目的探讨Fas/Fas1介导的外周血淋巴细胞凋亡在病毒性心肌炎发病中的作用。方法选择临床诊断病毒性心肌炎患儿和正常小儿各25例,采用Annexin V/PI双参数法经流式细胞仪定量检测两组小儿外周血淋巴细胞凋亡百分率,并用双色流式细胞仪检测外周血淋巴细胞Fas、Fas1蛋白表达。结果心肌炎患儿外周血淋巴细胞凋亡率[(0.6976±0.1109)%]较正常对照组[(0.1288±0.1069)%]明显升高(t=18.459,P〈0.001),且与肌酸激酶同工酶、乳酸脱氢酶、肌酸激酶、羟丁酸脱氢酶升高程度均呈正相关(r=0.817,P〈0.01;r=0.785,P〈0.01;r=0.726,P〈0.01;r=0.712,P〈0.01);心肌炎患儿外周血淋巴细胞Fas、Fas1蛋白表达[(2.8804±0.302O)、(4.4496±0.4836)]较对照组[(2.1300±0.6200)、(0.1238±0.0384)]明显增加(t=17.321,P〈0.001;t’=44.577,P〈0.001);心肌炎患JLgb周血淋巴细胞凋亡百分率与Fas、Fas1蛋白表达均呈正相关(r:0.972,P〈0.01;r=0.958,P〈0.01)。结论病毒性心肌炎患儿外周血淋巴细胞凋亡增加,可能为细胞免疫功能下降的原因之一,且与心肌损害程度相关。Fas、Fas1基因在病毒性心肌炎患儿外周血淋巴细胞凋亡中起着重要作用。  相似文献

目的 研究小儿急性肾小球肾炎患儿超氧化物歧化酶(SOD)、脂质过氧化物(LPO),维生素E(VitE)含量的变化及其在治疗上的意义。方法 采用放射免疫分析方法监测血SOD;LPO则采用改奶TBA法测定;VitE应用高压液相法测定。结果 1.肾炎患儿急性期SOD活力降低与恢复期及正常组比较差异显著(P<0.01),SOD在恢复期活力明显提高,但仍低于正常对照组(P<0.01)。2.LPO急性期显著升高与对照组相比非常显著(P<0.01),恢复期LPO下降,但尚未能正常水平(P<0.01)。3.肾炎患儿血VitE浓度显著低于正常对照组(P<0.01)。结论 氧化-抗氧化功能失衡参与了肾脏损伤及恢复,可为临床治疗提供依据。  相似文献

本文测定了22例肾小球疾病患儿血及尿中可溶性白细胞介素2受体(sIL-2R)含量,结果显示疾病组急性期血及尿液sIL-2R明显高于对照组(P<0.01),且血与尿液sIL-2R含量有明显相关性(P<0.01),恢复期sIL-2R显著下降,与急性期相比有显著意义(P<0.01)。提示sIL-2R为评价肾小球疾病免疫失衡的一项重要临床参考指标。  相似文献

目的 观察增生性肾炎患者肾小球系膜细胞(MC)在不同增生程度时细胞周期调控蛋白表达水平的变化。方法 采用免疫组织化学技术,检测34例肾不小球肾炎患者肾刺标本中细胞正性调控蛋白周期素D1(cyclinD1)、负性调控蛋白P21^CIP1(P21)、P27^KIP1(P27)和增殖细胞核抗原(PCNA)表达。结果 在正常对照及非增生性肾炎组中,cyclin D1、P21不表达,P27呈高表达。在增生性肾炎组中,cyclin D1、P21和P27阳性表达主要在系膜区,随着系膜增生程度的加重,肾小球cyclinD1、P21表达增加,并与PCNA呈正相关(P均<0.01);P27则表达减少并与PCNA呈负相关(P<0.05);肾小球P27表达还与血肌酐之间呈显著性负相关(P<0.01)。结论 在人类增生性肾炎中,cyclin D1、P21和P27参与MC的增生;P27在肾组织的表达量可能与疾病进展密切相关。  相似文献

