研究煤焦油组分时,为了防止组分相互之间的干扰,在测定前用几种不同极性的溶剂:石油醚;80%石油醚-20%甲苯;60%石油醚-40%甲苯;40%石滑醚-60%甲苯;20%石油醚-80%甲苯;甲苯;氯仿;50%氟仿-50%甲醇;甲醇进行洗提,从煤焦油中得到9个级分。然后用荧光法从9个级分中鉴定出28个组分:茶、苊、联苯、三甲基酚、菲、异喹啉、芴、对氨基酚、联苯胺、蒽醌、四氢萘、2,5-二甲酚、蒽、咔唑、间苯二酚、二乙基苯胺、喹啉、吲哚醌、1,3,5-三甲苯、1,2,4-三甲苯、萘醌、菲醌、窟、苾、氧芴、1,2-苯并蒽、2,3-苯并蒽、3,4-苯并苾。同时定量了氯仿和甲醇级分中氧芴、(艹屈)、联苯胺、苾、菲、蒽、咔唑、四氢蔡咔、四氢萘、苯醌、喹啉、8-羟基喹啉等12个组分的含量。 相似文献

目的 :探讨TrKA和p75神经营养因子受体在嗅神经母细胞瘤的表达及其临床意义。方法 :应用免疫组化和双荧光标记法对 10例鼻腔嗅神经母细胞瘤及 5例正常鼻腔嗅黏膜及 5例鼻咽癌组织进行TrKA和p75神经营养因子受体检测。结果 :免疫组化检测发现 10例鼻腔嗅神经母细胞瘤TrKA和p75蛋白均呈阳性表达 ,5例正常鼻腔嗅黏膜及 5例鼻咽癌组织均为阴性表达 ;双荧光标记显示所有肿瘤组织中TrKA和p75蛋白均在同一细胞表达。结论 :TrKA和p75神经营养因子受体的过度表达可能与嗅神经母细胞瘤的发生和发展有关 ;TrKA和p75神经营养因子受体检测可作为嗅神经母细胞瘤的一种辅助诊断方法 相似文献

The wheat--Thinopyrum amphiploid 'Agrotriticum # 3425' (AT 3425), which is highly resistant to Cephalosporium stripe, was identified to carry seven pairs of Thinopyrum chromosomes, three pairs of wheat--Thinopyrum translocated chromosomes and 18 pairs of wheat chromosomes. Fluorescence genomic in situ hybridization (FGISH), C-banding, sequential C-banding and FGISH, and denaturing polyacrylamide gel electrophoresis (SDS-PAGE) were used to characterize and identify the chromosomes. The Thinopyrum chromosomes in AT 3425 were designated as T1 through T7 based on their C-banding patterns. The FGISH and C-banding patterns of mitotic chromosomes in AT 3425 and meiotic chromosomes in the hybrid between AT 3425 and wheat cultivar 'Chinese Spring' (CS) revealed that wheat chromosomes 1D, 2B and 3D were involved in the three wheat-Thinopyrum chromosome translocations designated as (W-T)1, (W-T)2, and (W-T)3 respectively. The analysis of high-molecular-weight glutenin subunits in single seeds of AT 3425 confirmed the involvement of wheat chromosome 1D in the translocation (W-T)1. The designations 1DSuu.1DL-1TL, 2BSuu.2BL-2TL and 3DSuu.3DL-3TL were suggested for the wheat--Thinopyrum translocated chromosomes (W-T)1, (W-T)2 and (W-T)3 in AT 3425 respectively. 相似文献

用荧光偏振法测定17例慢性肺心病急性发作期患者及17名健康人红细胞膜流动性,用微孔泸膜法测定其中10例红细胞变形性。结果,肺心病患者及正常人红细胞膜微粘度分别为4.64±1.56及3.51±1.05η(p<0.05);红细胞滤速分别为384±136及233±97(p<0.01)。红细胞滤速与红细胞膜粘度间呈明显正相关(r=0.802,p<0.01)。结果提示,肺心病患者红细胞膜流动性下降可能对红细胞变形性下降起一定作用。 相似文献

