CD34 cells from human umbilical cord blood were purified by Dynal beads M-450 CD34 immunoselection system and cultured in the presence of various cytokines alone or in combination, including stem cell factor (SCF), interleukin-6 (IL-6) and erythropoietin (EPO). The results revealed that: (D In methylcellulose culture, the plating efficiencies of purified cord blood CD34 cells were much different when stimulated by various cytokines. IL-6 alone had the lowest colo-ny yield, while the combination of SCF, IL-6 and EPO had the highest yield. ② In the suspension culture, IL-6 alone or IL-6 EPO had little expanding effect on cord blood CD34 celis, the other cytokine combinations could expand cord blood CD34 celis at different Ievels. Among them, the combination of SCF, IL-6 and EPO had the maximal expanding effect on cord blood CD34 celis, the number of progenitor celis peaked at day 21, about 29-fold increase and nucleated celis increased approximately 3676-fold at day 28. Aim . Molecular imaging studies with benzamide radioligands can reveal competition from endogenous binding at D2/3‐receptors in living brain. However, single photon emission computed tomography (SPECT) methods suffer from limited spatial resolution, and [11C]‐labeled ligands are only available at positron emission tomography (PET) research sites with cyclotron‐radiochemistry facilities, whereas [18F] can be transported, due to its longer physical half‐life. Therefore, we endeavored to characterize the vulnerabilities of the benzamide antagonist [18F]desmethoxyfallypride (DMFP) and its high‐affinity congener [18F]fallypride (FP) to competition from endogenous dopamine in living mouse brain. Methods . Groups of awake mice were pretreated with saline, amphetamine (10 mg/kg), or reserpine (5 mg/kg), followed by i.v. tracer injections. Mice were killed at 2.5–90 min (DMFP) or 2.5–180 min (FP) circulation times. Brains were dissected and regional radioactivity concentration measured by gamma counting. Other groups of mice were anesthetized for dynamic microPET recordings with DMFP or FP. Binding potentials (BPND) were calculated using cerebellum as reference region. Results . With 90‐min circulation, DMFP BPND in striatum was 2.4 by dissection and 2.2 by microPET, which showed a 62% decrease in response to amphetamine‐evoked dopamine release and a 33% increase after reserpine‐evoked dopamine depletion. With 120‐min circulation, FP BPND in striatum was 24.1 by dissection and 9.2 by microPET, which showed a 31% decrease in the amphetamine group, but no effect of reserpine. Dissection showed similar sensitivities for FP binding, but only a 29% amphetamine‐evoked reduction for DMFP. Conclusions . Relative to gold standard ex vivo results, microPET estimates of DMFP BPND were unbiased, whereas FP BPND in striatum was substantially underestimated. 目的:研究白饭树水提取物对肝纤维化(HF)大鼠血清学的影响。方法:采用皮下注射(SC)四氯化碳(CCl4)油溶液造成HF模型,以秋水仙碱(colchicine)为阳性对照,观察白饭树水提取物对血清丙氨酸转氨酶(ALT)、天门冬氨酸转氨酶(AST)、总蛋白(TP)、白蛋白(ALB)、球蛋白(GLO)、透明质酸(HA)、层粘连蛋白(LN)、Ⅲ型前胶原(PCⅢ)含量的影响。结果:白饭树水提取物能降低HF模型大鼠血清中ALT、AST含量,提升ALB含量,并且有效降低HF模型大鼠血清中HA、LN、PCⅢ的含量,高剂量组总体效果优于低剂量组。结论:白饭树水提取物对HF大鼠有保肝降酶、改善蛋白脂质、抗HF作用,值得深入研究。  相似文献

目的 测定鄂西北地区不同生长条件和品种柴胡中多糖的含量.方法采用蒽酮 硫酸比色法,以620 nm波长为检测波长,分别测定柴胡中可溶性多糖和粗多糖,计算总多糖含量.结果鄂西北地区野生竹叶柴胡多糖含量高于同地区种植柴胡,但低于山西产北柴胡和湖北南漳种植的三岛柴胡.结论不同生长条件的柴胡药材中柴胡多糖含量不同,但差异不明显;不同品种柴胡药材中的多糖含量有明显差异.  相似文献

目的:确定灵芝菌固体发酵青木香的发酵终点,为灵青菌质的制备工艺提供实验依据。方法:运用双向固体发酵技术,青木香与灵芝菌共同发酵后,定期取样,测定基质转化率,并分别采用反相高效液相色谱法和紫外分光光度法测定发酵产物灵青菌质中马兜铃酸Ⅰ和总马兜铃酸的含量。结果:基质转化率、样品中马兜铃酸Ⅰ和总马兜铃酸的含量随发酵时间呈现动态变化:发酵至第28天灵青菌质的基质转化率小幅下降,第28~35天相对稳定,此后又大幅下降;HPLC法测定结果表明,0~28 d马兜铃酸Ⅰ含量基本无变化,28 d后含量开始明显下降;UV法测定结果表明,0~28 d总马兜铃酸的含量保持下降的趋势,28~35 d含量基本保持恒定,35 d后开始上升。结论:灵芝发酵青木香药材的发酵终点基本可判定为第28~35天。双向固体发酵技术可有效地应用于青木香的减毒。  相似文献

Aim

In critically ill patients, reduced anti-FXa plasma activity following subcutaneous administration of enoxaparin or nadroparin has been described. In this study, we aimed to investigate the bioactivity of enoxaparin in critically ill patients and controls.

Methods

A prospective, controlled, open label study was performed on a medical intensive care unit (ICU) and a general medical ward. Fifteen ICU patients (male = 12, median age 52 years [IQR 40−65], with a median Simplified Acute Physiology Score of 30 [IQR 18−52]) and sex- and age-matched medical ward patients were included. The anti-FXa plasma activity was measured after a single subcutaneous dose of 40 mg enoxaparin. The thrombus size of a clot formed in an ex vivo perfusion chamber and endogenous thrombin potential (ETP) were measured.

Results

The anti-FXa plasma activity increased significantly after enoxaparin administration, with peak levels at 3 h after treatment, but was comparable between the ICU and medical ward groups (median 0.16 IU ml−1[IQR 0−0.22 IU ml−1]vs. 0.2 IU ml−1[IQR 0.15−0.27 IU ml−1], respectively, P= 0.13). The area under the anti-FXa activity curve from 0–12 h was similar between the groups (median 0.97 IU ml−1 h [IQR 0.59−2.1] and 1.48 IU ml−1 h1[IQR 0.83−1.62], P= 0.42 for the ICU group compared with the control group, respectively). The ETP was lower in the ICU group (P < 0.05) at baseline, but it was comparable at 3 h between the groups. Thrombus size decreased at 3 h compared with predose (P= 0.029) and was not different between the groups.

Conclusion

Similar bioactivity was achieved with a standard dose of subcutaneous enoxaparin in this selected cohort of ICU and general ward patients with normal renal function.  相似文献

目的:为研究和开发蝶形花科千斤拔属植物锥序千斤拔Flemingia paniculata Wall. ex Benth.提供部分生药学研究的理论依据。方法:采用性状鉴别、显微鉴别、理化鉴别的方法对锥序千斤拔进行生药学初研。结果:锥序千斤拔根横切面主要特征为:木栓层4~7列细胞组成;韧皮部分泌细胞较多;木质部导管多呈单个或数个相聚;薄壁细胞及射线细胞有的含淀粉粒,棕色物及草酸钙方晶。结论:为同属植物的鉴别提供了参考依据。  相似文献
