Role of immune complexes in thyroid diseases

Calculation of cyclic voltammetric responses for the catalytic reduction of substrates via intramolecular electron transfer within a catalyst-substrate complex formed on electrode surfaces

Cyclic voltammetric responses are calculated for the case where the reduction of a substrate proceeds only via intramolecular electron transfer to the substrate molecule when it is bound to reduced catalyst molecules present in a single monolayer on the electrode surface. The dependences of the voltammetric responses on the equilibrium constant for the formation of the catalyst-substrate complex, the rate of its formation and the rate of intramolecular electron transfer are discussed. Procedures are described for estimating the rate constant for the intramolecular electron transfer reaction and for discriminating between mechanistic possibilities.     

胸骨"T"形锯开微创切口保留后叶装置二尖瓣置换术临床探讨

目的 介绍胸骨中下段“T”行锯开微创切口、保留后叶装置、连续缝合法人工机械瓣二尖瓣置换术的临床观察结果。方法 本组42例患应用胸骨中下部“T”形锯开微创切口行保留二尖瓣后叶装置人工机械瓣置换术，其中男性18例，女24例，年龄31～56岁。结果 应用该手术技术的患，手术顺利，无术后并发症及死亡。结论 本术式具有安全、创伤小、术后心脏功能恢复快、并发症少等优点。     

FKBP6基因278C/A单核苷酸多态性与原发无精症相关研究

目的对FKBP6基因第3、4外显子进行突变和多态性筛查,研究第3外显子278C/A位点及第2内含子C/T位点(rs7797242)在无精症患者和正常男性中的多态性,初步探讨与原发无精症的相关性。方法采用变性高效液相色谱和聚合酶链反应-限制性片段长度多态性方法,对第3、4外显子进行突变和多态性筛查,对177例无精症患者和231名正常男性的278C/A和C/T(rs7797242)多态性进行基因分型。结果278C和278A等位基因频率符合Hardy-Weinberg平衡。无精症患者278A显著低于正常对照,差异有统计学意义(P<0.05)。C/T多态性在两组中均未检出,第3、4外显子未筛查到新的变异。结论278A等位基因可能与原发无精症相关。C/T(rs7797242)及370G/A,430G/C,467T/C,468G/A在中国人群中非常罕见。     

简单灵敏的免疫复合物抗原特异性固相酶联检测法

本文将聚乙二醇(PEG)比浊法和固相酶联免疫法(ELISA)结合,建立了—较灵敏的免疫复合物(IC)直接固相吸附抗原特异性检测法。利用牛清蛋白(BSA)为已知抗原组份的IC模型,分别对IC直接固相吸附的条件和影响因素、方法的灵敏度、重复性等进行了研究。结果发现IC在解离状态下直接固相吸附后的抗原特异性检测灵敏度明显高于未解离者。该法具有简单易行,灵敏度较高、适于临床测定大量血清样品等优点。     

脂膜微泡结合凝血酶原复合物的制备

目的:制备结合凝血酶原复合物(PCC)的阴离子脂膜微泡,评价微泡的理化性质。材料和方法:采用机械振荡直接连接法,分两组(于机械振荡之前或之后加入),制备结合PCC的阴离子脂膜微泡,观察并检测洗涤前后微泡的理化性质。结果:微泡与PCC的结合率及Ⅸ因子活性:两组间比较无明显差异(P>0.05),但洗涤后结合率由洗涤前的95%降为80%,活性由90%降为19%。结论:直接连接法可制备结合PCC的阴离子脂膜微泡,微泡与PCC结合率较高,且洗涤前能保持Ⅸ因子较高的活性。     

Mechanism of uptake and retention of F-18 BMS-747 158-02 in cardiomyocytes: a novel PET myocardial imaging agent

