Objective To explore the feasibility of labeling and tracing in vitro goat bone marrow mesenchymal stem cells (BMSCs) by bromodeoxyuridine (BrdU) on the basis of investigation of its optimal concentration, incubating time and cytotoxicity. Methods A healthy goat, aged 10 months old, male, weighing 32 kg, was used in this study. Bone marrow was aspirated. BMSCs were isolated and cultured using the adherence method in vitro. The fourth passage of BMSCs (P4) were incubated with BrdU at 5, 10, 15, 20 μmol/L as 5, 10, 15, 20 μmol/L BMSC groups. Cells were not labeled by BrdU as negative control. The following parameters were measured: induction, differentiation and determination of goat BMSCs; the optimal mass concentration and incubation time of 5-BrdU labeling; cell positive rate at 12, 24, 48 and 72 hours in each group using immunofluoreseenee; the cell survival rate after various concentrations of BrdU ladling by trypan-blue exclusion. Results The morphology of the primary and passage goat BMSCs was fusiform in shape. Goat BMSCs could differentiate into osteoblasts and chondrocytes following induction. BMSC nucleus showed green fluorescence under fluorescence microscope after being labeled by BrdU. The mean labeling rate increased with the increase in the concentration and incubation time of BrdU, and reached to (93.32± 3.25)% after incubation in 15 μmol/L, BrdU for 48 hours. There were no significant differences between 15 μmol/L BrdU for 72 hours, 20 μmol/L BrdU for 48 hours and 72 hours (P > 0.05), or between the other groups or time points (P < 0.05). The labeling rate of the blank control group was 0. The cell survival rate was all above 90% (P > 0.05). Conclusions BrdU can be used as a labeling marker for goat BMSCs. When the concentration is 15 μmol/L and the incubation time is 48 hours, the optimal labeling effect can be achieved. Goat BMSCs labeled with BrdU is of high efficiency and safety. 相似文献

目的:评价自制不透X线标志物的临床应用价值,并确定正常人胃排空及胃肠道通过时间的正常值。方法:采用X线法对60例健康志愿者餐后1 h、2 h3、h的胃排空及24 h4、8 h肠道标志物排出情况进行观察。结果:正常人标准餐后3 h胃排空率为41.1±30.0%,24 h和48 h肠道标志物排出率分别为73.5±29.8%、99.0±20.9%。结论:自制的标志物具有良好的屏蔽功能;X线法胃排空及胃肠通过时间测定方法简单实用,值得临床推广。 相似文献

OBJECTIVES: To determine electrocardiogram (ECG) predictors of positive cardiac markers and short-term adverse cardiac events in an undifferentiated chest pain population presenting to emergency departments (EDs). The authors hypothesized that specific ECG findings, other than those previously identified in higher-risk populations, would be predictive of cardiac outcomes and positive cardiac markers. METHODS: This study used data from a prospectively collected, retrospectively analyzed Internet-based data registry of undifferentiated chest pain patients (i*trACS). Logistic regression modeling was performed to determine the ECG findings that were predictive of 1) positive cardiac markers and 2) short-term adverse cardiac events. RESULTS: ST-segment elevation (STE), ST-segment depression (STD), pathological Q-waves (PQW), and T-wave inversion were associated with increased odds of percutaneous coronary intervention or catheterization, myocardial infarction, or coronary artery bypass grafting. The odds of creatine kinase-MB (CK-MB) measuring positive were increased if STE, STD, or PQW were present [odds ratio (OR) 2.495, 2.582, and 1.295, respectively]. A right bundle branch block tended to decrease the odds of CK-MB measuring positive (OR 0.658). A similar pattern of results was observed for troponin I (OR 3.608 for STE, 3.72 for STD, 1.538 for PQW). Troponin T showed an increased odds of measuring positive if any of STE, STD, left bundle branch block, or T-wave inversion were evident (OR 2.313, 2.816, 1.80, and 1.449, respectively). CONCLUSIONS: Initial ECG criteria can be used to predict short-term cardiac outcomes and positive cardiac markers. These findings can be important aids in the risk-stratification and aggressive treatment regimens of chest pain patients presenting to EDs. 相似文献

目的评价血清肿瘤标记物联合检测对胃癌和大肠癌诊断和监测的价值.方法用PC-12多种肿瘤标志物蛋白芯片检测系统检测179例胃癌和大肠癌患者、82例胃和结直肠良性疾病患者及160例健康人血清中12种常见肿瘤标志物CA19-9、NSE、CEA、CA242、铁蛋白(Ferritin)、Beta-HCG、AFP、free-PSA、PSA、CA125、HGH、CA153的水平.结果肿瘤组的肿瘤标志物水平显著高于良性疾病组及健康组(P<0.01),其中CA19-9、CEA、CA242、CA125和CA153 5项在胃癌和大肠癌患者中水平较两对照组明显升高,差异有统计学意义(P<0.01).采用平行检测法,可以提高检测的敏感度(72.07%)和阴性预测值(79.25%);采用系列检测,可提高检测的特异度(92.15%)和阳性预测值(84.67%).肿瘤根治术后,肿瘤标记物水平显著下降.术后未降至正常者,复发或转移率为68.89%,而术后再升高者复发或转移率为77.78%.结论多种肿瘤标记物对胃癌和大肠癌的诊断、监测肿瘤复发和转移有一定的价值. 相似文献