目的 探讨外周血单个核细胞(PBMC)呼吸道病毒感染与激素敏感型单纯性肾病综合征(SRSNS)发病的相关性。方法 采用逆转录-聚合酶链反应(RT-PCR)、APAAP法和ELISA法,分别对42例SRSNS患儿(活动期24例、缓解期18例)、20例肾炎性肾病、25例毛细支气管炎和19例健康对照儿童单个核细胞(PBMC)呼吸道病毒基因、病毒抗原及血清病毒抗体进行检测。结果 (1)SRSNS患儿活动期组PBMC中呼吸道病毒基因、病毒抗原及血清病毒抗体检出率均高于RSNS患儿缓解期组、肾炎性肾病组及正常对照组,差异有显著意义(P<0.05)。(2)SRSNS患儿活动期组PBMC中呼吸道合细胞病毒检出率最高,其次为流感病毒。(3)SRSNS患儿病毒基因、抗原的表达检出率与激素应用无关,血清病毒抗体的检出率降低与激素应用有关。(4)病毒基因与病毒抗原检查结果有较好一致性(x^2=5.714,P<0.05)。结论 激素敏感型单纯性肾病活动期患儿PBMC中存在呼吸道感毒。  相似文献

激素耐药型和激素依赖型肾病患儿肾脏病理计量分析   总被引:43,自引:3,他引:40  
目的 探讨病理计量分析评价激素耐药型(SR)和激素依赖型(SD)肾病综合征患儿的肾脏病理损害及其临床应用意义。方法 采用自行研制的评分法,对73例SR型和SD型肾病患儿的肾脏病理从病理类型、肾小球病变、小管间质病变、肾脏总的病理损害4个方面计量分析,并以血尿素氮为应变量,临床表现和肾脏病理计分为自变量进行逐步回归分析。结果 (1)病理类型评分结果:微小病变、局灶节段肾小球硬化、膜性肾病、系膜毛细血管性肾小球肾炎和系膜增生性肾小球肾炎五组间的年龄、病程、复发次数、血尿素氮、胆固醇、白蛋白、尿蛋白定量、肾脏总的病理损害的差异均无显著意义,而五组患儿的肾小球病变计分分别为1,5,5,6,5,微小病变组与其他各组比较差异有显著意义(H=19.278,P<0.01)。(2)73例患儿肾小球病变计分结果:正常2例(3%),轻度53例(73%),中度17例(23%),重度1例(1%)。且正常、轻度、中度三组间血尿素氮、小管间质病变的差异有显著意义(H=8.40,P,0.01;H=11.56,P<0.05)。(3)肾小管间质病变计分结果:轻度23例(34%),中度37例(18%),且轻、中、重三组间病程、复发次数、血尿素氮、肾小球病变计分的差异均有显著意义。(H值分别为25.016,38.775,14.944,10.625,P值均<0.01)。(4)肾脏损害的总分结果:轻度33例(45%),中度34例(7%),重度6例(8%),轻、中、重三组间病程、复发次数、血尿素氮的差异有显著意义(H值分别为19.42,14.335,18.923,P值均<0.01)。(5)以血尿素氮为应变量行逐步回归分析,小管间质及肾小球损害对血尿素氮均有显著影响,小管间质病变的回归系数为0.862(P<0.01);肾小球病变的回归系数为0.212(P<0.05)。结论 采用评分法对肾脏病理进行主分评价其病变程度,具有临床实用性,尤其对小管间质病变的计分评价对判断肾功能受损更有价值。  相似文献

目的 探讨保肾录对尿微量白蛋白(mALB)、尿转铁蛋白(TRF)、视黄醇结合蛋白(RBP)、β2-微球蛋白(β2-MG)的影响与预防紫癜性肾炎的价值。方法 88例过敏性紫癜分为一般治疗组和保肾录治疗组,定期观察尿上述四项蛋白与尿白常规的变化。结果 治疗3个月时,对照组与治疗组尿mALB、TRF、β2-MG比较有显著性差异(P<0.05,P<0.01,P<0.01)。治疗6个月时对照组与治疗组尿mALB、TRF比较仍有显著性差异(P<0.05、P<0.01)。治疗3、6个月时尿常规异常人数具有显著性差异(P均<0.05)。结论 保肾康可缩短尿mALB、TRF、β2-MG升高的时间,降低紫癜性肾炎的发生率。  相似文献

