Leu-19 antigen, which seems to be identical with neural cell adhesion molecule (N-CAM), plays a major role in the innervation of muscle cells, and in adult muscle appears after denervation and during regeneration of muscle fibres, where it acts as part of a signalling system increasing the probability of re-innervation. This combined enzyme-histochemical and immunohistochemical study examined whether this signalling process was regulated in a uniform or differential pattern for type 1 and type 2 muscle fibres. The subscapular nerve of 18 rabbits was transsected with subsequent complete denervation of the supraspinatus muscle. Leu19 and N-CAM immunohistochemistry was performed 2 to 64 days after surgery. Whereas in normal muscle there are virtually no Leu-19/N-CAM positive muscle fibres; from day 2 after denervation an increasing proportion of fibres expressed Leu-19/N-CAM, prior to any neurogenic atrophy. In the early stage of denervation Leul9/N-CAM expression was confined to type 1 fibres. After 11 days nearly all fibres were Leul9/N-CAM positive irrespective of their fibre type. Sixty-four days after denervation type 1 fibres became Leu19/N-CAM negative, while atrophic type 2 fibres showed intensive staining. Thus, expression of Leu-19 antigenity is differently regulated in both fibre types.  相似文献

An unusual case of malignant peripheral nerve sheath tumor (MPNST) arising in the posterior mediastinum of a 59-year-old man is reported. Histopathologically, the tumor showed an admixture of a dense proliferation of small round cells resembling a primitive neuroectodermal tumor (PNET) and a pleomorphic spindle cell sarcomatous area. Abortive rosettes, primitive neural tube-like structures, and a few glandular structures were found in the small round cell area. Small round cells were immunoreactive for neural cell adhesion molecule and synaptophysin, but were not immunoreactive for MIC2 and neuron-specific enolase. Pleomorphic spindle cells were occasionally arranged in a storiform pattern and were diffusely immunoreactive for S-100 protein. The MPNST of small round cell type is distinguishable from PNET by its negative immunoreactivity for MIC2, and the present tumor is assumed to be derived from primitive neuroectodermal cells in the peripheral nerve capable of bidirectional (neuron and Schwann cell) differentiation.  相似文献

L1 is a representative of a family of carbohydrate neural cell adhesion molecules. The expression of L1 was studied during postnatal development of the rat pyramidal tract by immunohistology using polyclonal antibodies to L1 in spinal cord cervical intumescences. On postnatal day 1 (P1), L1 immunoreactivity was present in the entire dorsal funiculus, consisting of the ascending fasciculus gracilis and fasciculus cuneatus and the descending pyramidal tract. At that time the cervical pyramidal tract contains the first outgrowing corticospinal axons. At P4 both the fasciculus gracilis and the pyramidal tract are immunoreactive whereas the fasciculus cuneatus is negative. At P10 the pyramidal tract is intensely labelled whereas both ascending bundles are negatively stained. In the period between P4 and P10 the pyramidal tract is characterized by a massive outgrowth of corticospinal axons. During pyramidal tract myelination, between P10 and the end of the third postnatal week (P21), L1 immunoreactivity is progressively reduced. These observations suggest that L1 may play a prominent role in outgrowth, fasciculation and the onset of myelination of rat pyramidal tract axons. The differential L1 immunoreactivity of the pyramidal tract and the earlier developing ascending systems in rat dorsal funiculus indicate that this polyclonal antiserum is a useful differentiating marker for outgrowing fibre tracts.  相似文献

A new anti-p58 monoclonal antibody (mAb), termed CH-L, has been used to characterize a minor subset of T lymphocytes co-expressing p58 and CD3 molecules. In two-color immunofluorescence analysis, CH-L+CD3+ cells represented 0.5 to 6% of the peripheral blood lymphocytes (in 20 healthy donors). Clonal analysis showed that most CD3+CH-L+ T cell clones expressed the CD8+4? T cell receptor (TcR) α/β+ phenotype, while only a few were CD8?4+ TcR α/β+, CD8?4? TcR α/β+ or CD8?4? TcR γ/δ+. Western blot analysis indicated that the CH-L mAb identifies the same 56-58-kDa diffuse band in both T and natural killer cell (NK) clones. A minority of T cell clones also expressed other NK-related markers such as CD16, CD56 and CD94 and two clones also reacted with the anti-p58 mAb EB6. Interestingly, most clones displayed cytolytic activity in an anti-CD3 mAb-triggered redirected killing assay against the Fcδ receptor+ P815 target cells and NK-like activity against K562 and Raji cells. In contrast, the IGROV-1 ovarian carcinoma cell line was resistant to cytolysis by all of these clones. Since p58 molecules have previously been shown to exert regulatory functions on NK-mediated lysis, we investigated whether anti-p58 mAb could also influence cytotoxicity mediated by CD3+p58+ T lymphocytes. Lysis of P815 target cells, triggered by anti-CD3 mAb, could be inhibited by anti-p58 mAb in 8 out of 12 cytolytic clones tested, while 4 clones were not inhibited. In addition, anti-p58 mAb enhanced the cytolytic activity of 3 clones against IGROV-1 and of 4 other clones against Raji target cells. Taken together, these data indicate that p58+ T cells express heterogeneous phenotypes and different forms of TcR and, in most instances, display cytolytic functions. Perhaps more importantly, the p58 molecule appears to modulate the cytolytic activity triggered via the CD3/TcR complex.  相似文献

