Green tea polyphenol (GTP) is one of the most promising chemopreventive agent for cancer; it can inhibit cancer cell proliferation and induce apoptosis through p53-dependent cell signaling pathways. Unfortunately, many tumor cells lack the functional p53, and little is known about the effect of GTP on the p53-deficient/mutant cancer cells. To understand the p53-independent mechanisms in GTP-treated p53-dificient/mutant cancer cells, we have now examined GTP-induced cytotoxicity in human hepatoma Hep3B cells (p53-deficient). The results showed that GTP could induce Bax and Bak activation, cytochrome c release, caspase activation, and necroptosis of Hep3B cells. Bax and Bak, two key molecules of mitochondrial permeability transition pore (MPTP), were interdependently activated by GTP, with translocation and homo-oligomerization on the mitochondria. Bax and Bak induce cytochrome c release. Importantly, cytochrome c release and necroptosis were diminished in Hep3B cells (Bax?/?) and Hep3B cells (Bak?/?). Furthermore, overexpression of Bcl-2 could ameliorate GTP-induced cytochrome c release and necroptosis. Together, the findings suggested that GTP-induced necroptosis was modulated by the p53-independent pathway, which was related to the translocation of Bax and Bak to mitochondria, release of cytochrome c, and activation of caspases.  相似文献

目的探讨应用沙美特罗/氟替卡松粉吸入剂(舒利迭)在慢性阻塞性肺疾病(COPD)稳定期治疗中对肺功能和全身炎症标记物的影响。方法选取2011年11月至2012年11月在潍坊市人民医院治疗的COPD稳定期患者60例作为研究对象。将所有患者随机分为实验组和对照组,每组各30例。对照组患者给予为期8周的常规止咳、平喘、吸氧等对症支持治疗;实验组在与对照组患者相同治疗的基础上加用沙美特罗/氟替卡松粉吸入剂(舒利迭)治疗,为期8周,单次剂量为50/500μg,每日两次。观察患者治疗前及治疗8周后血清可溶性细胞间黏附分子-1和白细胞介素IL-6、IL-8、IL-17等指标,同时观察两组患者肺功能的变化。结果实验组稳定期COPD患者血清可溶性细胞间黏附分子-1浓度在治疗前后变化差异有统计学意义(P<0.05)。实验组患者治疗后血清IL-6、IL-8、IL-17的水平较治疗前明显降低,差异有统计学意义(P<0.05)。实验组患者肺功能指标FVC、FEV1、FEV1/FVC%在治疗前后有明显统计学差异(P<0.05)。结论通过吸入沙美特罗/氟替卡松粉吸入剂可以明显改善慢性阻塞性肺疾病患者肺功能,降低患者血清中可溶性细胞间黏附分子-1及IL-6、IL-8、IL-17的水平,减轻气道炎性细胞浸润。  相似文献

目的 了解学龄期哮喘儿童的病情控制情况及睡眠状况,研究哮喘控制与睡眠紊乱的相关性,以期为哮喘儿童睡眠障碍的防治提供依据。方法 对2018年10月-2019年10月在潍坊市人民医院小儿内科就诊的200例6~11岁支气管哮喘儿童及其家长进行问卷调查,包括哮喘儿童一般情况问卷、儿童哮喘控制测试问卷(C-ACT)、儿童睡眠紊乱量表(SDSC)。根据C-ACT得分将哮喘患儿分为完全控制组(≥23分,75例)、部分控制组(20~22分,46例)、未控制组(≤19分,79例),对3组哮喘儿童睡眠紊乱情况进行比较,并将SDSC总分及各维度得分与C-ACT得分进行相关性分析。结果 哮喘儿童的完全控制率为37.5%,部分控制率为23.0%,未控制率为39.5%。完全控制组、部分控制组和未控制组哮喘儿童SDSC总得分分别为34.21±3.60、38.74±2.86、40.09±5.37,差异有统计学意义(F=38.857,P<0.001)。睡眠紊乱的检出率分别为未控制组 49.4%、部分控制组40.0%、完全控制组20.0%,差异有统计学意义(χ2=15.224,P<0.001);哮喘儿童C-ACT得分与SDSC得分呈负相关(r=-0.623,P<0.01)。结论 本地区哮喘儿童的哮喘控制水平欠佳;哮喘儿童睡眠紊乱的检出率较高;完全控制组哮喘儿童睡眠质量明显优于部分控制组和未控制组的患儿,儿科医护人员在对哮喘儿童进行整体治疗时应更加关注哮喘控制状况,通过良好的哮喘控制改善其睡眠状况,从而促进患儿的生长发育。  相似文献

