目的探讨葛根素的雌激素样作用。方法采用成年雌性SD大鼠,摘除双侧卵巢,分成6组假手术组(SHAM)、去卵巢组(OVX)、17β-雌二醇替代治疗组(ERT,0.5mg.kg-1.2d-1)、高剂量葛根素组(PH,100mg.kg-1.d-1)、中剂量葛根素组(PM,50mg.kg-1.d-1)和低剂量葛根素组(PL,20mg.kg-1.d-1)。去卵巢术后1周开始皮下注射给药,连续7周。通过观察大鼠阴道涂片情况、动情周期的改变、子宫内膜的变化及子宫系数的测定来观察葛根素的雌激素样作用。结果与OVX组相比,高、中剂量葛根素能使成年去卵巢大鼠子宫增重、阴道上皮细胞发生角化并增加动情周期的发生次数,但程度明显低于SHAM组及ERT组;而低剂量葛根素组无此改变。结论葛根素具有雌激素样作用,但作用强度弱于17β-雌二醇。  相似文献

目的 研究大鼠去卵巢前、后不同时间子宫肌壁内神经肽Y免疫反应阳性神经纤维表达变化与子宫湿重变化.方法 选取雌性SD大鼠40只,随机分为假手术对照组(sham)、卵巢切除后4周组(ovx1)、卵巢切除后6周组(ovx2)、卵巢切除后8周组(ovx3),每组10只.观察各组大鼠的子宫湿重变化,并采用SABC免疫组织化学法检测神经肽Y免疫反应阳性神经纤维在各组大鼠子宫肌壁内的表达.结果 各组大鼠子宫湿重经两两比较均P<0.05,表现为:ovx3组相似文献

目的:观察长期使用葛根素对去卵巢大鼠阴道壁结构的影响,为中西药葛根素与雌激素组合治疗Ⅰ型骨质疏松症提供实验依据。方法:5月龄健康雌性大白鼠40只,分成5个实验组(n=8只),除假手术组外均建立去卵巢动物模型。①假手术组(Sham);②去卵巢模型组(OVX);③葛根素-50组(Pr-50),皮下注射葛根素50 mg/kg,1次/天;④雌二醇-200组(E2-200),皮下注射雌二醇200μg/kg,2次/周;⑤葛根素-25+雌二醇-100组(Pr-25+E2-100),皮下注射葛根素25 mg/kg,1次/天和雌二醇100μg/kg,2次/周。在第20周,取大鼠血清和阴道壁,观察血清雌二醇(E2)水平和阴道壁结构的变化。结果:OVX组E2水平、阴道上皮及肌层厚度明显低于其他各组(P﹤0.05);E2-200组E2水平、阴道上皮及肌层厚度明显高于其他各组(P﹤0.05);Pr-25+E2-100组E2水平较Pr-50组高(P﹤0.05),但两组间阴道上皮及肌层厚度差异无统计学意义(P﹥0.05)。结论:较大剂量雌二醇可明显促进去卵巢大鼠阴道上皮和肌层增生;小剂量的雌二醇与葛根素联合、较大剂量的葛根素对去卵巢大鼠阴道上皮和肌层增生作用不明显。  相似文献

目的探讨去卵巢联合高脂饮食致雌性大鼠脂肪组织炎症激活是否与成纤维细胞生长因子21(FGF21)信号通路相关。方法 30只雌性SD大鼠随机分为假手术对照组(SHAM)、去卵巢组(OVX)、去卵巢+高脂组(OVX+HFD),高脂饲料喂养28周,GTT和ITT法分析大鼠的胰岛素抵抗,检测血脂指标,观察肾周脂肪细胞的体积变化,免疫组化和蛋白印迹法分析肾周脂肪组织中的磷酸化c-Jun氨基端激酶(P-JNK)、血管内皮生长因子(VEGF)、β-Klotho和FGF21的表达。结果与SHAM组相比,OVX和OVX+HFD组大鼠出现胰岛素抵抗,血脂水平上升,肾周白色脂肪组织脂肪细胞的体积增大,P-JNK的表达也增加,VEGF、β-Klotho和FGF21的表达下调,以OVX+HFD组病理学和基因表达变化最为明显。结论雌激素缺乏可造成胰岛素抵抗、脂肪组织内炎性激活和FGF21信号通路的衰减,若联合高脂饮食则病变更加明显。  相似文献

