不同浓度的金属硫蛋白对离体豚鼠乳头肌缺氧、复氧电生理特性的影响 总被引:1,自引:0,他引:1
为探讨金属硫蛋白(MT)对豚鼠乳头肌缺血再灌注损伤所致心律失常的影响,利用标准玻璃微电极技术,采用缺氧及复氧豚鼠乳头肌模型,模拟体内缺血再灌注损伤,观察不同浓度MT对豚鼠乳头肌电生理特性的影响。结果显示低浓度的MT(0.002mmol/L)对正常及缺氧和复氧豚鼠乳头肌的动作电位(AP)有关参数及自律性均无影响;中等浓度的MT(0.02mmol/L)仅使正常乳头肌的AP复极达50%时程(APD50)缩短24%(P<0.05),但使缺氧乳头肌的AP复极达20%时程(APD20)、APD50和AP复极达90%时程(APD90)分别缩短68%、56%和43%(P均<0.01),并使静息电位(RP)、AP幅值(APA)和0相最大上升速率(Vmax)分别增加30%、30%和45%(P均<0.01);高浓度的MT(0.1mmol/L)使正常豚鼠乳头肌的APD20、APD50和APD90分别缩短57%、54%和50%(P均<0.01),并且RP、APA及Vmax分别下降22%(P<0.05)、28%(P<0.01)和29%(P<0.05),而使缺氧豚鼠乳头肌的APD20、APD50和APD90分别延长92%、78%和50%(P均<0.01),对RP、APA及Vmax无明显影响。在复氧期间,0.02mmol/L的MT可使自律性的发生率从77.8%降至55.6%(P<0.05);而0.1mmol/L的MT则使自律性的发生率从77.8%降至22.2%(P 相似文献

The effects of oestrogen administration on 1 h post‐exercise cardiac muscle myeloperoxidase (MPO) and calpain activities were determined in female rats. Rats were ovariectomized and implanted for 2 weeks with either oestrogen (25 mg 17‐oestradiol) or placebo pellets or left with ovaries intact. Rats were then run for 1 h at 21 m min–1, 12% grade, killed 1 h post‐exercise and cardiac muscle and blood samples were removed. Control animals from each group were killed without prior exercise. Serum oestrogen levels in the order of the highest to lowest were; ovariectomized oestrogen replaced rats > intact ovaries rats > ovariectomized placebo rats. Oestrogen induced significant (P < 0.05) elevations in cardiac MPO activity at rest and at 1 h post‐exercise in ovariectomized rats. No significant elevations in cardiac MPO activity were evident in placebo ovariectomized or normal ovary rats at rest or post‐exercise. Cardiac calpain activities were similar in all unexercised groups. Ovariectomized placebo and intact ovary rats had significantly (P < 0.05) elevated cardiac calpain activities 1 h post‐exercise while calpain activity was not significantly elevated in hearts from ovariectomized oestrogen rats. These results demonstrate that oestrogen supplementation in ovariectomized rats induces elevations in cardiac muscle MPO activities at rest and at 1 h post‐exercise. This is opposite to the effect of oestrogen in post‐exercise skeletal muscle and implies a greater neutrophil infiltration into cardiac muscle caused by oestrogen. This effect cannot be explained by changes in 1 h post‐exercise cardiac muscle calpain activity, the elevation of which was suppressed by oestrogen administration. Oestrogen influences cardiac calpain activity similarly to its effect in skeletal muscle. Abstract: This paper describes a device used to measure the isometric forces generated during electrical stimulation of the canine latissimus dorsi muscle in vivo with a preserved neurovascular supply. This device uses 2 strain gauge force sensors linked to a movable alignment frame to which the muscle is attached. The muscle length is controlled by the application of known weights to the system. 目的 :观察大鼠骨骼肌钝挫伤后 ,不同干预方法对骨骼肌修复过程中肌电信号的影响 ,研究肌电图在骨骼肌损伤愈合过程中的评价作用。方法 :10 8只雄性SD大鼠 ,利用自制的重物坠落打击装置造成右侧腓肠肌钝挫伤。随机分成 3组 ,即胰岛素样生长因子 - 1(IGF - 1)组、中药组和自然愈合组 ,分别于损伤局部注射IGF - 1、黄芪丹参注射液、不作处理。于伤后第 2、5、7、10、14、2 1、2 8、35、5 6天观察并比较双侧腓肠肌的肌电图变化 ,记录纤颤电位和正尖波等自发活动 ,分析复合肌肉动作电位 (CMAP)并测量其潜伏期和波幅 ,观察伤后CMAP的变化。结果 :(1)各组大鼠均于伤后第 2~ 5天出现纤颤电位和正尖波 ,约至第 7~ 10天达到高峰 ,然后逐渐减少。以IGF - 1组最早消失 ,中药组次之。 (2 )各组大鼠的伤侧肌电图CMAP中的潜伏期逐渐缩短 ,波幅逐渐增高。以IGF - 1组最快 ,中药组次之 ,自然愈合组最慢。 (3)伤后第 8周 ,IGF - 1组和中药组大鼠损伤侧CMAP波幅接近正常 ,自然愈合组则显著低于其它两组。结论 :(1)肌电图检测证实 ,IGF - 1和黄芪丹参注射液均具有促进骨骼肌损伤愈合的作用 ,但以IGF - 1的作用更明显。 (2 )损伤肌肉局部肌电信号的变化 ,可以用来评价损伤骨骼肌的修复进程和愈合质量 相似文献

