目的探讨重楼提取物(PPEE)抗骨肉瘤作用及其机制。方法不同质量浓度的PPEE处理骨肉瘤细胞,采用MTT法、Hoechst 33342染色、流式细胞术、Matrigel培养、免疫印迹法分别检测细胞增殖抑制率、细胞凋亡、细胞周期、血管生成拟态(VM)形成及相关蛋白表达情况。裸鼠移植瘤模型观察PPEE的体内抑瘤效果。结果 PPEE对骨肉瘤细胞半数抑制浓度(IC50)为10~60μg/m L,对成骨细胞增殖影响较小。PPEE可浓度依赖性地将骨肉瘤143B细胞阻滞在G2/M期,上调细胞周期相关蛋白p-CDK1、p-Cdc25C、p-Chk2表达,下调cyclinB1表达;促进细胞凋亡,上调cleavedCaspase-3、8、9和PARP表达,上调Bax/Bcl-2;抑制细胞体外VM形成,下调FAK、Mig-7、MMP-2和MMP-9表达。体内实验显示PPEE能明显抑制骨肉瘤生长和体内VM形成,延长荷瘤裸鼠生存期。结论 PPEE在体内外均具有良好的抗骨肉瘤活性,其作用机制可能与诱导骨肉瘤细胞凋亡、阻滞细胞周期及破坏骨肉瘤VM形成有关。  相似文献

Intervertebral disc degeneration (IDD) is associated with dysregulated expression of microRNAs (miRNAs). However, the precise molecular mechanisms underlying this disorder remain unclear. Therefore, we tested the hypothesis that miRNAs modulate IDD through effects on the IL‐6/STAT3 signaling pathway, a potential regulator of IDD. The miRNA expression profile was determined in nucleus pulposus (NP) tissues from patients with IDD and controls, employing miRNA microarray and quantitative real‐time PCR (RT‐qPCR). Biological functions of differential expression miRNAs were further investigated using immunofluorescent staining. Luciferase reporter assays and Western blotting were performed to determine miRNA targets. We identified 41 miRNAs that were differentially expressed in patients compared with controls. Following RT‐qPCR confirmation, miR‐98 was significantly downregulated in degenerative NP tissues. Moreover, its level was inversely correlated with grade of disc degeneration. Through gain‐of‐function and loss‐of‐function studies, miR‐98 was shown to significantly promote type II collagen expression in NP cells. Interleukin‐6 (IL‐6) was identified as a target of miR‐98. Knockdown of IL‐6 induced effects on NP cells similar to those induced by miR‐98. In contrast, IL‐6 treatment abrogated the effects induced by miR‐98 upregulation. Moreover, miR‐98 dramatically suppressed expression of STAT3 target gene, MMP2. IL‐6 treatment antagonized this effect, whereas knockdown of IL‐6 by IL‐6 short hairpin RNA (shIL‐6) induced inhibitory effects on the expression of p‐STAT3 and its main target genes, similar to miR‐98. The mRNA level of IL‐6 was inversely correlated with that of miR‐98 in degenerative NP tissues. These results suggest the downregulation of miR‐98 could promote IDD through the IL‐6/STAT3 signaling pathway. Our findings also highlight miR‐98 as a novel hopeful therapeutic target for IDD. © 2015 American Society for Bone and Mineral Research.  相似文献

Timely detection is crucial for successful treatment of cancer. The current study describes a new approach that involves utilization of the tumor cell environment for bioimaging with in-situ biosynthesized nanoscale gold and iron probes and subsequent dissemination of Au-Fe nanoclusters from cargo exosomes within the circulatory system. We have isolated the Au-Fe cargo exosomes from the blood of the treated murine models after in situ biosyntheses from their respective pre-ionic solutions (HAuCl4, FeCl2), whereas Na2SeO3 supplementation added into Au lethal effect. The microarray data of various differentially expressed genes revealed the up-regulated tumor ablation and metal binding genes in SGC-7901 cell lines after treatment with Au-Fe-Se triplet ionic solution. The isolation of Au-Fe nanoclusters cargo exosomes (nano in nano) after secretion from deeply seated tumors may help in early diagnosis and reveal the tumor ablation status during and after the relevant treatment like radio-chemo therapies et al.  相似文献