目的:观察橄榄油对去势后大鼠骨代谢和骨密度的影响,探讨并分析橄榄油防治绝经后骨质疏松症(PMOP)的有效性及机理。方法:将30只5~6个月龄清洁型SD雌性大鼠进行随机分成四组:①假手术组(Sham组)、②去卵巢组(OVX组)、③去卵巢+橄榄油组(OVX+Olive)、④去卵巢+雌激素组(OVX+E)。治疗组用药:(OVX+Olive)组及(OVX+E)组均采用经口灌胃方式进行用药,按1 ml/100 g体重,1次/天灌胃,连续12周。12周后分别左心室取血,检测血中雌二醇(E2)、血钙、血碱性磷酸酶(ALP)、白介素-6(IL-6)水平,放血处死后取出腰椎及左侧股骨行双能X线骨密度(DEXA)测定。结果:OVX组中血E2明显低于Sham组和治疗组,差异有统计学意义(P<0.01、P<0.05),ALP、IL-6值显著高于其余三组,差异有统计学意义(P<0.05);治疗组中E2、ALP、IL-6与Sham组差异无统计学意义(P>0.05);四组中血钙值差异均无统计学意义(P>0.05)。去势橄榄油组与雌激素组骨密度平均值均较OVX组高,差异有统计学意义(P<0.05),但该两组间差异无统计学意义(P>0.05)。结论:橄榄油能有效地减轻大鼠卵巢切除术引起的骨质丢失,可能通过补充植物雌激素发挥拟雌激素样作用。  相似文献

目的:探讨雌激素对去势雌性Sprague-Dawley大鼠单核细胞趋化因子1(MCP-1)及Ras同源基因家族成员A(Rho A)水平的调控及其机制,研究雌激素对心血管的保护作用。方法:将45只3月龄雌性Sprague-Dawley大鼠随机分为空白对照组(CON,n=10)、假手术组(SHAM,n=10)、去势对照组(OVX,n=12)和去势实验组(OVX+E_2,n=13)。去势实验组给予200μg/(kg·d)17β-雌二醇灌胃,余3组给予等量生理盐水灌胃。灌胃16周后酶联免疫吸附测定(ELISA)法测定血清雌二醇(E_2)、卵泡刺激素(FSH)、MCP-1及Rho A水平;ELISA法检测心脏组织沉默调节蛋白1(SIRT1)、腺苷酸活化蛋白激酶(AMPK)水平。免疫组织化学法检测心脏组织MCP-1及Rho A水平。结果:灌胃16周后去势实验组血清及心脏组织MCP-1、Rho A水平低于去势对照组(P0.05);去势实验组心脏组织匀浆SIRT1、AMPK水平高于去势对照组(P0.05)。结论:外源性17β-雌二醇可下调去势雌性大鼠血清及心脏组织MCP-1、Rho A表达水平,从而发挥其心血管保护作用。其可能的机制是17β-雌二醇通过上调AMPK/SIRT1调控MCP-1及Rho A的表达水平。  相似文献

目的:研究植物雌激素葛根素和鸡豆黄素A对17β-雌二醇诱发去卵巢大鼠子宫内膜渗出和血管通透性的影响.方法:将36只健康雌性SD大鼠随机分为6组(n=6):假手术组、卵巢切除组、17β-雌二醇组、17β-雌二醇+他莫昔芬组、17β-雌二醇+葛根素组、17β-雌二醇+鸡豆黄素A组,除假手术组外,其他各组大鼠摘除双侧卵巢后,分别皮下注射芝麻油、17β-雌二醇、17β-雌二醇+葛根素、17β-雌二醇+鸡豆黄素A、17β-雌二醇+他莫昔芬21天后取组织,比较不同处理组子宫内膜渗出变化和血清VEGF的浓度.结果:给予植物雌激素葛根素、鸡豆黄素A和他莫昔芬后,子宫内膜嗜酸性粒细胞的渗出减少,血清VEGF浓度降低.结论:植物雌激素葛根素和鸡豆黄素A可发挥雌激素拮抗作用,使血清VEGF浓度减小,可抑制外源性雌激素诱导的子宫内膜的渗出.  相似文献