目的构建人OX40-Fc基因的真核表达质粒,并表达有生物学活性的纯化人OX40-Fc融合蛋白。方法从活化的人T细胞cDNA文库中以PCR方法扩增OX40膜外区cDNA编码基因,并导入真核表达载体pcDNA3中。测序证实后,进行酶切并与人IgG1Fc基因一起装入真核表达载体pcDNA3中,采用脂质体法稳定转染COS-7细胞,用夹心ELISA检测其表达情况,经蛋白A亲和纯化,用SDS-PAGE与Western blotting进行鉴定。检测其对活化后Jurkat细胞增殖的抑制活性。结果测序证实构建的OX40膜外区与OX40-Fc cDNA阅读框完整,连接部位序列正确;ELISA证实OX40-Fc蛋白的表达;SDS-PAGE与Western blotting证实其为OX40-Fc蛋白。增殖实验证明其对活化后的Jurkat细胞有抑制增殖作用。结论成功构建了人OX40-Fc真核表达载体,并使有生物学活性的OX40-Fc融合蛋白在COS-7细胞上获得了稳定表达。为进一步研究该蛋白在自身免疫性疾病治疗中的作用及调节免疫自稳机制中的地位奠定了基础。  相似文献

为在纳米尺度对 NMDA受体蛋白分子进行神经细胞膜表面原位定位和探讨原子力显微镜在生物单分子操纵和调控中的应用 ,本研究应用原子力显微镜分别对分布在云母表面的膜 NMDA受体蛋白分子标记物抗 NMDAR1Ig G-葡萄球菌蛋白 A-胶体金复合物分子和结合标记物分子后的神经元膜进行扫描 ,三维形貌测定 ,通过颗粒度分析结果 ,明确标记物分子的特征性三维形貌 ,对比确定经过免疫胶体金结合后的 NMDA受体蛋白单分子在神经元膜表面的定位。结果显示 ,空白云母表面标记物分子为分散均匀的平均粒径为 49nm的球形颗粒 ,在神经元膜表面结合 NMDA目的受体蛋白分子后 ,免疫复合物分子呈现出粒径为 5 3 nm的散在分布球形或短棒状颗粒 ,长径约为宽径的 2倍 ,长轴截面可见典型的双峰三维结构。上述结果表明 ,NMDA受体蛋白单分子可以结合 1个或 1个以上的胶体金标记物分子 ;原子力显微镜可以在纳米尺度对神经元膜 NMDA受体蛋白进行标记和其免疫复合物的三维形貌测定。胶体金颗粒标记 ,原子力显微镜测定是免疫细胞化学新方法。  相似文献

目的研究中国汉族人群中细胞间黏附分子1(intercellular adhesion moleculel,ICAM1)基因K469E多态性与冠状动脉粥样硬化性心脏病(简称冠心病)的关联。方法采用聚合酶链反应.限制性片段长度多态性方法检测了173例冠心病患者和141名对照的ICAM1基因K469E基因型和等位基因的分布。结果基因型频率符合Hardy-Weinberg平衡。冠心病组的KK基因型的频率显著高于对照组(64.2%比48.9%,P〈0.01),同样,冠心病组K等位基因的频率显著高于对照组(79.2%比69.9%,P〈0.01)。经Logistic回归分析排除年龄,性别,和冠心病其它危险因素的影响后,KK纯合子患冠心病的危险性是KE和EE基因型的2.35倍(95%CI:1.03-5.36,P〈0.05)。结论ICAM1基因K469E多态性与中国汉族人冠心病的危险性相关,其中K等位基因可能是冠心病的遗传危险因素。  相似文献

Expression of CD4-like molecule on vitelline membrane of murine eggs was demonstrated by indirect immunofluorescence (IIF) test and immunoprecipitation corresponding to the expression of major histocompatibility complex (MHC) class II molecule on murine sperm detected by immunoblotting. This molecule showed slightly larger size than that of the authentic CD4 molecule from T-cells on SDS-PAGE. This molecule was suggested to bind to MHC class II structure on sperm during fertilization because anti-CD4 monoclonal antibody (mAb) blocked in vitro fertilization (IVF). In addition, src-related tyrosine protein kinase (p56lck) was demonstrated in the inner vitelline membrane of eggs by means of IIF with anti-p56lck mAb and immune-complex kinase assay. 探讨与滋养层侵入有关的细胞外基质分子相关基因在先兆子痫胎盘中的表达,采用分别点样有220余种人细胞因子相关基因和人类激素相关基因cDNA片段的两款基因芯片,检测经过严格配伍的先兆子痫和正常胎盘组织的基因表达谱差异。结果显示:钙粘蛋白、胶原、整合素、选择蛋白等18种细胞外基质分子基因的表达在先兆子痫和正常胎盘组织间相差2倍以上,且全部表现为在先兆子痫胎盘中的表达增强。先兆子痫患者的胎盘组织中基质金属蛋白酶(MMP)-10、-13、-15和金属蛋白酶组织抑制因子(TIMP)-2、TIMP-3、纤溶酶原、纤溶酶原激活物等的表达均较正常者高。提示胎盘中细胞外基质分子及其降解酶基因表达异常可能与先兆子痫的病理发生关系密切。  相似文献