BackgroundIn order to avoid excessive treatment of thyroid nodules in the clinic, it is necessary to find a simple and practical analysis method to comprehensively and accurately reflect benign or malignant thyroid nodules. This study aimed to construct and validate a comprehensive and reliable network-based predictive model using a variety of imaging and laboratory criteria for thyroid nodules to stratify the risk of malignancy prior to surgery.MethodsWe retrospectively analyzed data from patients who underwent surgical treatment for thyroid nodules at the Thyroid and Breast Diagnosis and Treatment Center of Weifang Hospital of Traditional Chinese Medicine between January 2018 and December 2020. Binary logical regression analysis was performed to predict whether nodules were malignant or benign. The developmental dataset included 457 patients (January 2018–December 2020). The validation set included separate data points (n = 225, January 2018–December 2020).ResultsIn this study, criteria that showed significant predictive value for malignant nodules included TI-RADS: 4b (p = 0.065); Bethesda IV, Bethesda V, Bethesda VI (P < 0.0001); BRAFV600E mutation (P < 0.0001); Calcitonin>5 pg/ml (p = 0.0037); and FNA-Tg>30 ng/ml (p = 0.0003). A 10-grade risk scoring system was developed. The risk of malignancy risk ranged from 2.06% to 100% and was positively associated with increasing risk grade. The areas under the receiver-operating characteristic curve of the development and validation sets were 0.972 and 0.946, respectively.ConclusionA simple, comprehensive and reliable web-based predictive model was designed using a variety of imaging and laboratory criteria to stratify thyroid nodules by probability of malignancy.  相似文献

Background: Parkinson’s is a common, progressive, neurodegenerative movement disorder of the central nervous system. Research has consistently shown that dancing can have a positive impact on the lives of people living with Parkinson’s (PwP).PwP who dance report that current measurement of the effects fails to appreciate the multifactorial presentation of the condition as well as the biopsychosocial potential of dance holistically.Aim: To explore the feasibility of combining measurements of whole-body movement using 3-dimensional motion analysis with the lived experience of dance for PwP, to see if it is possible to ‘see (measure)’ what PwP say they ‘feel (experience)’ when dancing, in a ‘mind/body approach’Method: Thirteen PwP and ten Carers took part in three community delivered ‘Parkinson’s Dance Science’ classes followed by focus group reflection. Three PwP wore a 3-dimensional, motion capture suit that recorded whole-body movement throughout the class and were interviewed about their experiences.Data were analysed using a ‘Convergent Parallel’ mixed methods design, mapping participant’s experiences and reflections over six biomechanical movement variables.Results: Quantitative results indicate that it is possible to use 3-dimensional motion capture to measure biomechanical change during and following dance in PwP but the amount and direction of change was different for each participant.Qualitative results showed PwP felt the methods were feasible and reported a positive immediate and long-term effect of dance in a complete ‘mind/body experience’.The sample size did not permit further analysis, but areas of development suggest it may not be the change in physical ability that is important, but more the perception of change following dance that is unique to each individual. Thus single measures of the effects of dance for PwP are likely to be insufficient when trying to understand the effect holistically.Conclusion: A ‘Convergent Parallel’ mixed methods research design, mapping the experience of dance for PwP (‘what I feel’) and their peers against biomechanical change (‘what you see’) following dance is feasible.Further research is required to develop the method with a larger sample to truly begin to understand the potential and effects of dance for PwP.  相似文献

Icariin (ICA) is obtained from Epimedium brevicornu maxim and exploited to remedy miscellaneous cancers. But the role of ICA in medulloblastoma remains hazy. The research delved into the antitumor activity of ICA in medulloblastoma DAOY cells. ICA with diverse concentrations was utilized to stimulate DAOY cells, and the biological functions of ICA in medulloblastoma DAOY cells were examined. Then, the relative SPARC expression was determined in ICA‐managed DAOY cells, and the pc‐SPARC vector was transfected into DAOY cells to further probe the influence of SPARC and JAK1/STAT3 and PI3K/AKT pathways in ICA‐managed DAOY cells. A xenograft model was established to investigate the function of ICA in vivo. ICA restrained cell viability, expedited apoptosis, prohibited cell migration and invasion, and meanwhile affected the associative factors expression in DAOY cells. Additionally, SPARC expression was declined in ICA‐stimulated DAOY cells. Overexpressed SPARC reversed the functions of ICA in above‐involved cell behaviors of DAYO cells and the correlative protein levels. Besides, ICA notably frustrated JAK1/STAT3 and PI3K/AKT activations in DAOY cells. Beyond that, ICA prohibited tumor formation in vivo. The results concluded that ICA exhibited the antitumor activity in DAOY cells via decreasing SPARC and inactivating JAK1/STAT3 and PI3K/AKT pathways.  相似文献

