Objective: The objective of this study is to investigate the inhibitory effect of peony and licorice decoction and its compatibility components on the Nav1.4 voltage?gated sodium channels (VGSCs). Materials and Methods: Writhing test was carried out with ICR mice. Paeonia lactiflora and Glycyrrhiza uralensis group were administrated 0.2 ml of solution of freeze?dried powder dissolved in normal saline with the concentration of 2.94 mg/ml, 1.47 mg/ml, and 0.74 mg/ml using intragastric administration, respectively. Peony and licorice decoction groups were administrated 0.2 ml of solution of freeze?dried powder dissolved in normal saline with the concentration of 5.89 mg/ml, 2.94 mg/ml, and 1.47 mg/ml using intragastric administration, respectively. For electrophysiology studies, each freeze?dried powder was dissolved in DMSO to make 10 mg/ml and 50 mg/ml stock solutions. The electrophysiological recordings were obtained under visual control of a microscope. For UPLC analysis, the freeze?dried powder was dissolved in methanol and then determines the contents of the nine marker compounds. Results: The effect of G. uralensis on incubation period and writhing frequency was significantly better than that of peony and licorice decoction group and P. lactiflora group. The inhibition rate of 50 mg/ml water extracts of the three samples was significantly higher than that of the 10 mg/ml group. Moreover, the water extract of G. uralensis at 50 mg/ml had the strongest inhibitory effect on INav1.4 of the three. Conclusion: The possible mechanism of peony and licorice decoction in relieving spasm and pain is most likely by inhibiting Voltage?Gated Sodium Channel Subtype 1.4.  相似文献

目的:优选白花蛇舌草中总三萜酸的提取纯化工艺。方法:以总三萜酸提取率为指标,选取乙醇用量、乙醇体积分数及回流时间为考察因素,采用正交试验优选白花蛇舌草中总三萜酸的提取工艺;通过静态吸附和解吸试验筛选树脂型号,采用单因素试验考察总三萜酸的大孔树脂纯化工艺。结果:最佳提取工艺为加10倍量70%乙醇回流提取1.5 h。采用AB-8型大孔树脂,其纯化工艺为上样流速2 BV.h-1,上样液pH 5,上样液质量浓度0.25 g.L-1,洗脱剂pH 7,洗脱乙醇体积分数80%,洗脱速度2 BV.h-1。结论:优选的工艺操作简单、稳定可行、重复性好,可推广于工业化生产应用。  相似文献

Objective: To observe the therapeutic efficacy of puncturing Houxi (SI 3) toward Hegu (LI 4) plus lumbar movement in treating acute lumbar sprain. Methods: A total of 212 patients with acute lumbar sprain were randomized into two groups. There were 106 subjects in the observation group who were treated with puncturing Houxi (SI 3) toward Hegu (LI 4) plus lumbar movement; the other 106 patients in the control group were treated with puncturing Houxi (SI 3) toward Hegu (LI 4) alone. Therapeutic efficacy was evaluated after 2 treatment courses. Results: The total effective rate was 95.3% in the observation group versus 88.7% in the control group, and the difference was statistically insignificant between the two groups (P>0.05). However, in comparing the treatment times for achieving recovery and effectiveness, differences between the two groups were statistically significant (both P<0.05), indicating that integrated use of acupuncture and exercise is faster in realizing therapeutic effectiveness. Conclusion: By selecting fewer acupoints, puncturing Houxi (SI 3) toward Hegu (LI 4) is effective in treating acute lumbar sprain, and with the help of lumbar movement, it can achieve even better treatment effect.  相似文献

目的:从紫玉盘Uvaria macrophylla var. microcarpa中分离具有抗癌活性成分。方法:用色谱方法分离紫玉盘中化学成分,用波谱技术(IR,ESI-MS,1H-NMR,13C-NMR,HMQC,COSY,HMBC, NOESY)进行鉴定结构。结果:从紫玉盘中分离得到4个化合物, uvarimacrins A (1), B(2)and C(4)和uvamalols D(3)。结论:化合物(1),(2),(4)均为新化合物,(3)首次从紫玉盘中分离得到。  相似文献