目的探讨儿童乙型肝炎病毒相关性肾炎(HBV-GN)的临床和病理特点。方法收集经肾活检确诊为HBV—GN的74例患儿入院时临床表现(水肿、血压)及实验室检查结果(尿常规、24h尿蛋白定量、乙肝5项、血清清蛋白、血HBVDNA等),并对其肾组织进行光镜(常规染色及特殊染色)及免疫组织化学检查(IgG、IgA、IgM、C3e、C4e、HBsAg、HBcAg)。根据血清HBVDNA水平不同将74例HBV—GN患儿分为高病毒载量组(HBVDNA〉1.0×10^8copies/L)58例和低病毒载量组(HBVDNA〈1.0×10^8copies/L)16例,并比较分析其临床病理特点。结果临床表现:肾病综合征(NS)52例(70.3%);血尿和蛋白尿13例(17.6%);单纯性蛋白尿6例(8.1%);单纯性血尿3例(4.1%)。病理分型:膜性肾病(MN)56例(75.7%);系膜增生性肾小球肾炎(MsPGN)12例(16.2%);膜增生性肾小球肾炎(MPGN)5例(6.6%);毛细血管内增生性肾小球肾炎(EnPGN)1例。高病毒载量组与低病毒载量组入院时血清清蛋白[(22.04±7.69)g/L vs(31.95±5.83)g/L],C3[(0.759±0.252)g/L vs (0.986±0.330)g/L]比较均有统计学差异(Pa〈0.05)。高病毒载量组病理类型多样,而低病毒载量组相对单一,以MsPGN、MN为主。结论HBV—GN患儿学龄期多见,且男多于女,临床表现以NS为主,病理类型以MN多见,血清HBVDNA水平与HBV—GN病变密切相关。  相似文献

目的 探讨白细胞介素10(IL-10)在急性肾小球肾炎(AGN)发病机制中的作用及其临床意义。方法 应用逆转录-多聚酶链反应(RT-PCR)及双抗体夹心ELISA法检测了33例AGN患儿外周血单个核细胞(PBMC)IL-10mRNA和蛋白水平。结果 1.AGN急性期IL-10 mNRA和蛋白水平较恢复期和正常对照组显著升高,恢复期降至正常水平。2.IL-10mRNA和蛋白水平与血清补体C3呈显著负相关。结论 AGN患儿存在Th1/Th2细胞免疫功能的失衡。PVMC IL-10可作为观察病情活动状态的免疫学指标之一。  相似文献

目的 探讨肾病综合征(NS)儿童外周血单个核细胞(PBMC)中活化蛋白1(AP-1)和糖皮质激素受体(GR)的DNA结合活性的变化,体外实验加入地塞米松(DEX)以及泼尼权治疗后对二者的影响。方法 用凝胶电泳迁移率变化分析法(EMSA)和同位素放射性自显影法分析了NS儿童PMBC中AP-1、GR的DNA结合活性。结果 (1)NS组基础状态AP-1DNA活性为264.1(0.6-351.8),对照组为0.8(0-1.1),两组比较差异有非常显著意义(Z=2.72,P<0.01);佛波酯刺激状态下AP-1DNA活性为514.8(1.1-616.7),对照组为1.1(0-36.6),两组比较差异有非常显著意义(Z=2.64,P<0.01);经DEX作用后NS组AP-1的DNA活性与对照组比较,差异无显著意义(Z=0.64,P>0.05)。(2)NS组基础状态GR的DNA活性为7.6(0.2-11.7),对照组为45.2(8.1-95.2),两组比较差异有显著意义(Z=2.56,P<0.05);TPA刺激状态GR的DNA活性为5.3(0.0-10.7),对照组为78.1(7.8-97.4),两组比较差异有显著意义(Z=2.40,P<0.05),经DEX作用后GR的DNA活性与对照组比较,差异无显著意义(Z=0.96,P>0.05)。(3)经泼尼松治疗尿蛋白转阴1周后,AP-1 DNA活性为1.05(0.95-1.44),与治疗前[2.87(1.00-4.20)]比较,差异有显著意义(Z=2.24,P<0.05)。治疗后GR的DNA活性为3.91(1.00-4.70),与治疗前[1.04(0.73-1.24)]比较,差异有显著意义(Z=2.0,P<0.05)。结论 (1)NS儿童的PBMC中AP-1 DNA结合活性异常升高,而GRDNA结合活性降低;(2)糖皮质激素可抑制AP-1、增强GR的DNA结合活性。  相似文献

