Nasopharyngeal carcinoma (NPC) is one of the most common malignancies of the head and neck. It arises from the nasopharynx epithelium and is associated with high morbidity and mortality. Long non‐coding RNA (lncRNA) have been reported to regulate gene interaction and play critical roles in carcinogenesis and progression. LncRNA‐ROR, a recently identified lncRNA, has been shown to be involved in initiation, progression and metastasis of several tumors, including hepatocellular carcinoma, breast cancer and glioma. However, whether lncRNA‐ROR is associated with the progression of NPC remains unknown. Resistance to radiotherapy and chemotherapy is the primary cause of NPC patients’ death. In this study, we found that lncRNA‐ROR was significantly upregulated in NPC tissues compared with normal tissues. Next, our study proved that lncRNA‐ROR was highly associated with the proliferation, metastasis and apoptosis of NPC. The enrichment of lncRNA‐ROR played a critucal functional role in chemoresistance. The mechanism by which NPC resists chemotherapy might be that lncRNA‐ROR suppress p53 signal pathway. Taken together, these data suggested that lncRNA‐ROR played an important role in the progression of NPC; thereby it might become a therapeutic target and reduce chemoresistance for NPC.  相似文献

目的:研究长链非编码 RNA H19对骨肉瘤 MG63细胞增殖、侵袭的影响。方法:RNA 干涉技术沉默MG63细胞中 H19,荧光实时定量 PCR(RT -qPCR)检测骨肉瘤 MG63细胞中 H19的表达情况;流式细胞术检测细胞周期的变化;MTT 实验检测沉默 H19表达对细胞增殖的影响;Transwell 侵袭实验检测细胞侵袭能力的变化。结果:H19-siRNA 可有效沉默 H19在骨肉瘤细胞 MG63中的表达,沉默 MG63细胞中 H19的表达可以明显抑制骨肉瘤细胞的细胞周期,降低骨肉瘤细胞的增殖、侵袭能力。结论:H19在骨肉瘤的发生发展及侵袭中发挥重要作用。  相似文献

目的:分析长链非编码RNA (long non-coding RNA,lncRNA)在高氧诱导支气管肺发育不良(bronchopulmonary dysplasi,BPD)小鼠模型肺中表达谱的变化?方法:构建高氧诱导支气管肺发育不良新生小鼠模型,利用lncRNA芯片技术检测正常小鼠肺及BPD小鼠肺组织中lncRNA表达谱的变化,经过对原始数据进行预处理,筛选出差异表达的lncRNA?结果:与对照组相比,模型组差异表达的lncRNA(差异倍数≥2倍且P < 0.05)共1 769条,其中882条上调,887条下调;5倍以上上调的共140条,下调的共71条;10倍以上上调的共28条,下调的有2条?结论:高氧诱导BPD发生时,肺组织中lncRNA的表达谱发生了显著变化;这些差异表达的lncRNA,可能参与了BPD发生?发展过程?  相似文献

PurposeThis work explores the abnormal expression of long noncoding RNAs (lncRNAs), microRNAs (miRNAs) and messenger RNAs (mRNAs) in diabetic corneal epithelial cells (CECs) and constructs an associated competitive endogenous RNA (ceRNA) network. Moreover, we revealed that Rik may exert advantageous effects on diabetic corneal epithelial wound closure by sponging miR-181a-5p.MethodsWe obtained the profiles of differentially expressed lncRNAs (DELs) of CECs of type 1 diabetic versus control corneas by microarray and summarized the differentially expressed miRNAs (DEmiRs) and differentially expressed genes (DEGs) data by published literature. Subsequently, the ceRNA network was constructed using bioinformatics analyses. The levels of lncRNA ENSMUST00000153610/3632454L22Rik (Rik) and miR-181a-5p were verified. The localization of Rik was identified with fluorescence in situ hybridization (FISH), and dual-luciferase assays proved the targeted relationship between Rik and miR-181a-5p. Furthermore, we validated the functional impact of Rik in vitro.ResultsOverall, 111 upregulated and 117 downregulated DELs were detected in diabetic versus control CECs. The level of Rik located in both the cytoplasm and the nucleus was clearly downregulated, whereas miR-181a-5p was upregulated in vitro and in vivo in the diabetic group versus the control group. Rik can act as a ceRNA to bind to miR-181a-5p, thus promoting diabetic corneal epithelial wound healing in vitro.ConclusionsThis work investigated the expression profile of DELs and constructed ceRNA networks of diabetic CECs for the first time. Furthermore, we revealed that Rik may positively impact diabetic corneal epithelial wound healing by sponging miR-181a-5p, providing a novel potential therapeutic target of diabetic keratopathy (DK).  相似文献

目的 本研究旨在探讨lncRNA介导的ceRNA网络在变应性鼻炎(AR)病理生理过程中的作用及其在程序性细胞死亡(PCD)中的潜在调控功能。方法 纳入AR患者与正常人,取全血进行RNA测序分析,筛选差异表达的lncRNA、miRNA和mRNA用于构建基于ceRNA理论的lncRNA-miRNA-mRNA调控网络。采用基因本体(GO)和KEGG富集分析来预测ceRNA网络中mRNA的潜在生物学功能。随后,将差异表达的mRNA与PCD相关基因相互取交集,并依据交集mRNA构建新的ceRNA网络,同时对ceRNA网络中mRNA进行靶器官定位网络分析。结果 基于生物信息学分析,共提取5个lncRNA、4个miRNA和46个mRNA,构建AR中lncRNA介导的上调及下调的ceRNA网络。功能富集分析显示其在“钙信号通路”、“PI3K/AKT信号通路”及“ERK1和ERK2级联”等中显著富集。并对AC008394.1及人类组织相容性白细胞抗原复合物P5(HCP5)2个lncRNA构建PCD相关ceRNA调控网络。在对ceRNA网络中mRNA进行靶器官定位分析中发现,它们多在神经免疫相关器官中表达。结论 本研究构建了AR中新的ceRNA网络,初步探索了HCP5介导的ceRNA网络可能通过影响细胞凋亡对AR发挥作用。为AR发病机制的研究提供了新的见解和潜在的研究方案。  相似文献

BackgroundExpression of the TAZ gene is closely related to the prognosis of glioma patients. We hoped to find long noncoding RNAs (lncRNAs) related to TAZ and a new target for glioma treatment.MethodsTAZ‐related genes were found by dual‐luciferase reporter gene assay, and the correlation of each gene was analyzed by the Pearson method. Human glioma cell lines U87 MG and U251 and glioma rats were used for cytology assays, and the related genes were transfected. We conducted immunohistochemistry, RT‐qPCR, Western blotting, CCK8 test, flow cytometry, transwell assays, clone formation analysis, and tumor weight measurements to verify the above relationship.ResultsWe found that miR‐125a‐5p was closely related to the TAZ gene, and the lncRNA MIR4435‐2HG was closely related to miR‐125a‐5p. Both MIR4435‐2HG‐OE and TAZ increased the expression of the TAZ gene, activated the Wnt signaling pathway, inhibited apoptosis, and promoted migration and proliferation in glioma cells. Besides, it also increased the tumor volume of gliomas in a rat model subcutaneously inoculated with glioma cells. We also found miR‐125a‐5p could block the effect of MIR4435‐2HG‐OE and TAZ.ConclusionsLncRNA MIR4435‐2HG obstructs the functions of miR‐125a‐5p and promotes neuroglioma development by upregulating the TAZ gene.  相似文献