The effects of physical-therapy intervention on the motor function of upper limbs and the quality of life in patients with Parkinson’s disease (PD) are not fully understood. We evaluated the effects of a progressive muscle-strengthening protocol for upper limbs on the functionality and quality of life. Patients were divided into two groups: Intervention (n = 6) and Control (n = 7). Assessment tools used were: Unified Parkinson’s Disease Rating Scale (UPDRS), Parkinson’s Disease Questionnaire, Nine-Hole Peg Test (9HPT), Test d’Évaluation des Membres Supérieurs de Personnes Âgées (TEMPA), 10-Repetition Maximum (10-RM) and handgrip dynamometer, which were applied pre- and post-intervention, with follow-up for one month after the last training session. Only, the Intervention group (post-intervention) showed significant statistical differences, with the following outcomes: UPDRS III (p = 0.042); 9HPT, right (p = 0.028) and left side (p = 0.028); TEMPA for total right side (p = 0.028), left side (p = 0.028) and total bilateral tasks (p = 0.028); TEMPA task 2 – open a jar and take a spoonful of coffee (p = 0.028), task 3 – pick up a pitcher and pour water into a glass for right (p = 0.046) and left side (p = 0.028), task 5 – write on an envelope and stick on a stamp (p = 0.028), and task 6 – shuffle and deal playing cards (p = 0.028). We observed significant statistical differences between groups (post-intervention) for TEMPA task 6 (p = 0.032), total right side (p = 0.032), and total bilateral tasks (p = 0.032). An increase in the maximum load in the post-intervention stage, based on the 10-RM test, was observed on the right (p = 0.003) and left (p = 0.007) sides. Our results showed an improvement in upper-limb functionality in PD patients submitted to progressive muscle-strength training, although not in quality of life.  相似文献

【摘要】 目的 探讨外源性胆绿素对中波紫外线(UVB)照射的HaCaT细胞光损伤的保护作用。方法 将HaCaT细胞分为加入0、0.1、1、10 μmol/L胆绿素并照射UVB的UVB组、0.1 μmol/L UVB组、1 μmol/L UVB组、10 μmol/L UVB组及不做处理的对照组。UVB照射剂量为30 mJ/cm2,照射后继续培养24 h,分别检测细胞活性氧(ROS)水平、超氧化物歧化酶(SOD)活力、丙二醛(MDA)含量,ELISA法检测各组细胞的炎症因子白细胞介素6(IL-6)、IL-8水平。多组间均数比较采用单因素方差分析,组间两两比较采用LSD-t检验。结果 UVB组、0.1 μmol/L UVB 组、1 μmol/L UVB组、10 μmol/L UVB组、对照组细胞ROS水平(3 613.33 ± 206.61、2 958.67 ± 193.87、2 678.33 ± 178.24、2 274.67 ± 118.81、1 905.67 ± 250.25)、SOD活力(24.41 ± 1.78、28.96 ± 2.21、29.75 ± 1.75、30.19 ± 2.29、37.52 ± 2.31)、MDA含量(5.61 ± 0.32、5.46 ± 0.55、4.65 ± 0.22、2.55 ± 0.93、1.31 ± 0.05)、IL-6水平、IL-8水平差异均有统计学意义(F值分别为 34.02、57.36、214.09、29.73、11.40,均P < 0.05),UVB组ROS水平、MDA含量及IL-6、IL-8水平均高于另4组(均P < 0.05),SOD活力均低于另4组(均P < 0.05)。结论 外源性胆绿素减轻UVB引起的HaCaT细胞的氧化损伤、减轻炎症反应和抑制脂质过氧化作用,对细胞光损伤有一定的保护作用。  相似文献

Background & objective: The effects of green coffee bean extract (GCBE) supplementation on inflammatory biomarkers have been widely spread. The purpose of this article was to assess the impact of GCBE supplementation on C-reactive protein (CRP) levels.MethodsThe literature search was performed in four databases (Scopus, PubMed, the Cochrane Library, and Google Scholar) to identify studies that examined the influence of GCBE supplementation on CRP levels up to August 2019. Mean and standard deviation (SD) of the outcomes were used to estimate the weight mean difference (WMD) between intervention and control groups for the follow-up period.ResultsFive (5) studies, with 6 arms, reported CRP as an outcome. Statistically, the use of GCBE supplements resulted in a significant change in CRP levels (WMD: −0.017 mg/dL, 95 % CI: −0.032, −0.003, p = 0.018), whose overall findings were obtained from random-effects model. In addition, a significantly greater reduction in CRP was noted for studies with doses of GCBE supplements ≥ 1000 mg/d (WMD: −0.015 mg/dL, 95 % CI: −0.020, −0.010, p < 0.000), length of intervention < 4 weeks (WMD: -0.015 mg/dL, 95 % CI: −0.020, −0.010, p < 0.001), and for non-healthy subjects (WMD: −0.019 mg/dL, 95 % CI: −0.027, −0.011, p < 0.001). Dyslipidemia, hypertension and non-alcoholic fatty liver disease were the ailments of the studies that encompassed non-healthy patients.ConclusionsThis meta-analysis shows that the use of GCBE supplements resulted in a statistical decrease in CRP levels, mainly for non-healthy subjects. However, due to the limited number of studies, further randomized clinical trials are crucial in this regard.  相似文献