Methods??Analyze the clinical manifestations??pathological features??therapies??prognosis of patients who were diagnosed with C3 glomerulonephritis from September??2010 to June??2016 retrospectively. Results??Clinical characteristics and laboratory examination??2 patients’ first symptom was hematuria??4 patients’ first symptom was hematuria and proteinuria??3 patients presented as acute nephritic syndrome??one presented as nephrotic syndrome. All the patients showed that the level of serum complement C3 was reduced??while sernm complement C4 was normal. Pathological character??6 patients showed strong positive complement C3 deposition under immunofluorescence. Lightmicroscopy showed mesangial proliferative glomerulonephritis in 5 cases??1 case was diagnosed as endocapillary proliferative glomerulonephritisin??and 3 patients presented electrondense depositionin under electron microscope. Treatment and prognosis??after conventional treatment??2 patients who were with crescent were treated with glucocorticoid .After a follow-up from 6 months to 42 months ??the prognosis was pretty good. Conclusion??Children with C3GN are usually presented with hematuria and ??or??proteinuria??characterized by strong positive C3 deposition. Lightmicroscopy always shows mesangial proliferative glomerulonephritis. Electron microscope show electron dense deposition??and short-term prognosis is pretty good.  相似文献

目的 探讨硫化氢 (H2 S)对低氧性肺动脉高压 (HPH)大鼠肺动脉结构重建和肺动脉平滑肌细胞增殖、凋亡的调节作用。方法 选取体重 180~ 2 0 0g的健康雄性Wistar大鼠 2 4只 ,随机分为常氧组、低氧组、低氧 硫氢化钠 (NaHS)组。监测其血液动力学变化及肺动脉结构重建 ,采用免疫组织化学方法检测增殖细胞核抗原 (PCNA)和Bcl 2凋亡蛋白表达 ,并对肺动脉平滑肌细胞增殖与凋亡进行定位和半定量分析。结果 低氧组大鼠平均肺动脉压 (mPAP)明显升高 (P <0 .0 1) ,右室 / (左室 室间隔 )比值显著增加 (P <0 .0 1) ,肺动脉相对中膜厚度 (RMT)和相对中膜面积 (RMA)增加 (P均 <0 .0 1) ;NaHS可明显降低肺动脉压力及抑制肺动脉重建 (P <0 .0 1)。低氧组大鼠肺小动脉平滑肌细胞PCNA和Bcl 2凋亡蛋白表达分别较对照组增高 (P <0 .0 1) ;NaHS可明显下调肺小动脉平滑肌细胞PCNA和Bcl 2蛋白表达 (P <0 .0 1)。结论 新型气体信号分子H2 S可降低肺动脉压力 ,抑制肺动脉平滑肌细胞增殖 ,在HPH及肺血管结构重建的形成中发挥重要作用  相似文献