目的:探讨Parkin介导的线粒体自噬在球形脂联素(globular adiponectin,gAPN)减轻间歇低氧(intermittent hypoxia, IH)所致H9C2心肌细胞损伤中的作用及相关机制。方法:建立H9C2细胞IH模型来模拟阻塞性睡眠呼吸暂停低通气综合征。 采用流式细胞术检测细胞凋亡。采用JC-1试剂盒检测线粒体膜电位(mitochondrial membrane potential,MMP)。采用免疫印迹法检测Parkin、Bax、Bcl-2蛋白表达。采用Parkin siRNA抑制Parkin基因表达。采用LC3与线粒体红色荧光探针进行荧光共定位检测线粒体自噬。结果:IH暴露后,H9C2细胞凋亡率、Bax及Parkin蛋白表达水平、线粒体自噬明显升高,MMP下降,Bcl-2 蛋白表达水平下降;给予gAPN保护后,IH+gAPN组细胞凋亡率、Bax蛋白表达水平较IH组下降,MMP较IH组升高,Bcl-2及 Parkin蛋白表达水平、线粒体自噬也较IH组升高。抑制Parkin基因表达后,线粒体自噬明显下降,线粒体损伤、心肌细胞凋亡率明显增加。结论:gAPN通过上调Parkin介导的线粒体自噬减轻IH所致H9C2心肌细胞损伤。  相似文献

探讨橄榄苦苷对circMBOAT2/miR-106a-5p 信号通路的调控作用及其对口腔鳞癌细胞CAL27增殖和凋亡的影响。方法 应用实时定量聚合酶链反应(RT-qPCR)检测circMBOAT2和miR-106a-5p在口腔鳞癌组织和癌旁组织中的表达水平。将口腔鳞癌细胞CAL27分为橄榄苦苷(0、200、400、800 μg/mL)组、si-NC组、si-circMBOAT2组、橄榄苦苷800μg/mL+pcDNA-circMBOAT2组。采用RT-qPCR检测各组细胞中circMBOAT2和miR-106a-5p的表达水平,检测并验证circMBOAT2与口腔鳞癌的相关性。应用CCK-8法、集落形成实验、流式细胞术测定CAL27细胞的增殖活力、集落形成能力和凋亡率。应用双荧光素酶实验确定 circMBOAT2和miR-106a-5p靶向关系。结果 口腔鳞癌组织中circMBOAT2表达显著高于癌旁组织(P<0.05),miR-106a-5p表达显著低于癌旁组织(P<0.05)。与橄榄苦苷0μg/mL组比较,橄榄苦苷(200、400、800 μg/mL)组CAL27细胞集落形成数、circMBOAT2水平显著降低(P<0.05),抑制率、凋亡率、miR-106a-5p水平显著升高(P<0.05)。与si-NC组比较,si-circMBOAT2组CAL27细胞集落形成数显著降低(P<0.05),抑制率、凋亡率、miR-106a-5p水平显著升高(P<0.05)。circMBOAT2与miR-106a-5p直接特异性结合。与橄榄苦苷800 μg/mL组比较,橄榄苦苷800μg/mL+pcDNA-circMBOAT2组CAL27细胞集落形成数显著升高(P<0.05),抑制率、凋亡率显著降低(P<0.05)。结论 橄榄苦苷通过下调circMBOAT2/miR-106a-5p通路可抑制口腔鳞癌细胞CAL27增殖,诱导细胞凋亡。  相似文献

