目的 :探讨 L-精氨酸对体外培养的心肌细胞和内皮细胞一氧化氮合酶 (NOS)活性的影响。方法 :体外培养 SD大鼠的心肌细胞和心脏微血管内皮细胞 ,72 h后分组 ,实验组在生长液中加入 1 mol/L的 L-精氨酸 ,分别在加入 L-精氨酸后 1 /2、1、2、4h取细胞 ,反复冻融 3次使细胞破裂 ,用分光光度法测细胞内 NOS活性。结果 :加入 1 mol/L的 L-精氨酸后 ,心肌细胞的 NOS活性在 1 /2 h升高至对照组的 1 2 8% (P <0 .0 5) ,2 h时升高至 1 37% (P <0 .0 5) ,4h后下降 ;而内皮细胞在加入 L-精氨酸后 1 /2、1、2、4h时的 NOS活性分别为对照组的 1 2 2 %、1 38%、1 2 8%和1 0 3% ,且内皮细胞 NOS活性较心肌细胞为高。结论 :L-精氨酸可升高心肌细胞和心脏微血管内皮细胞的 NOS活性 相似文献

The effects of dexamethasone (Dex) on cultured rat beating heart cells infected with 100 TCID-50 Coxsackie virus B-2 (CB2V) were observed. The beating % began to decrease in the infected group 2 or 3 d post-challenge. Meanwhile, the cytopathic effect (CPE) appeared rapidly from 1+ to 3+. In the infected and Dex-treated group 1 h after inoculation, the beating % and CPE in the whole flask were significantly higher and less, respectively, than that in the group infected (P less than 0.05) at the same intervals. At 5 d after challenge, the beating % in the whole flask was significantly higher than that in the infected group. The cardiac enzyme-aspartate aminotransferase (AST) in the infected group was higher than that in the infected and Dex-treated group (P less than 0.01) through 3-5 d post-challenge. Moreover, the AST levels in these 2 groups were also higher than that in the uninfected group, Dex control group at the same intervals (P less than 0.01). Ultrastructural findings were parallel to the results of CPE through 1-5 d post-challenge in these 4 groups. It is suggested that the protective effect of Dex on cultured beating heart cells infected with CB2V occurred in the early stages after infection. It is surmised that steroids can probably save the lives of patients with severe myocarditis if the conventional therapy for protecting the myocardium and improving immunity were administered together. 相似文献

观察了维拉帕米(Verapamil,Ver)对感染柯萨奇B_3病毒(CVB_3)的大鼠培养心肌细胞Ca~(2+)内流及CVB_3-RNA复制的影响。结果发现在感染48h后,Ver对感染细胞及正常对照的Ca~(2+)内流均有显著的抑制作用(P<0.01);若在病毒感染同时加入Ver,经48h培养后,细胞中CVB_3-RNA含量显著高于病毒对照组(P<0.05)。提示钙拮抗剂(如Ver)可减少病毒感染引起的心肌Ca~(2+)内流增加,有可能减轻感染细胞的继发性Ca~(2+)损伤;但Ver会促进病毒RNA的复制,提示在急性病毒性心肌炎临床上用Ver治疗心律失常时宜慎重。 相似文献

本文报道用微量板中和试验以结晶紫染色固定细胞后在微量酶标自动读数仪上按细胞染色深浅所得不同光密度,自动将细胞病变定量读数记录,检测40例急性病毒性心肌炎患者早期及恢复期血清中柯萨奇B组病毒中和抗体,并与微量板中和试验在倒置显微镜下观察结果相比较。二法总符合率为96.8%,但前法有正确、敏感、重复性强、省时、省视力,能快速阅读结果,又有客观记录可资备查等优点。 相似文献