目的 观察卡维地洛对免疫性心肌炎小鼠心肌凋亡蛋白Bcl-2、Bax及Fas的影响,探讨其在免疫性心肌炎中的作用.方法 60只4~5周龄近交系雄性BALB/C小鼠,随机将小鼠分为3组:免疫性心肌炎模型组(模型组),卡维地洛干预组(干预组),空白对照组(对照组),每组20只.眼球取血后处死,留取心脏及血液标本.光镜观察各组小鼠心肌组织病理学改变,电镜观察心肌细胞超微结构,化学发光免疫法检测血清心肌肌钙蛋白I(cTnI)水平;免疫组化法检测心肌Bcl-2、Bax及Fas蛋白表达的含量;原位缺口末端标记(TUNEL)法检测心肌细胞凋亡情况.结果 模型组小鼠心肌细胞光镜下见大量炎症细胞浸润,电镜下可见细胞核固缩、染色质边集,对照组小鼠心肌细胞光镜下未见炎症细胞浸润,电镜下染色质边集不明显.模型组Bcl-2、Bax及Fas蛋白表达较对照组高(23.48±2.24 vs 6.64±1.60,26.15±2.02 vs 5.09±0.85,21.22±3.62 vs 5.86±1.37,P<0.01);干预组与模型组比较,小鼠心肌组织炎症积分轻(2.60±0.3l vs 2.02±0.26,P<0.05),电镜下细胞核固缩轻,心肌细胞Bcl-2、Bax及Fas蛋白表达低(17.13±1.94 vs 23.48±2.24,17.66±2.62 vs26.15±2.02,16.79±2.83 vs 21.22±3.62,P<0.05),凋亡指数低[(16.61±4.67)%vs(24.51±4.70)%,P<0.05],cTn-I水平低[(1.878±0.48)ng/ml vs(1.102±0.23)ng/ml,P<0.05].结论 卡维地洛可减轻免疫性心肌炎小鼠心肌细胞凋亡,对免疫性心肌炎起一定的保护作用.
Abstract:
Objective To observe the effects of carvedilol on the expression of Bcl-2, Bax and Fas in autoimmune myocarditis (AM).Methods A total of 60 inbred male BALB/C mice 4-5 weeks of age were divided at random into 3 groups as follows: AM group ( n = 20), carvedilol group ( n = 20 ) and control group (n = 20).The mice were sacrificed after gathering blood specimens by taking out the eyeballs and hearts tissue.The histological and ultrastructural changes were observed under light microscope and electron microscope.The concentrations of cardiac troponin Ⅰ (cTn Ⅰ ) were detected by chemiluminescence immunoassay (CLIA).Immunohistochemistry (IHC) was performed to analyze the contents of Bcl-2, Bax and Fas, TUNEL to detect the apoptotic index in myocardial cells.Results There were large number of lymphocyte and monocyte infiltrates under light microscope and karyopyknosis and chromatin gathered along the nuclear membrane under electron microscope in AM group.There were no inflammations and chromatin gathering in group C.Compared with control group, the Bcl-2, Bax and Fas protein expression significantly elevated in AM group ( 23.48 ± 2.24 vs.6.64 ± 1.60,26.15 ± 2.02 vs.5.09 ± 0.85,21.22 ± 3.62 vs.5.86 ± 1.37, P < 0.0l ) . The histopathologic scores ( 2.60 ± 0.31 vs.2.02 ± 0.26, P < 0.05 ) and karyopyknosis of carvedilol group decreased as compared with AM group.The Bcl-2, Bax and Fas protein expression (17.13 ±1.94 vs.23.48 ±2.24,17.66 ±2.62 vs.26.15 ±2.02,16.79 ±2.83 vs.21.22 ±3.62, P < 0.05 ), AI [( 16.61 ± 4.67 ) % vs.( 24.51 ± 4.70) %, P < 0.05]and contents of cTnI [( 1.878± 0.48 ) ng/ml vs. ( 1.102 ± 0.23 ) ng/ml, P < 0.05]also decreased in carvedilol group compared with AM group.Conclusion Carvedilol could protect against AM by alleviating cardiomyocyte apoptosis.  相似文献

Ninety five children with chronic glomerulonephritis were studied to analyze the correlation between hypertension and functional deterioration in each of the following histology groups; minor glomerular abnormalities (MGA), proliferative glomerulonephritis (PGN), membrano-proliferative glomerulonephritis (MPGN), membranous glomerulonephritis (MGN) and focal segmental glomerulosclerosis (FSGS). Among the 95 patients, 14 patients revealed to be hypertensive. All patients with MGA and MGN revealed normal blood pressure. There were no statistically significant differences in the prevalence of hypertension in patients with PGN, MPGN and FSGS. The overall incidence of renal insufficiency was significantly higher in the hypertensives (50.0%) than in the normotensives (4.9% p less than 0.0001). The incidences of renal insufficiency were significantly higher in the hypertensives than in the normotensives in patients with PGN (P less than 0.01) and MPGN (P less than 0.05). These data suggest that there is a significant correlation between functional deterioration and hypertension, and that the hypertensive patients with PGN and MPGN are at risk for progression to renal insufficiency.  相似文献

