To investigate the representation of motor sequence, we tested transfer effects in a motor sequence learning paradigm. We hypothesize that there are two sequence representations, effector independent and dependent. Further, we postulate that the effector independent representation is in visual/spatial coordinates, that the effector dependent representation is in motor coordinates, and that their time courses of acquisition during learning are different. Twelve subjects were tested in a modified 2x10 task. Subjects learned to press two keys (called a set) successively on a keypad in response to two lighted squares on a 3x3 display. The complete sequence to be learned was composed of ten such sets, called a hyperset. Training was given in the normal condition and sequence recall was assessed in the early, intermediate, and late stages in three conditions, normal, visual, and motor. In the visual condition, finger-keypad mapping was rotated 90 degrees while the keypad-display mapping was kept identical to normal. In the motor condition, the keypad-display mapping was also rotated 90 degrees, resulting in an identical finger-display mapping as in normal. Subjects formed two groups with each group using a different normal condition. One group learned the sequence in a standard keypad-hand setting and subsequently recalled the sequence using a rotated keypad-hand setting in the test conditions. The second group learned the sequence with a rotated keypad-hand setting and subsequently recalled the sequence with a standard keypad-hand setting in the test conditions. Response time (RT) and sequencing errors during recall were recorded. Although subjects committed more sequencing errors in both testing conditions, visual and motor, as compared to the normal condition, the errors were below chance level. Sequencing errors did not differ significantly between visual and motor conditions. Further, the sequence recall accuracy was over 70% even by the early stage when the subjects performed the sequence for the first time with the altered conditions, visual and motor. There were parallel improvements thereafter in all the conditions. These results of positive transfer of sequence knowledge across conditions that use dissimilar finger movements point to an effector independent sequence representation, possibly in visual/spatial coordinates. Initially the RTs were similar in the visual and the motor conditions, but with training RTs in the motor condition became significantly shorter than in the visual condition, as revealed by significant interaction for the testing stage and condition term in the repeated measures ANOVA. Moreover, using RTs for single key pressing in the three conditions as baseline indices, it was again observed that RTs in the visual and motor conditions were not significantly different in the early stage, but motor RTs became significantly shorter by the late testing stage. These results support the hypothesis that the motor condition benefits more than the visual because it uses identical effector movements to the normal condition. Further, these results argue for the existence of effector dependent sequence representation, in motor coordinates, which is acquired relatively slowly. The difference in the time course of learning of these two representations may account for the differential involvement of brain areas in early and late learning phases found in lesion and imaging studies. 相似文献

To investigate a potential role of osteopontin (OPN) in developing rat brain, the expression of OPN mRNA and protein in the developing rat brain relative to the distribution of brain macrophages was investigated using in situ hybridization and immunohistochemistry, and the phagocytic capability of OPN-expressing cells was accessed using rhodamine isothiocyanate (RhIc) as a tracer. OPN-expressing cells appeared from embryonic day 16. During the first week of postnatal life, OPN-labeled cells increased markedly, and peaked around P7, then declined and had completely disappeared by the end of the second postnatal week. The spatiotemporal distribution pattern of OPN mRNA closely matched that of OPN protein. Their morphology and localization were compared with those of cells expressing the established microglial marker OX-42 in adjacent sections, and double-labeling studies demonstrated that OPN was localized to the amoeboid microglia which stain with the lectin GSI-B4, another marker for microglia. Furthermore, OPN-labeled cells were confirmed to be active phagocytes emitting RhIc fluorescence indicating that the tracer into the brain tissues was engulfed by phagocytosis. Therefore, these results provide the first evidence that OPN is transiently expressed in active brain macrophages in the embryonic and early postnatal brain, and suggest that OPN may contribute to the migration and phagocytic function of brain macrophages in the developing brain.This work was supported by a Korea Research Foundation grant (KRF-2002-015-EP0106) 相似文献

Langerhans cell histiocytosis (LCH) has been thought to be a disorder of immune regulation, and increasingly, evidence showing
that the tissue damage in LCH involves lymphokines and pro-inflammatory cytokines is reported. We detected human cytomegalovirus
(HCMV)-DNA in LCH cells in the foci of LCH lesions by immunohistochemistry, in situ hybridization and PCR. HCMV was detected
in the nuclei and/or cytoplasm of LCH cells in 9 of 27 LCH cases by immunostaining. HCMV was probably an early antigen. In
situ hybridization revealed signals for HCMV-DNA only in the nuclei of LCH cells in 10 of the 27 LCH cases. PCR analysis was
performed in 20 of the LCH cases, and HCMV-DNA was detected in 7 of these. All 7 positive cases were also positive for HCMV
by ISH and IHC. These findings suggested that early phase infection or reactivation of HCMV occurred in the LCH lesions. HCMV
infection may be accompanied by impaired cytokine production. Our study also suggested a relationship between HCMV infection
and expression of TNFα. In tissues affected by LCH, dermatopathic lymphadenopathy or malignant fibrous histiocytoma and in
normal tissues no signals for Epstein-Barr virus-RNA were detected. These findings suggest that in some cases LCH is associated
with HCMV infection.

