Prevalence and severity of erectile dysfunction (ED) increase with aging and are often associated with illnesses, like diabetes mellitus, heart disease, and hypertension, pathologically characterized by endothelial dysfunction and whose prevalence increases with age. The assumption that ED is mainly a neurovascular disease is supported by the evidence that specific phosphodiesterase type 5 (PDE5) inhibition produces an efficient erection in a wide range of ages and conditions. The availability of specific PDE5 inhibitors has enabled the development of effective treatment strategies, in this contest, tadalafil may be considered as the least “typical” PDE5 inhibitor. In clinical trials, tadalafil significantly enhanced, in patients of different ages, all efficacy outcomes across disease etiologies and severities. With an effectiveness lasting up to 36h, tadalafil allows patients to choose when to have sexual activities without the need to time it, showing positive feedback in terms of quality of life related to the treatment. Headache and dyspepsia were the most frequent side-effects of tadalafil, followed by back pain, nasal congestion, myalgia, and flushing, but the impact that long time action could have on effectiveness and safety is not yet entirely defined. The aim of this article is to critically review the available evidence from the tadalafil clinical research program and give the physician a rational approach for intervention in the treatment of ED and related diseases.  相似文献

Recently the term 'global aging', which refers to the universal trend of aging of the world population, is being widely used. First of all, I will shed some light on this trend of world population aging from the viewpoint of regional populations and their future projections.
Second, I will discuss about the determinants of population aging in the Asia and Oceania region. So far as the region is concerned, it will be divided into three groups of countries according to the level of population aging. The first group is the most advanced aged countries such as Japan, Australia, and New Zealand whose percentage of the elderly aged 65 and over is more than 10% in the year of 2000. The second group is the aging countries such as Korea, Taiwan, Singapore, and China whose percentage of the elderly is between 5 and 10%. and the third group is the remaining countries in this region whose percentage is less than 5%. The demographic aspects of each group are analyzed and the relationships between the fertility reduction and aging are also discussed in the perspectives of the demographic transition.
Third, I will discuss about the consequences of population aging in this region with special reference to Asian developing countries. In this section, I will mention the problems of dependency burden, the so-called population bonus, health expectancy, and social security systems. Fourth, some prominent characteristics concerning the living conditions and norms or attitudes of the elderly will be indicated, and I will also raise such points like family systems, living arrangements, social participation, and Ikigai (the worth of life) peculiar to Asian countries. Fifth and finally, I will touch on the topics of the issues and policies of the aging society. At present the social status of the elderly is improving in Asian countries and it is in the hope that the myth of aging will disappear in the near future.  相似文献

Mitomycin C (MMC) at a dose of 8 mg/m2 was administered by short intravenous infusion to 14 cancer patients. All patients had normal renal, hepatic, cardiac and bone marrow functions. The plasma level of MMC, which was determined by high-performance liquid chromatography over a 24–h period after the infusion, fitted the 2-compartment model curve except for one patient. There was a significant correlation between the area under the time versus concentration curve (AUC) of MMC and the age of patients ( r = 0.558, P < 0.05). Pharmacokinetic parameters in one of the older (a 69-year-old male) patients were extremely different from those of the other 13 patients. This patient experienced severe myelosuppression, probably due to the markedly increased AUC of MMC. Our results suggest that a patient's age has a significant influence on the pharmacokinetics of MMC in patients with normal major organ functions and that in some patients, MMC pharmacokinetics may be altered, possibly due to interpatient variation in the activation or metabolic pathway of MMC.  相似文献

目的 观察松花粉的抗衰老作用。方法 用含松花粉的饲料喂养衰老小鼠 ,测定小鼠血清中超氧化物歧化酶 (SOD)、过氧化氢酶 (CAT)、谷胱甘肽过氧化物酶 (GSH Per)的活性 ,测定脑组织中丙二醛 (MDA)含量及脑组织和肝脏中的脂褐质 (Lf)含量 ,同时测定胸腺和脾脏的重量及网状内皮系统的吞噬功能。结果 松花粉能明显增加衰老小鼠血清中SOD、CAT、GSH Per活性 ,降低脑组织中MDA含量及脑组织和肝脏中的Lf含量 ,并增加胸腺和脾脏质量 ,提高网状内皮系统吞噬功能。结论 松花粉具有抗衰老作用  相似文献