目的探讨脑肿瘤干细胞(BTSCs)体外分化过程中的回逆现象,为研究其分化抑制机制奠定基础。方法利用CD133免疫磁珠筛选系统,从肿瘤组织中分离获得的CD133^+细胞(BTSCs)分成4组进行培养:(1)含10%胎牛血清(FCS);(2)10%FCS+丙戊酸钠注射液(VPA);(3)无FCS+生长因子;(4)无FCS+生长因子+VPA。取不同时间点上的细胞,相差显微镜观察其形态变化:流式细胞术检测与分化相关的标志物、细胞周期和DNA倍体变化;利用免疫激光共聚焦分析与分化相关标志物的共表达情况。结果无FCS条件下培养的BTSCs呈悬浮球状生长,高表达CD133和巢蛋白(nestin),不表达胶质纤维酸性蛋白(GFAP)和β-微管蛋白Ⅲ(β-TubulinⅢ)。G0/G1期细胞占大多数,G2/M期细胞接近0%,DNA都是异倍体,对VPA反应不敏感。含FCS培养的原本悬浮的细胞约4h开始贴壁。均呈圆形。此后逐渐向多形性分化,至7d时分化的细胞部分又返回至圆形。至10d-21d时,有的还能重新恢复球形,并呈悬浮生长。培养3d、7d、10d和21d时,CD133、nestin阳性细胞数先降后升,GFAP^+和β-TubulinⅢ^+细胞数始终处于较低水平。含FCS培养液中加入VPA。细胞形态上未见上述的回逆现象,CD133和nestin表达的先降后升现象消失,GFAP和β-TubulinⅢ在第7天以后表达明显升高,但极大部分细胞共表达nestin。而神经干细胞(NSCs)在含FCS培养至10d时,即以GFAP和β-TubulinⅢ表达为主,未见CD133^+细胞。此外,含血清培养时BTSCs仍以异倍体为主。含少量的G2/M期细胞,加VPA诱导后细胞周期和DNA倍体变化不明显。结论BTSCs在含血清条件下培养出现的多向分化表型不稳定,时有去分化所导致的回逆。加入诱导分化剂VPA培养,虽然能阻止回逆现象出现,并有代表星形胶质细胞和神经元标志物表达上升.但因其共表达nestin而仍属于未完全分化细胞,表明BTSCs分化始终处于受抑状态。 相似文献

This research is designed to evaluate a number of biological markers to estimate harmful exposure on coal miners from different mining regions in France and to relate the outcome to differences in prevalence of coal worker pneumoconiosis (CWP) between these regions. Eight epidemiological groups of active and ex-miners (smokers and non-smokers) have been selected in the French collieries (North, Lorraine and Provence) according to their occupational and pneumoconiotic status. The following biomarkers have been evaluated: cellularity of sputum, elementary analysis of particles in TEM/EDAX, plasma neutral metalloendo peptidase elastase type (NMEP), leucocyte elastase (HLE), fibronectin (FN) and elastin peptides. Pulmonary alveolitis, expressed by sputum cellularity, is different between active workers groups but not related to the general background of pneumoconiosis prevalence in the French collieries. In the plasma parameters, fibronectin, HLE and NMEP significantly increased in all groups of coal mine workers as compared to the control group, except for fibronectin parameter in Lorraine collierie. The degree of increase of these parameters allow us to discriminate the different groups and suggest that plasma FN, HLE and NMEP may be considered as biological markers of chronic inhalation of coal mine dust particles. The decrease of elastin peptides level in the Lorraine group alone suggests a specific alteration of elastin metabolism. These parameters were not related to the development of pneumoconiosis and its degree of severity. 相似文献

Dermal exposures of methanol were administered in a clinical study designed to compare several biological indicators. Four
subjects were exposed in five exposure sessions of varying length. In each session, a sequence of measurements of methanol
concentrations in blood, breath, and headspace samples of air at exposed and unexposed skin were collected before and after
dermal exposures. Skin headspace samples, collected in gas sampling bags, were designed to reflect equilibrium skin: air partitioning.
At exposed skin, headspace samples were highly elevated for at least 8 h following exposure, indicating the presence of a
methanol reservoir in skin. After exposure, methanol concentrations at exposed skin showed a rapid initial decline, then a
slower first-order decrease. Methanol concentrations were clearly detectable in headspace samples at unexposed skin. Substantial
transfer from exposed skin occurred due to mechanical contact and washing. When transfer was restricted, surface concentrations
at unexposed skin were similar to levels in breath and were strongly correlated to methanol concentrations in blood. While
results are preliminary due to the small sample sizes and several unresolved experimental issues, the simple, rapid, and noninvasive
skin headspace measurements appear useful as a biological exposure indicator that clearly shows the presence and site of a
dermal exposure, and measurements at unexposed skin reflect concentrations in blood.

Summary Analysis of hemoglobin adducts in blood samples is suitable for the biological monitoring of genotoxic chemicals. The method is specific because the compound to which the individual was exposed is identified. The sensitivity of the method depends on the analytical procedure applied, but is hardly limiting since large amounts of the protein can be obtained. The method provides not only information about the internal exposure to the environmental chemical, but also about the individual's capacity to generate ultimate genotoxic metabolites from it. Since macromolecular damage in blood cells is correlated to that in potential target tissues, this information is relevant to risk assessment, insofar as macromolecular damage produced by a specific chemical can be correlated with the development of tumors. 相似文献
