in fracture repair. Polarized macrophage populations differentially promote and regulate inflammation in other tissues, but little is known about the various macrophage subtypes and their signaling activities following a bone fracture. The authors hypothesized that classically activated (M1 subtype) and alternatively activated (M2 subtype) macrophages are active during the early repair process to initiate and regulate the inflammatory response. To test our hypothesis, bone marrow was collected from intact femurs (naïve group), contralateral and fractured femurs of mice on days 0, 1, 2, 4, and 7 postfracture. Macrophages were isolated from the bone marrow and macrophage subtypes were identified using flow cytometry with antibodies to F4/80, MHC II, CD86, CD11c, and CD40. Bone marrow cytokine levels were measured using xMAP. Flow cytometry revealed dynamic changes in M1 subtype (F4/80+/MHC II+/CD86+), M2 subtype (F4/80+/MHC II−/CD86−), and dendritic cell (DCs; MHCII+/CD11c+/CD40+) populations following fracture as compared to naïve controls. M1 subtype levels were correlated with IL-1α, IL-1ß, IL-2, IL-17, Eotaxin, and MCP-1, while DCs were correlated with IL-6, G-CSF, LIF, KC, and VEGF-A. The results indicate that M1 and M2 subtypes and DCs are recruited to the fracture site early during the repair process and consequently may work in tandem to regulate the inflammatory response required to recruit osteogenic cells needed for later stages of repair.  相似文献

目的检测妊娠期糖尿病(GDM)患者血清25-羟维生素D3[25-(OH)2-VitD3]水平,评估其水平与机体炎症因子、血脂的相关关系。方法选取2018年2~9月在中国医科大学航空总医院产检并确诊为GDM的孕妇178例作为GDM组,同期在本院进行产检且糖耐量检查正常的孕妇100例作为正常对照组。对比两组孕妇血清中25-(OH)2-VitD3、炎症因子及血脂指标水平的差异,采用Pearson检验评估GDM患者血清25-(OH)2-VitD3水平与机体炎症因子、血脂指标的相关关系。结果GDM组血清中25-(OH)2-VitD3的含量显著低于正常对照组(P<0.05)。GDM组血清中炎症因子白介素-1β(IL-1β)、IL-6、IL-18和肿瘤坏死因子α(TNF-α)水平显著高于正常对照组(P均<0.05);血脂指标总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、载脂蛋白B(Apo-B)和载脂蛋白CⅢ(Apo-CⅢ)的水平显著高于正常对照组,高密度脂蛋白胆固醇(HDL-C)、载脂蛋白A1(Apo-A1)的水平显著低于正常对照组(P均<0.05)。相关性分析发现,GDM孕妇血清中25-(OH)2-VitD3水平与炎症因子、血脂指标均相关(P均<0.05)。结论GDM患者血清25-(OH)2-VitD3水平明显降低,且其水平与机体炎症因子、血脂指标相关,这在GDM发生及病情评估中具有重要意义。  相似文献

Background: Millettia aboensis (Hook. F.) Baker (Fabaceae) is popular in ethnomedicine for its acclaimed efficacy in a number of disease conditions. This study evaluated the immunomodulatory effect of the leaf extract as a possible mechanism of its ethnomedicinal uses.

Methods: Humoral and cellular immune responses of Balb/c mice to tetanus toxoid and cyclophosphamide, respectively, were used to monitor immunomodulatory activities of the ethanol leaf extract and fractions of M. aboensis at 200, 300 and 400 mg/kg. Active (butanol) fraction of the extract was subjected to chromatographic purifications to isolate the active compound and the structure elucidated by a combination of 1D and 2D NMR and mass spectrometry. Stimulation of specific T-lymphocytes using intracellular cytokine staining technique was used to evaluate immune-enhancing activity of the isolated compound.

Results: The extract and fractions evoked increase in both humoral and cellular immunity. At 400 mg/kg of butanol fraction, the normalized mean secondary production of IgG1 and IgG2a antibodies were 9.0 and 7.7, respectively. Serum cytokine production by butanol fraction following secondary challenge with tetanus toxoid showed that IL-12, IL-17A and IFN-γ were expressed by 48.14, 41.37 and 38.22%, respectively. Structural elucidation of the active compound revealed presence of isomeric mixtures of quercetin-3-O-rutinoside and quercetin-3-O-robinobioside (Compound 1a/b). Compound 1a/b exhibited in vitro upregulation of specific CD4+ T-lymphocytes that were largely IFNγ releasing with up to 43.7% stimulation at 6.25 μg/mL compared to the baseline effect in DMSO vehicle control group.

Conclusion: M. aboensis expressed strong immune-enhancing properties, which may explain its ethnopharmacological use in disease management.  相似文献
