The objective of this study is to express the carbohydrate recognition domain (CRD) of the asialoglycoprotein receptor (ASGPR) H1 and H2 subunits of Marmota himalayan in vitro, and develop polyclonal antibodies against the recombinant proteins. RT-PCR was used to amplify ASGPR CRDH1 and CRDH2 from the liver tissue of Marmota himalayan. The products of amplification were subcloned into prokaryotic expression vector pRSET-B, and expressed in E.coli BL21(DE3)plysS. The recombinant proteins were purified using Ni-NTA spin column. The purified proteins were inoculated into BALB/c mice to develop polyclonal antibodies. The sensitivity and specificity of antibodies were evaluated by enzyme-linked immunosorbent assay (ELISA), Western blotting and immunohistochemical staining (IHC). The polyclonal antibodies showed high sensitivity and specificity against both denaturated and native ASGPR proteins. We successfully amplified and expressed the ASGPR CRDs of Marmota himalayan. The nucleic sequences of ASGPR CRDH1 and CRDH2 of Marmota himalayan have been submitted to Genbank and the sequence ID are DQ 845465 and DQ845466, respectively. The proteins and antibodies prepared can be used for targeting gene therapy in a new animal model-Marrnota himalayan—— for the research of infectious diseases of hepatitis viruses and liver cancer treatment.  相似文献

The pharmacological effect about the recipe of ancient Chinesemedicine and herb drugs—Gegen Qinlian Decoction(GGQLD)was studied.The results showed that it could markedly lower the body temperature of feverishrabbits caused by infection of pentavaccine.The effect is as strong as that ofAspirin compound(APC),there is no statistical significance between animaltreated by GGQLD and those by APC.It could also inhibit the growth of manykinds of bacteria(Staphylococcus;Bacillus dysenteriae;Pneumococcus)in vivoand in vitro.It was observed that the percentage of ANAE~+ lymphocytes in thespleen and lymphonodes and the number of PFC in the spleen of mouse could beincreased by GGQLD.Meanwhile,the phagocytic effect of peritoneal macrophagesin mice treated by GGQLD was strengthened,which suggested that the antifebrileand antibiotic effects of this medicine are partly related to its efficiency to enhancethe immunity against pathogens.  相似文献

using a WZD—1 microcirculation microscope,we observed the tongue tipand nail-bed microcirculation of uremic patients with deficiency of kidney-yangbefore and after blood dialysis.The results showed that the pathological basis ofuremia with deficiency of kidney-yang may be microcirculation disturbance,in-crease of metabolites and water retention.The effusion from capillary loops intongue tip may be one of the pathogenetic factors of a swollen and jaggedtongue.The flow rate of blood in capillary loops in the nail-bed was apparentlyabnormal and markedly improved after blood dialysis.The weighted integral ofnail-bed microcirculation decreased from moderately abnormal (4.99±1.52) toapproximately normal (1.98±0.97) (P<0.01).Microcirculation seems to be a sensi-tive criterion for reflecting the pathologic changes of uremia with deficiency ofkidney-yang.  相似文献

The effect of Batroxobin on spatial memory disorder of left temporal ischemic rats and the expressionof HSP32 and HSP70 were investigated with Morri's water maze and immunohistochemistry methods.The results showed that the mean reaction time and distance of temporal ischemic rats in searching agoal were significantly longer than those of the sham-operated rats and at the same time HSP32 andHSP70 expression of left temporal ischemic region in rats was significantly increased as compared withthe sham-operated rats.However,the mean reaction time and distance of the Batroxobin-treated ratswere shorter and they used normal strategies more often and earlier than those of ischemic rats.Thenumber of HSP32 and HSP70 immune reactive cells of Batroxobin-treated rats was also less than thatof the ischemic group.In conclusion,Batroxobin can improve spatial memory disorder of temporalischemic rats;and the down-regulation of the expression of HSP32 and HSP70 is probably related tothe attenuation of ischemic injury.  相似文献

The guanine and cytosine content of some pathogenic species and varieties of Cryptococcus was reported here as determined by a thermal denaturation technique. The results range from 47.82 to 61.98, which are similar to those in the literature. This study indicated that different species of Cryptococcus and different varieties of C. neoformans seem to be reflected in their G+C mol% content values. Especially the differentiation of GC% of both C neoformans var. Shanghainess and C. neoformans var. gatti were over the range of variation that permitted in this species (1-4%)..The determination of G+C mol% content can raise the level of taxonomy of Cryptococcus from morphology to nucleotide molecule, and the value is not affected by environment and etc. It is a standard of heredity for the classification and identification of fungi.  相似文献

The expression of interleukin-17(IL-17) in lung and peripheral blood of asthmatic rats and the influence of dexamethasone,and the role of IL-17 in the pathogenesis of asthma were investigated.Thirty Sprague-Dawley(SD) adult rats were randomly divided into three groups(n=10 in each group):normal group,asthmatic group,and dexamethasone-interfered group.Rat asthmatic model was established by intraperitoneal(i.p.) injection of 10% ovalbumin(OVA) and challenge with 1% OVA via inhalation.Rats in dexamethasone-interfered group were pretreated with dexamethasone(2 mg/kg,i.p.) 30 min before each challenge.The expression of IL-17 protein in serum and bronchoalveolar lavage fluid(BALF) was detected by ELISA.The expression of IL-17 mRNA in peripheral blood mononuclear cells(PBMC) and BALF cells was semi-quantitatively detected by RT-PCR.The expression of IL-17 protein in serum and BALF of asthmatic rats was significantly elevated as compared with normal rats and dexamethsone-interfered rats(P<0.01),and there was significant difference between normal rats and dexamethsone-interfered rats(P<0.05).The expression of IL-17 mRNA in PBMC and BALF cells of asthmatic rats was markedly increased as compared with normal rats and dexamethsone-interfered rats(P<0.01),and significant difference was found between normal rats and dexamethsone-interfered rats(P<0.05).It was concluded that the expression of IL-17 was increased significantly in asthmatic rats and could be inhibited partly by dexamethasone,suggesting that IL-17 might play an important role in the pathogenesis of asthma as an inflammation regulation factor.  相似文献

Objective To compare the effects of autotransfusion and homologous transfusion on milieu interieur and secretion of erythropoietin (EPO). Methods Thirty patients aged 8-21 yr, undergoing surgical correction of kyphoscoliosis were randomly divided into two groups of fifteen:group A and group B. In group A patients received homologous transfusion during operation, while in group B shed blood was collected from operative field and after filtration RBCs were separated by a differential centrifugation and washed in normal saline and reinfused. Anesthesia was induced with fentanyl 2 μg· kg-1 ,propofol 1 mg·kg-1 and vecuronium 0.1 mg·kg-1 and maintained with inhalation of isoflurane and 50 % N2O in oxygen and intermittent iv boluses of vecuronium and fentanyl. Blood routine, blood gases, electrolytes, plasma osmotic pressure and activated clotting time (ACT) were measured before operation, 2 h after skin incision and at the end of surgery.