目的 探讨外周血单个核细胞 (PBMC)呼吸道病毒感染与糖皮质激素敏感型单纯性肾病综合征 (SSNS)的发生和反复的关系。方法 采用免疫组化、原位杂交、酶联免疫吸附和病毒分离技术 ,对青岛大学医学院附属医院儿科和滨州医学院附属医院 2 0 0 0年 1月至 2 0 0 2年 12月收治的 36例初发组、15例反复组和 2 1例缓解组SSNS患儿检测了PBMC病毒分子表达和血清中病毒特异性IgM抗体水平。结果 初发组与反复组比较 ,PBMC中呼吸道合胞病毒抗原和肠道病毒基因表达的阳性率差异均无显著性 ;而初发组与缓解组和对照组比较及反复组与缓解组和对照组比较 ,PBMC中两种病毒感染的阳性率差异均有显著性 (P <0 0 5 ,P <0 0 1)。初发组与其他各组比较 ,不同病毒的特异性IgM抗体阳性率明显增高 ( P <0 0 5 ) ;而反复组与缓解组和对照组比较 ,多种病毒的特异性IgM抗体的阳性率差异均无显著性。 结论 初发和反复SSNS患儿的PBMC均易被病毒感染 ,糖皮质激素的应用并不影响细胞内病毒抗原和基因的表达 ,但可使病毒特异性IgM抗体的产生明显减少 ;PBMC内病毒抗原或基因表达阳性可做为SSNS有反复倾向的重要分子标志之一。  相似文献

目的 探讨Bcl-2、Fas蛋白在不完全梗阻性黄疸大鼠胆总管的表达及其与胆总管上皮细胞凋亡的关系.方法 60只SD大鼠,雌雄不拘,随机分为不完全梗阻性黄疸组40只(A组)和实验对照组20只(B组).A组不完全结扎大鼠胆总管建立动物模型,B组仅打开腹腔游离胆总管末端即可.二组分别于术后3、7、14、21 d处死10只和5只,检测血清胆红素(TBIL)的变化,观察胆总管病理组织学的改变,TUNEL法观察胆总管上皮细胞凋亡,免疫组化法检测大鼠胆总管上皮中Bcl-2、Fas蛋白的表达.结果 A组随着梗阻性黄疸时间的延长,血清胆红素逐渐增高,胆总管上皮细胞凋亡数逐渐增多;B组变化不明显.A组与B组比较有显著性差异(P<0.05).A组Bcl-2、Fas蛋白在结扎后3 d均为弱阳性表达,7、14 d均为阳性表达,21 d均为强阳性表达;B组均为阴性表达.结论 不完全梗阻性黄疸时细胞凋亡增加是机体维持自身稳定的一个重要机制.Fas、Bcl-2蛋白对不完全梗阻性黄疸大鼠胆总管上皮细胞凋亡有调节作用.  相似文献

The glomerulonephritides are a group of disorders characterized by intraglomerular inflammation, manifested clinically by hypertension, haematuria and oedema. The disease severity varies from asymptomatic microscopic haematuria to rapidly progressing renal failure. The common causes of glomerulonephritis (GN) in children include acute post streptococcal GN (APSGN), Henoch-Schönlein purpura nephritis (HSPN) and IgA nephropathy (IgAN). Focussed clinical evaluation and key investigations are vital in establishing a timely diagnosis. Although many patients with GN require minimal medical input, a sub-section require urgent nephrology input for life-saving interventions. Prognosis depends on the underlying cause of GN. APSGN and HSPN are associated with excellent prognosis in the vast majority of affected children whereas majority of those presenting with rapidly progressive crescentic nephritis are likely to develop chronic kidney disease.  相似文献