Background: The hippocampus is a center of cognitive function and therefore hippocampal atrophy is the major factor in cognitive decline. Analysis of hippocampal size may make it possible to predict progression in cognitive impairment. To address this point, the present study investigated the relationship between hippocampal atrophy and dementia using magnetic resonance (MR) images and the Hasegawa Dementia Scale‐Revised (HDS‐R). Methods: The present study was performed on 274 subjects (14–97 years old; average, 66 years; 106 male and 168 female) who had no focal neurological deficit. Hippocampal area and whole brain area were measured in three series of coronal MR images taken from a 5‐mm slice rostrally along dorsal edge of the pons, and hippocampal size normalized by calculating summated hippocampal areas as percentages of summated whole brain areas. Dementia was screened for using HDS‐R. Results: Hippocampal size decreased and HDS‐R reduced with age. Hippocampal atrophy was highly correlated with cognitive deterioration; a critical normalized hippocampal size for HDS‐R of less than 20, which corresponds with mild cognitive impairment and dementia, was found in 65% of over 60‐years old subjects and 98% of subjects with HDS‐R of less than 20 were over 60 years old. Conclusion: There is a high probability that over 60‐year‐old people with a normalized hippocampal size of less than 1.0 would develop dementia in the future, even though their HDS‐R might presently be over 20. Measurement of hippocampal size with coronal MR imaging may therefore contribute to prospective diagnoses of age‐related dementia.  相似文献

目的 探讨老年排尿功能障碍的神经机制。方法 利用水浴条件下离体逼尿肌条张力测定技术,观察老龄Wistar大鼠膀胱逼尿肌条对不同浓度卡巴可、去甲肾上腺素、ATP的收缩反应改变;阿托品对卡巴可诱导逼尿肌收缩的舒张反应改变;异丙肾上腺素对电刺激诱导逼尿肌收缩的舒张反应改变。结果 与青龄组相比,老龄大鼠膀胱逼尿肌对ATP的收缩反应显著增强,对异丙肾上腺素的舒张反应在高浓度时增强;对卡巴可、阿托品、去甲肾上腺素的反应没有差异。结论 老龄大鼠排尿功能障碍可能与β-肾上腺素能、嘌呤能神经改变有关。  相似文献

本文对韦氏成人智力量表中国修订本25岁及其以上成年人城乡样本共1976人在校正各年龄组的文化程度以后,比较了年龄和教育因素对成年人智力发展的影响。结果发现:与晶体智力有关的测验成绩并不随年龄增大而下降,某些测验(如知识;领悟等)甚至呈上升趋势;与流体智力有关的测验成绩随年龄的增大而下降;而测量精神运动速度(数字符号测验)的成绩受年龄影响最为明显。看来流体-晶体智力理论能较好地解释成年人的典型智力发  相似文献

In aging brain, degenerative as well as compensatory regenerative processes are believed to occur. The neural cell adhesion molecule NCAM is involved in developmental and regenerative processes in the brain. However, the role of NCAM in aging brain has not been characterized. In this study, the expression of NCAM mRNAs and polypeptides was investigated in aging rat brain. The 7.4 and 6.7 kb NCAM mRNAs were selectively downregulated during postnatal development, and the 5.2 and 2.9 kb NCAM mRNAs were upregulated. However, from postnatal day 40 to old age no change in NCAM mRNA classes was observed. The fraction of NCAM mRNA containing the VASE exon increased postnatally but remained stable during adult life. VASE, which is believed to modulate the binding capacity, seemed to be relatively more abundant in the 7.4 and 6.7 kb NCAM mRNAs, encoding transmembrane NCAM forms, than in the 5.2 and 2.9 kb NCAM mRNAs, coding for glycosyl phosphatidylinositol (GPI) linked NCAM. Conversely, insertion of exons a and AAG between exons 12 and 13, a region containing two fibronectin type III repeats, seemed to be more pronounced in 5.2 and 2.9 kb NCAM mRNAs than in the 7.4 and 6.7 kb mRNAs. During postnatal development an increase in the fraction of 6.7 kb NCAM mRNA containing the exons a and AAG was observed. However, during aging the fraction of NCAM mRNAs containing this exon combination seemed constant. At the protein level, NCAM-A was downregulated both during development and aging. No changes were observed during aging in the composition of soluble NCAM forms in the brain, cerebrospinal fluid or blood plasma. The amount of NCAM in rat brain decreased during postnatal development, but remained at a constant level from postnatal day 40 to old age.To conclude, several changes in NCAM expression occur during early postnatal development emphasizing the important role of this molecule in the morphogenetic processes. During aging, a significant selective downregulation of NCAM-A was observed indicating that in general only minor regenerative processes occur in the brain.  相似文献

本文通过对3月龄、18月龄、24月龄雄性大鼠下丘脑去甲肾上腺素(NE)、多巴胺(DA)、5-羟色胺(5-HT)、5-羟吲哚乙酸(5-HIAA),和下丘脑垂体促甲状腺释放激素(TRH)、垂体、血甲状腺释放激素(TSH)、血甲状腺激素(T_3、T_4)的测定,研究了大鼠下丘脑神经递质与甲状腺轴机能的增龄性变化,并在此基础上探讨了补益中药对老龄大鼠下丘脑—垂体—甲状腺机能的作用。结果提示:老龄大鼠下丘脑机能的减退是甲状腺轴机能减退的重要原因;补益中药可能改善了老龄大鼠下丘脑的调控机能,进而延缓了甲状腺轴机能的老年性变化。  相似文献