目的:观察雌二醇与不同剂量的葛根素组合对去卵巢大鼠骨组织的影响,为中西医结合的雌激素与葛根素最佳组合剂量治疗Ⅰ型骨质疏松症提供实验依据。方法:5月龄健康雌性大白鼠70只,分成7个实验组(每组10只),除假手术组外均建立去卵巢动物模型。葛根素-50组(Pr-50),皮下注射葛根素50 mg/kg,1次/天;雌二醇-200组(E2-200),皮下注射雌二醇200μg/kg,2次/周;雌二醇+葛根素-10组(E2+Pr-10),皮下注射雌二醇100μg/kg,2次/周,葛根素10 mg/kg,1次/天;雌二醇+葛根素-25组(E2+Pr-25),皮下注射雌二醇100μg/kg,2次/周和葛根素25 mg/kg,1次/天;雌二醇+葛根素-40组(E2+Pr-40),皮下注射雌二醇100μg/kg,2次/周和葛根素40 mg/kg,1次/天。各实验组在第10周和第20周,随机取5只大鼠取左股骨切片观察骨组织,取右股骨测量骨密度、骨钙、骨磷含量。结果:OVX组在第10周和第20周的骨密度和骨钙、骨磷明显低于假手术组和5个用药组(P<0.05),骨组织呈骨质疏松的病理改变;E2+Pr-25组在第10周和第20周的骨密度和骨钙、骨磷明显高于E2+Pr-10组(P<0.05);而与假手术组、Pr-50、E2-200和E2+Pr-40相比无统计学差异(P>0.05)。结论:E2+Pr-25组葛根素与雌二醇的组合剂量治疗去卵巢大鼠骨质疏松效果明显优于E2+Pr-10组的小剂量,与假手术组、相对较大剂量的E2+Pr-40组和单一较大剂量Pr-50、E2-200组相比治疗效果无明显差异。  相似文献

目的观察雌激素对去卵巢大鼠肝细胞胰岛素受体底物-1(IRS-1)和胰岛素受体底物-2(IRS-2)表达的影响,以探讨其改善胰岛素抵抗的机制。方法取6周SD雌性大鼠随机分为3组:假手术组(Sham组)、手术组(OVX组)和戊酸雌二醇组(E_2组),E_2组每天灌胃戊酸雌二醇水溶液(1 mg/kg),而其他组给予相应体积的蒸馏水,连续给药12周。然后采用葡萄糖氧化酶法测定血糖,实时荧光定量PCR法检测肝细胞IRS-1和IRS-2 mRNA的表达,免疫组织化学法检测肝细胞IRS-1和IRS-2蛋白的表达。结果 OVX大鼠终末体重、肝重、内脏脂肪指数KITT显著高于Sham组,而葡萄糖利用常数KITT显著低于Sham组(P0.05);与OVX组大鼠相比,E_2组大鼠终末体重、肝重、内脏脂肪指数显著降低,而葡萄糖利用常数显著升高(P0.05);免疫组化结果显示,OVX大鼠肝组织IRS-1、IRS-2表达阳性细胞数目及染色程度较Sham组显著减少,而雌激素替代治疗后肝组织IRS-2阳性表达细胞数目及染色程度显著改善,但IRS-1改善不显著;OVX大鼠肝细胞IRS-1和IRS-2mRNA表达水平显著低于Sham组,而E_2组大鼠IRS-1和IRS-2 mRNA表达水平显著高于OVX组(P0.05)。结论雌激素可上调去卵巢大鼠肝细胞IRS-1和IRS-2的表达水平,进而改善胰岛素抵抗。  相似文献

目的:探讨雌、孕激素对STAT3在小鼠子宫内膜表达的影响。方法:将小鼠卵巢切除,随机分为对照组(腹腔注射油)、E2组(腹腔注射苯甲酸雌二醇)、P组(腹腔注射黄体酮)和E2+P组(腹腔注射苯甲酸雌二醇和黄体酮)。用RT-PCR方法检测STAT3 mRNA在各组小鼠子宫内膜的表达。同时用免疫组化方法检测STAT3和磷酸化STAT3蛋白在各组小鼠子宫内膜的表达。结果:STAT3 mRNA在E2组和E2+P组有明显的表达,而在P组和对照组无表达。STAT3蛋白在E2组和E2+P组有明显的表达,而在P组和对照组无表达。P-STAT3蛋白在E2组有强表达;在P组和E2+P组也有明显的阳性表达;而在对照组则无表达。结论:在小鼠子宫内膜,雌激素促进STAT3 mRNA和蛋白的表达;孕激素无促进STAT3 mRNA和蛋白表达的作用,但可以促进STAT3的磷酸化。在胚胎种植的过程中,STAT3的表达和活化受到雌、孕激素的调控。  相似文献

