The potential role of free chitosan in bone trauma and bone cancer management

Bone defects caused by fractures or cancer-mediated destruction are debilitating. Chitosan is commonly used in scaffold matrices for bone healing, but rarely as a free drug. We demonstrate that free chitosan promotes osteoblast proliferation and osteogenesis in mesenchymal stem cells, increases osteopontin and collagen I expression, and reduces osteoclastogenesis. Chitosan inhibits invasion of endothelial cells, downregulating uPA/R, MT1-MMP, cdc42 and Rac1. Better healing of bone fractures with greater trabecular bone formation was observed in mice treated with chitosan. Chitosan induces apoptosis in osteotropic prostate and breast cancer cells via caspase-2 and -3 activation, and reduces their establishment in bone. Chitosan is pro-apoptotic in osteosarcoma cells, but not their normal counterpart, osteoblasts, or chondrosarcoma cells. Systemic delivery of chitosan does not perturb angiogenesis, bone volume or instinctive behaviour in pregnant mice, but decreases foetal length and changes pancreatic secretory acini. With certain controls in place, chitosan could be useful for bone trauma management.     

细胞黏附分子CD99在人髓母细胞瘤中的表达及意义

目的 探讨细胞黏附分子CD99在人髓母细胞瘤组织中的表达特点及其临床意义。方法 采用免疫组化法检测107例髓母细胞瘤及15例瘤旁正常脑组织标本中CD99的表达。所有病人均随访至死亡或截止至2013年12月，随访时间为12~80个月，利用Kaplan-Meier生存曲线进行生存分析。结果 髓母细胞瘤CD99阳性表达率（57.94%，62/107）明显高于瘤周正常脑组织（0%，0/15；P<0.05）。CD99的表达水平与病人性别、年龄、肿瘤的病理组织学分型和分子亚型均无明显相关性（P>0.05）。CD99的高表达与髓母细胞瘤病人的预后呈显著负相关（P<0.05）。结论 CD99高表达与髓母细胞瘤预后显著相关，可作为预后判断的指标及治疗髓母细胞瘤潜在的新靶点。     

急性心肌梗死患者检测早期Mb、TnI指标的临床意义

测定心肌梗死患者早期血清肌红蛋白(Mb)与血清肌钙蛋白(TnI)指标可作为早期诊断的标的物。急性心肌梗死患者由于早期心肌损伤,心肌营养不良以及并发严重的心力衰竭等体征,促使患者肌红蛋白值(Mb)与肌钙蛋白值(TnI)增高;采用金标法试剂盒检测心肌梗死患者血清中的肌红蛋白与肌钙蛋白指标,结临床确诊提供可靠的依据,方法简单迅速。     

针灸治疗神经衰弱的实效性分析

目的探讨针灸治疗神经衰弱实效性分析。方法随机抽取本院2010年3月至2011年8月共收治的72例神经衰弱患者的临床资料,将其随机分为两组,对照组与治疗组,每组各36例,其中对照组患者采用常规抗神经药物治疗神经衰弱,治疗组患者应用对照组常规治疗的基础上,加入针灸治疗。结果治疗组患者有效率(94.44%)明显高于对照组有效率(72.22%),经临床统计学分析,有显著性差异(P<0.05)。结论针灸配合临床药物治疗神经衰弱疾病,其临床取得满意疗效,无不良反应,值得临床长期推广应用。     

内路小梁切开联合白内障超声乳化吸除术治疗原发性开角型青光眼的短期疗效

目的 对比内路小梁切开联合白内障超声乳化吸除术和单纯内路小梁切开术治疗合并白内障的原发性开角型青光眼（primary open angle glaucoma，POAG）的早期手术效果和并发症。设计 病例对照研究。研究对象2018年3月至2019年3月在北京同仁眼科中心住院手术治疗的合并白内障的POAG患者36例（36眼）。方法 根据患者选择的手术方式，将患者分为两组：行内路小梁切开联合白内障超声乳化吸除术的POAG患者12例（12眼）为观察组，行内路小梁切开术的POAG患者24例（24眼）为对照组。记录并比较两组患眼术前、术后1、2、3、5天、1周、1个月时眼压、视力、并发症和使用降眼压药的种类。主要指标 眼压、视力、并发症和降眼压药物种类。结果 观察组和对照组术前最高眼压分别为（34.08±8.21）mmHg和（37.75±8.61）mmHg（P=0.46）,观察组术后第1天、1周、1个月的平均眼压分别为（20.75±11.89）mmHg、（21.23±8.72）mmHg、（15.00±2.22） mmHg，对照组术前、术后第1天、1周、1个月的平均眼压分别为（16.08±5.63）mm Hg、（22.08±11.48）mmHg、（16.05±5.25）mmHg，两组术后眼压整体变化趋势随时间降低（P<0.001）,两组间术后眼压波动虽然存在差异，但是无统计学意义（P=0.77）。观察组和对照组术后1个月降眼压幅度分别为53.09%±15.19%和55.68%±15.63%（P=0.64）。观察组和对照组术前降眼压药物使用种类分别为（2.67±0.99） 种和（3.08±0.72）种（P=0.16），术后1个月时分别下降至（0.17±0.58）种和（0.96±1.33）种（P=0.06）。观察组和对照组组术后眼压反跳发生率分别为16.67%和54.17%（P=0.03）。前房积血为术后最常见的并发症, 观察组发生率为83.3%，对照组为62.5%（P=0.21)。观察组和对照组睫状体脱离发生率分别为25.0%和75.0%（P=0.004）。观察组中，8眼（66.7%）术后生活视力提高，4眼（33.3%）视力恢复至术前水平，0眼视力降低；对照组中，20眼（83.3%）术后生活视力恢复至术前水平，4眼（16.7%）视力降低（P<0.001）。结论 对于合并白内障的开角型青光眼患者，内路小梁切开联合白内障超声乳化吸除术和单纯内路小梁切开术在术后1个月时降眼压作用相似。联合手术术后视力改善明显，眼压反跳发生率较单纯内路小梁切开术低，是一种安全有效的手术方式，远期效果有待进一步观察。     

Alantolactone,a sesquiterpene lactone,inhibits breast cancer growth by antiangiogenic activity via blocking VEGFR2 signaling

Alantolactone (ALA), a sesquiterpene lactone isolated from several medicinal plants such as Inula helenium, has been identified to have attractive anticancer activity. However, its role in the inhibition of angiogenesis during tumor development remains unclear. In this study, we found ALA can inhibit the proliferation, motility, migration, and tube formation of human umbilical vein endothelial cells. ALA also restrained angiogenesis in chick embryo chorioallantoic membrane and delayed the growth of human MDA‐MB‐231 breast cancer xenograft in mice through angiogenesis inhibition. Furthermore, ALA suppressed the phosphorylation of vascular endothelial growth factor receptor 2 and its downstream protein kinase including PLCγ1, FAK, Src, and Akt in endothelial cells. Taken together, the antiangiogenic activity of ALA and its molecular mechanism are identified for the first time, indicating that ALA may be a potential drug candidate or lead compound for antiangiogenic cancer therapy.