The endogenous activity of the local renin-angiotensin system(RAS) and the anti-ischaemic properties of captopril were investigatedin electrically driven rabbit Langendorff hearts (constant pressure:70 cmH2O, Tyrode solution, Ca2+ 1.8 mmol.l1). Cumulativeconcentration-response curves showed no significant difference(P>0.05) between the reduction of the global coronary flow(CF) by exogenous angiotensin-I or angiotensin-II (EC50 = 1010mol.l1). It is concluded that the local RAS in isolatedrabbit hearts is highly sensitive, whereas its endogenous activityis very low due to very low endogenous angiotensin-I content.Myocardial ischaemia (MI) was induced by the occlusion of aleft coronary artery branch and MI was quantified from NADHsurface fluorescence photography. MI was significantly enlarged(+35%) (P <0.05) by exogenous angiotensin-I (6x109mol.l1). The reduction in CF and the increment in MIby angiotensin-I could be completely prevented by adding captoprilat a low concentration (106 mol.l1) to the perfusionbuffer. In the absence of exogenous angiotensin-I, captoprilalone (106 mol.l1) neither significantly enhancedCF (P >0.05), nor diminished MI (P >0.05), supportingthe finding of very low endogenous activity of tile local RASin this model. We, moreover, conclude that at a low concentration(106 mol.l1) captopril does not possess directcardioprotective properties independent of its ACE inhibitingaction. 相似文献

The chicken genome, like those of most avian species, contains numerous microchromosomes that cannot be distinguished by size
alone. Unique properties attributed to the microchromosomes include high GC content and gene density, and an enhanced recombination
rate. Previously, microchromosome GGA 17 was shown to align with the consensus genetic linkage group E41W17, and bacterial
artificial chromosome (BAC) clones containing E41W17 markers were isolated and assigned on the physical BAC map as well as
the recently assembled draft chicken genome sequence. For this study, these same BACS were utilized as probes for fluorescence
in-situ hybridization (FISH) to develop the GGA 17 cytogenetic map. Here we detail the chromosome order of ten BAC DNAs, thereby
deriving a cytogenetic map of GGA 17 that is simultaneously integrated with both the linkage map and genome sequence. The
location of the FISH probes together with the morphological appearance of the chromosome suggested that GGA 17 is an acrocentric
chromosome whose cytogenetic map orientation is reversed from that currently indicated by the linkage map and draft genome
sequence. The reversed orientation and the centromere location of GGA 17 were confirmed experimentally by dual-colour FISH
hybridization using terminal BACs and the centromere-specific CNM oligonucleotide as probes. An advantage of this cyto-genomic
approach is the improved alignment of the sequence and linkage maps with cytogenetic features such as the centromere, telomeres,

Human papilloma virus (HPV) type 16 infections of the genital tract are associated with the development of cervical cancer (CxCa) in women. HPV16-derived oncoproteins E6 and E7 are expressed constitutively in these lesions and might therefore be attractive candidates for T-cell-mediated adoptive immunotherapy. However, the low precursor frequency of HPV16E7-specific T cells in patients and healthy donors hampers routine isolation of these cells for adoptive transfer. To overcome this problem, we have isolated T cell receptor (TCR) genes from four different HPV16E7-specific healthy donor and patient-derived human cytotoxic T lymphocyte (CTL) clones. We examined whether genetic engineering of peripheral blood-derived CD8+ T cells in order to express HPV16E711-20-specific TCRs is feasible for adoptive transfer purposes. Reporter cells (Jurkat/MA) carrying a transgenic TCR were shown to bind relevant but not irrelevant tetramers. Moreover, these TCR-transgenic Jurkat/MA cells showed reactivity towards relevant target cells, indicating proper functional activity of the TCRs isolated from already available T cell clones. We next introduced an HPV16E711-20-specific TCR into blood-derived, CD8+ recipient T cells. Transgenic CTL clones stained positive for tetramers presenting the relevant HPV16E711-20 epitope and biological activity of the TCR in transduced CTL was confirmed by lytic activity and by interferon (IFN)-gamma secretion upon antigen-specific stimulation. Importantly, we show recognition of the endogenously processed and HLA-A2 presented HPV16E711-20 CTL epitope by A9-TCR-transgenic T cells. Collectively, our data indicate that HPV16E7 TCR gene transfer is feasible as an alternative strategy to generate human HPV16E7-specific T cells for the treatment of patients suffering from cervical cancer and other HPV16-induced malignancies. 相似文献