Heparin has an apoptotic effect beside its anticoagulant, anti-inflammatory, antihypertensive, and antiproliferative effects. In this study, the authors detected the percentages of apoptotic lymphoblasts and the expressions of apoptotic Fas protein and antiapoptotic Bcl-2 protein with flow cytometry in vitro after the incubation of lymphoblasts with heparin. Eleven newly diagnosed acute lymphoblastic leukemia (ALL) children were included in the study. Lymphoblasts were incubated in all different levels of heparin concentrations (0, 10, and 20 U/mL) and the percentages of apoptotic lymphoblasts and the percentages of Fas protein and Bcl-2 proteins were simultaneously measured by flow cytometry at 0, 1, and 2 h. At 0, 1, and 2 h, apoptosis was determined when heparin was added in 10- and 20-U/mL concentrations (p <.05). The apoptotic effect of heparin on lymphoblasts was higher at the first hour than at 0 and 2 h in 10- and 20-U/mL heparin concentrations (p <.01). The highest apoptosis was detected in the 20-U/mL heparin concentration at the first hour. The expression levels of Fas protein on lymphoblasts were higher at the first hour than at 0 and 2 h in 10- and 20-U/mL heparin concentrations (p <.001). The highest expression of Fas protein was observed in the 20-U/mL heparin concentration at the first hour. The expression levels of Bcl-2 protein on lymphoblasts were lower at the first hour than at 0 and 2 h in 10- and 20-U/mL heparin concentrations (p <.001). The lowest expression of Bcl-2 protein was detected in the 20-U/mL heparin concentration at the first hour. Increased concentrations of heparin had an increasing effect on the percentages of apoptotic lymphoblasts. The expression percentages of Fas protein on lymphoblasts also increased, whereas the expression percentages of Bcl-2 protein on lymphoblasts decreased (p <.05). These results suggest that low-dose heparin may cause significant apoptosis of lymphoblasts in newly diagnosed ALL patients.  相似文献

目的:探讨血管紧张素Ⅱ1型受体拮抗剂(AT1RA)缬沙坦对阿霉素肾病肾硬化大鼠肾脏细胞凋亡及凋亡相关蛋白的影响。方法:36只SD大鼠随机分为模型组、治疗组和对照组。大鼠单侧肾切除加阿霉素注射诱导阿霉素肾病肾硬化模型。治疗组每日予缬沙坦(20 mg/kg)灌胃1次,共12周。对照组和模型组每日用等量生理盐水灌胃。采用TUNEL法检测肾脏细胞凋亡情况,计算出肾小球凋亡指数(GAI)和肾小管凋亡指数(TAI)。免疫组化法检测肾组织Fas和FasL表达。光镜下观察肾组织病理改变,并计算肾小球硬化指数(GSI)。结果:模型组较对照组肾小球硬化明显,GSI高于对照组(P<0.01);而治谱镚SI较模型组降低,但仍高于对照组(P<0.01)。模型组和治疗组GAI和TAI较对照组显著增高(P<0.01);而治疗组GAI和TAI均低于模型组(P<0.01),但仍高于对照组(P<0.01)。模型组和治疗组的Fas和FasL表达较对照组亦明显增强(P<0.01),其中治疗组低于模型组(P<0.01)。 结论:AT1RA缬沙坦可能通过降低凋亡相关蛋白Fas及FasL在肾组织表达而抑制肾脏细胞过度凋亡,从而发挥其延缓肾小球硬化的作用。  相似文献

目的 探讨Bcl-2、Fas蛋白在不完全梗阻性黄疸大鼠胆总管的表达及其与胆总管上皮细胞凋亡的关系.方法 60只SD大鼠,雌雄不拘,随机分为不完全梗阻性黄疸组40只(A组)和实验对照组20只(B组).A组不完全结扎大鼠胆总管建立动物模型,B组仅打开腹腔游离胆总管末端即可.二组分别于术后3、7、14、21 d处死10只和5只,检测血清胆红素(TBIL)的变化,观察胆总管病理组织学的改变,TUNEL法观察胆总管上皮细胞凋亡,免疫组化法检测大鼠胆总管上皮中Bcl-2、Fas蛋白的表达.结果 A组随着梗阻性黄疸时间的延长,血清胆红素逐渐增高,胆总管上皮细胞凋亡数逐渐增多;B组变化不明显.A组与B组比较有显著性差异(P<0.05).A组Bcl-2、Fas蛋白在结扎后3 d均为弱阳性表达,7、14 d均为阳性表达,21 d均为强阳性表达;B组均为阴性表达.结论 不完全梗阻性黄疸时细胞凋亡增加是机体维持自身稳定的一个重要机制.Fas、Bcl-2蛋白对不完全梗阻性黄疸大鼠胆总管上皮细胞凋亡有调节作用.  相似文献