The course of radiculopathy is sometimes associated with weakness and wasting of muscles. Very rarely in such cases, however, is hypertrophy of muscle fibres observed. Three cases are presented of sciatica with enlarged calves caused by hypertrophy of type 1 or types 1 and 2 muscle fibres. An electron microscopic study was done on abductor pollicis brevis muscles of 18 Rhesus monkeys after intramuscular injections of 0.75% bupivacaine, 2% mepivacaine, or 2% lidocaine + epinephrine. The muscles were examined for from 2 h to 28 days. Severe muscle fiber damage, consisting of breakdown of sarcolemma and myofibrils, was seen as early as 2 h. Phagocyte mediated fragmentation of the degenerating muscle fibers was at its peak during the third and fourth days. Myoblasts were abundant during the fourth day. Early myotubes appeared on the fifth and sixth days, and they matured during the second week. Satellite cells appeared alongside mature myotubes. Overall, the local anesthetic-induced breakdown and regeneration of skeletal muscle fibers in the monkey followed a course quite similar to that seen in the rat. 相似文献

Adhesion molecules were studied in regenerating skeletal muscle immunohistochemically and ultrastructurally after a standardized trauma. In normal muscle, extracellular matrix (ECM) protein tenascin was restricted to myotendinous junctions (MTJ), while the integrin beta 1-subunit was present also on the sarcolemma. After injury, tenascin increased on the outer surface of regenerating myofibers, where cellular fibronectin also accumulated. Later, tenascin concentrated at the tips of regenerating myofibers, where new MTJs were formed. The beta 1-subunit disappeared on necrotized myofibers and reappeared on regenerating fibers in a thicker layer. The regenerating myofibers were invested by a basal lamina, except for the growth cones at the distal ends, which were laminin-negative until the formation of MTJs occurred. These results indicate that regenerating muscle cells are attached to the ECM in a way that allows both growth of the muscle cells across the scar and their use before the regeneration is completed. 相似文献

Muscle fiber typing and in vitro contracture tests were performed in 59 patients investigated for susceptibility to malignant hyperthermia (MH). Eighteen patients were found to be susceptible to MH. There was no difference in age or fiber type distribution between MH susceptible and non-susceptible patients. No correlation was found between age and fiber type distribution. Separate analyses for each diagnostic group revealed no relationship between age or fiber type distribution and response to halothane or caffeine. When all caffeine results were pooled, however, there was a significant effect of age on the caffeine specific concentration (the concentration eliciting a contracture of 1 g), but not on the caffeine threshold concentration (the minimal concentration eliciting an increase in tension). It is concluded that age and fiber type distribution have no influence on MH diagnosis, if the protocol of the European MH Group for evaluation of susceptibility to MH is followed. 相似文献