Decompression operation of the optic canal via the nasal path under endoscope is widely used, but it is both a challenging and controversial method. Unsatisfactory results were largely associated with otolaryngologists' limited understanding of the real anatomical situations of the optic canal before operation. To provide otolaryngologists with the real situations and data preoperation, multislice helical CT was used to reconstruct the images of the optic canal. Using multislice helical CT‐aided three‐dimensional reconstructive methods in combination with direct anatomic measurement, we dissected and analyzed the shape of the optic canal and its anatomic relationship with the adjoining structures in 40 intact postmortem skull samples. The In‐Space technique clearly showed the structure and the related region of the optic canal. The virtual endoscopy technique showed superbly the spatial appearance and topography of the inner optic canal and also gave the inner structure of the optic canal optically. There was no statistic difference in three‐dimensional reconstructive data with that obtained by anatomical measurements and thus can be used to directly instruct the clinic operation. These results demonstrate that a combined In‐Space technique with virtual endoscopy can accurately define the subtle structure and the related region of the optical canal. Infants with Ureaplasma urealyticum in the lower respiratory tract are at risk for chronic lung disease (CLD) or bronchopulmonary dysplasia (BPD) but causality has been difficult to prove. The goal of this study was to identify ureaplasma in human neonatal lung tissue using the in situ hybridization (ISH) procedure described in Part 1 (Exp. Mol. Pathol., in press) of this report. By correlating their presence with the histopathologic findings, it may be possible to provide further evidence of the pathogenicity of ureaplasmas and their association with BPD. Lung autopsy tissue from seven infants with positive cultures and seven infants with negative cultures for ureaplasma were included in the study. All culture-positive infants were positive for ureaplasma on ISH and all had histopathologic evidence of BPD. Two of the seven infants with negative cultures were positive for ureaplasma with ISH. Of interest, these two infants were also found to have BPD at autopsy. The other five infants with negative cultures were also negative for ureaplasma on ISH and had no evidence of BPD. This study correlates the presence of U. urealyticum by ISH with the finding of BPD on histopathologic evaluation and provides evidence that it has a role in the development of CLD. 相似文献

Intracellular pH (pHi) and viability of gastric surface cells of the rat stomach in response to luminal acidification, and the role of Na+/H+ exchange in maintaining pHi homeostasis were studied in vivo using a fluorescent microscopic technique. pHi was measured during superfusion with buffers of pH 1.2–7.4. When the pH of the superfusate was 7.4, baseline pHi was unchanged. Superfusion with pH 3 buffer rapidly decreased pHi to 6.7, with subsequent recovery to baseline pHi within 15 min despite continuing acid exposure. Superfusion with buffers of pH 1.7 and 1.2 decreased pHi continuously to below 6.2 with no recovery observed. Despite the relentless decline in pHi during superfusion with pH-1.2 and –1.7 solutions, over 75% of the surface cells were still viable, as measured by exclusion of the vital dye propidium iodide. We then examined the role of Na+/H+ exchange in the regulation of pHi. Superfusion with amiloride did not affect recovery of pHi from intracellular acidification induced by a NH4Cl prepulse. Exposure to the potent, lipophilic Na+/H+ exchange inhibitor 5-(N,N-hexaniethylene)-amiloride (HMA), either in the superfusate or by close arterial perfusion, decreased baseline pHi from 7.1 to 6.8. Close arterial perfusion of HMA additionally attenuated the recovery of pHi to baseline during superfusion with pH 3 buffer. We conclude that luminal protons permeate into the cytoplasm of gastric surface cells, where they are eliminated by an Na+/H+ exchanger, most probably localized to the basolateral membrane. 相似文献

Summary Experiments are described in which we have investigated the responsiveness to GnRH of pituitary glands obtained from ovariectomised rats in which LH surges were induced by steroid priming (a) by estrogen implants and (b) by estrogen/progesterone injections. These treatments generate LH surges late in the light phase. Stimulation with GnRH in vitro revealed a large increase in responsiveness coincident with the surge of LH. We suggest that this effect is mediated through a self-priming effect of GnRH and that this in vitro technique may be used to detect hypothalamic release of GnRH.Supported by grants MA-7131 (M. 目的 探讨18q21在人胰腺导管腺癌和慢性胰腺炎中杂合性缺失(LOH)的情况及其相关因素.方法 选择18q21上的位点RP11-729G3和RP11-850A17作为目的 片段,选择接近18号染色体着丝粒的位点RP11-621L6作为参照位点,利用细菌人工染色体(BAC)提取、纯化相应位点的DNA,用切口平移法分别标记生物素和地高辛后制成双色探针,应用荧光原位杂交技术(FISH)检测30例胰腺导管腺癌和10例慢性胰腺炎石蜡包埋组织切片中18q21 LOH情况,并收集、整理相应临床病理资料进行相关性分析.结果 30例胰腺导管腺癌中在RP11-729G3位点有25例(83.3%)有LOH,在RP11-850A17位点26例(86.6%)有LOH,其中25例在RP11-729G3和fuP11-850A17两个位点均有LOH,1例仅在RP11-850A17位点有LOH.10例慢性胰腺炎中均未发现18q21 LOH.经统计学分析发现,18q21 LOH在慢性胰腺炎和胰腺导管腺癌中差异有统计学意义(P<0.01),且RP11-729G3和RP11-850A17两个位点LOH有高度相关性(Phj系数=0.877),但和临床病理各因素间未发现明显相关.结论 18q21 LOH在胰腺导管腺癌中属于高频事件,并且位点RP11-729G3和RP11-850A17的LOH有高度相关性,可能在胰腺导管腺癌发生发展中起重要作用.在临床诊断中18q21LOH也能作为较特异的标记辅助诊断. 相似文献