Background BMS-747158-02 is a novel fluorine 18-labeled pyridazinone derivative designed for cardiac imaging. The uptake and retention mechanisms of F-18 BMS-747158-02 in cardiac myocytes were studied in vitro, and the biodistribution of F-18 BMS-747158-02 was studied in vivo in mice. Methods and Results Fluorine 19 BMS-747158-01 inhibited mitochondrial complex I (MC-I) in bovine heart submitochondrial particles with an IC₅₀ of 16.6±3 nmol/L that was comparable to the reference inhibitors of MC-1, rotenone, pyridaben, and deguelin (IC₅₀ of 18.2±6.7 nmol/L, 19.8±2.6 nmol/L, and 23.1±1.5 nmol/L, respectively). F-18 BMS-747158-02 had high uptake in monolayers of neonatal rat cardiomyocytes (10.3%±0.7% of incubated drug at 60 minutes) that was inhibited by 200 nmol/L of rotenone (91%±2%) and deguelin (89%±3%). In contrast, an inactive pyridaben analog, P-0 (IC₅₀ value>4 μmol/L in MC-1 assay), did not inhibit the binding of F-18 BMS-747158-02 in cardiomyocytes. Uptake and washout kinetics for F-18 BMS-747158-02 in rat cardiomyocytes indicated that the time to half-maximal (t_1/2) uptake was very rapid (approximately 35 seconds), and washout t_1/2 for efflux of F-18 BMS-747158-02 was greater than 120 minutes. In vivo biodistribution studies in mice showed that F-18 BMS-747158-02 had substatial myocardial uptake (9.5%±0.5% of injected dose per gram) at 60 minutes and heart-to-lung and heart-to-liver ratios of 14.1±2.5 and 8.3±0.5, respectively. Positron emission tomography imaging in the mouse allowed clear cardiac visualization and demonstrated sustained myocardial uptake through 55 minutes. Conclusions F-18 BMS-747158-02 is a novel positron emission tomography cardiac tracer targeting MC-I in cardiomyocytes with rapid uptake and slow washout. These characteristics allow fast and sustained accumulation in the heart.     

The nocturnal jejunal migrating motor complex: defining normal ranges by study of 51 healthy adult volunteers and meta-analysis

Interdigestive human small bowel motility is characterized by the migrating motor complex (MMC). The aims of this study were to: (i) establish the normal range of variables of the nocturnal jejunal MMC and (ii) incorporate these data in a subsequent meta-analysis. Eighty-one recordings were performed by prolonged (24 h) ambulatory manometry in 51 subjects in two centres. Quantitative analysis was undertaken of 419 Phase III and 332 Phase II episodes. Adjusted mean values of seven variables were calculated using a mixed-effects model. Meta-analysis of pooled published data to generate a reliable 95% reference range was also performed. Adjusted mean values and confidence intervals are presented for all seven variables. Intrasubject variances were large in comparison with intersubject. Meta-analysis of 19 studies (356 pooled patients) meeting inclusion criteria produced wide reference ranges. At least five such ranges are useful for the detection of abnormality in the individual. This is the largest study of normal volunteers presented to date, with ranges for many variables produced using appropriate statistical methodology. A model for definition of abnormality has been proposed. We recommend that these data may be used by investigators in this field as a complement to other existing indicators of small bowel dysmotility.     

保留后方韧带复合体在胸腰椎骨折治疗中的意义

[目的]探讨保留后方韧带复合体在胸腰椎骨折治疗中的意义。[方法]1998～2002年单一节段胸腰椎爆裂骨折治疗中保留后方韧带复合体44例，同期行胸腰椎爆裂骨折治疗切除后方韧带复合体者35例。两种方法分别有15例（定义为A组）和12例（定义为B组）行2次手术取出内固定物，比较AB两组间2次手术前后腰椎功能JOA评分。[结果]2次手术前腰椎功能JOA评分：A组平均24．5分（19～29分）；B组平均22．8分（19～29分），二者无显著性差异（P〉0．05）。2次手术后JOA评分：A组平均24．8分（20～29分），B组平均17．5分（13～26分）。二组有显著性差异（P〈0．05）。[结论]胸腰椎骨折治疗中保留后方韧带复合体，利于术后腰椎功能恢复。     

重组耻垢分枝杆菌-Sj26GST疫苗的免疫保护作用研究

目的初步研究重组耻垢分枝杆菌疫苗rMc-Sj26GST(recombinant M.smegmatis mc2155-Sj26GST)对小鼠的免疫保护作用.方法用耻垢分枝杆菌重组疫苗rMc-Sj26GST免疫雄性BALB/c小鼠,检测小鼠淋巴细胞刺激指数(SI),腹腔巨噬细胞培养上清释放NO量,小鼠血清IFN-γ、IL-2的含量,并计数小鼠肝、肺活细菌数.结果小鼠脾淋巴细胞刺激指数(SI)为2.64±1.37,与Mc组(载体组)1.28±0.41相比,差异具有显著性;小鼠血清IFN-γ为196.43 pg*ml-1,与Mc组112.57 pg*ml-1相比,差异有显著性,较对照组高41%;小鼠血清中IL-2的浓度较对照组高.经rMc-Sj26GST疫苗免疫的小鼠受结核杆菌攻击后其肺、肝脏结核杆菌数较对照组少.结论耻垢分枝杆菌重组疫苗rMc-Sj26GST增强了小鼠细胞免疫功能,并使小鼠能抵抗结核杆菌的攻击.