Survivin是一种新的凋亡抑制蛋白(inhibitor of apoptosisprotein,IAP),具有细胞周期调控和凋亡抑制双重功能。在多种造血系统肿瘤组织中高表达,与诊断、预后和耐药密切相关。利用survivin致敏的树突状细胞疫苗、survivin反义核酸及Survivin阴性突变体可有效抑制肿瘤细胞生长,为采用生物学策略治疗造血系统肿瘤开辟了新途径.本综述重点阐述了survivin在造血系统肿瘤中的表达,survivin与造血系统肿瘤预后和耐药的关系及survivin在造血系统肿瘤治疗中的应用。  相似文献

目的探讨分析经脐单孔腹腔镜手术(laparoendoscopic single-site surgery,LESS)技术应用于全子宫切除术的安全性,并根据子宫大小、是否合并盆腔粘连分成单纯型和复杂型全子宫切除术绘制其学习曲线。 方法回顾性分析东南大学附属中大医院2017年3月至2019年11月的80例LESS全子宫切除术的临床资料,均由同一位熟练掌握妇科标准腹腔镜技术的术者施行。定义术中探查子宫大小如孕周数≤8周、无盆腔粘连者为单纯型全子宫切除术,子宫大小如孕周数>8周、合并盆腔粘连者定义为复杂型全子宫切除术。结合手术例数和手术时间绘制2组学习曲线。 结果80例手术全部顺利完成,无术中并发症发生,无术中更改手术方式。所有病例术后随访暂未发现切口感染、脐疝等术后并发症。观察2组学习曲线,单纯型全子宫切除术经过约20例后,手术时间渐趋平稳;复杂型全子宫切除术经过约26例后,手术时间渐趋平稳。 结论LESS在全子宫切除术中是安全、可行的,单纯型和复杂型全子宫切除术的学习曲线分别为20例和26例。  相似文献

Study objectiveWe aimed to determine the magnitude of peri-operative opioid sparing effect when general anesthesia is combined with spinal analgesia for laparoscopic gynecological surgery.DesignA prospective randomized controlled study; a three-group trial with two comparisons (each intervention group to control).SettingOperating room and postoperative recovery area.PatientsPatients aged between 18 and 65 years with American Society of Anesthesiologists physical status 1 or 2 who were scheduled for inpatient elective laparoscopic gynecological surgery with expected pneumoperitoneum duration of at least 20 min. Of 102 randomized patients, 99 completed the study.InterventionsPatients were randomized to general anesthesia alone (control group) or combined with very-low-dose (levobupivacaine 3.75 mg; sufentanil 2.5 μg) or low-dose (levobupivacaine 7.5 mg; sufentanil 2.5 μg) spinal analgesia.MeasurementsPrimary endpoints were perioperative opioid consumption and pain scores (11-point numeric rating scale) at 30 min, 1 h, 2 h, 4 h and 24 h post-surgery. Secondary endpoints were patient satisfaction with anesthetic care and participation in research, sevoflurane consumption and adverse effects.Main resultsIntra-operative sufentanil (median [95% CI]) consumption was 16.1 (10.5–22.6) μg/h in the control group versus 4.7 (3.2–9.2) μg/h in the very-low-dose and versus 2.9 (0.0–4.0) μg/h in the low-dose spinal analgesia groups (p < 0.001, for both comparisons). Median (95% CI) piritramide consumption at 24 h post-surgery was 7.5 (3–8) mg in the control group versus 5 (0–7.5) mg in the very-low dose spinal analgesia group (p = 0.182) and versus 2 (0–2.5) mg in the low-dose spinal analgesia group (p = 0.001). Postoperative pain scores were consistently <3 only in the low dose spinal analgesia group. Patient satisfaction with anesthetic care and participation in research was very high in all groups.ConclusionsLow-dose spinal analgesia in combination with general anesthesia reduces peri-operative opioid consumption in laparoscopic gynecological surgery in immediate postoperative period.  相似文献