目的:观察复方丹参合剂对抗去卵巢大鼠骨元素代谢紊乱的作用。方法:将健康4月龄雌性SD大鼠36只,随机分成6组。A组(正常对照);B组(假去卵巢);C组(去卵巢);D组(去卵巢+合剂小剂量);E组(去卵巢+合剂中等剂量);F组(去卵巢+合剂大剂量)。去卵巢后第2 d开始,连续用药11周。用测定骨元素含量的方法,分析了复方丹参合剂(三种不同浓度)对切除双侧卵巢后的大鼠骨矿物元素代谢的影响。结果:去卵巢大鼠骨Ca、S、Mg、Mn等元素明显降低,骨P含量增加,复方丹参合剂可使去卵巢大鼠降低的骨Ca、S、Mg、Mn等矿物元素均显著回升,骨P回降。结论:复方丹参合剂具有良好的对抗由于去卵巢引起骨元素代谢紊乱作用。  相似文献

目的:观察利骨素对去卵巢大鼠骨矿盐丢失的预防作用。方法:健康4月龄SD雌性大鼠24只,随机分成4组,A组(正常对照);B组(假去卵巢);C组(去卵巢);D组(去卵巢+利骨素)。取左股骨测定骨矿盐含量。结果:去卵巢大鼠骨干重、骨干重/体重、骨灰重、骨灰重/体重、骨灰重占骨干重(%)等指标均明显降低,持续服用利骨素11周,可使实验组大鼠上述指标明显高于去卵巢组。提示:利骨素有良好的预防去卵巢引起的骨矿盐丢失作用。  相似文献

At the level of prevention of bone mineral loss produced by ovariectomy, the aim of the present study was to determine the effect produced by supplementation of Ca in the diet and a moderate exercise programme (treadmill), simultaneously or separately, in ovariectomized rats, an experimental model of postmenopausal bone loss. Female Wistar rats (n 110, 15 weeks old) were divided into five groups: (1) OVX, rats ovariectomized at 15 weeks of age, fed a standard diet; (2) SHAM, rats sham operated at 15 weeks of age, fed a standard diet; (3) OVX-EX, ovariectomized rats, fed a standard diet and performing the established exercise programme; (4) OVX-Ca, ovariectomized rats fed a diet supplemented with Ca; (5) OVX-EXCa, ovariectomized rats with the exercise programme and diet supplemented with Ca. The different treatments were initiated 1 week after ovariectomy and were continued for 13 weeks for subgroup 1 and 28 weeks for subgroup 2, to look at the interaction of age and time passed from ovariectomy on the treatments. Bone mineral density (BMD) was determined, at the end of the study, in the lumbar spine (L2, L3 and L4) and in the left femur using a densitometer. Bone turnover was also estimated at the end of the study, measuring the serum formation marker total alkaline phosphatase (AP) and the resorption marker serum tartrate-resistant acid phosphatase (TRAP). As expected, OVX rats showed a significant decrease (P<0.05) in BMD, more pronounced in subgroup 2, and a significant increase in AP and TRAP with regard to their respective SHAM group. The simultaneous treatment with Ca and exercise produced the best effects on lumbar and femoral BMD of ovariectomized rats, partially avoiding bone loss produced by ovariectomy, although it was not able to fully maintain BMD levels of intact animals. This combined treatment produced a significant increase in AP, both in subgroups 1 and 2, and a decrease in TRAP in subgroup 1, with regard to OVX group. The exercise treatment alone was able to produce an increase in BMD with regard to OVX group only in subgroup 1 of rats (younger animals and less time from ovariectomy), but not in subgroup 2. In agreement with this, there was an increase of AP in both subgroups, lower than that observed in animals submitted to exercise plus Ca supplement, and a decrease of TRAP in subgroup 1, without significant changes in this marker in the older rats. Ca treatment did not produce any significant effect on BMD in OVX rats in both subgroups of animals, showing a decrease of AP and TRAP levels in the younger animals with no significant variations in markers of bone remodelling in the older female rats compared with their respective OVX group.  相似文献

ObjectivesPostmenopausal women develop obesity, insulin resistance, and potentially renal dysfunction because of decreased serum estrogen levels. We investigated the effects of genistein, an estrogen-like compound thought to exert antioxidative effects, on insulin resistance, renal lipid accumulation, and oxidative stress in ovariectomized rats.MethodsThree weeks after an ovariectomy or a sham surgery, rats were put on a high-fat diet containing 0% or 0.1% genistein for 4 wk. We examined the following treatment groups: sham surgery + high-fat diet (sham), ovariectomy + high-fat diet (OVX), and ovariectomy + high-fat diet with 0.1% genistein (OVX + G).ResultsThe OVX + G group had increased serum estradiol levels and renal expression of estrogen receptors-α and -β compared with the OVX group. OVX + G rats showed decreases in serum insulin levels and the insulin resistance index. OVX + G rats also exhibited decreased renal triacylglycerol and cholesterol levels, which may have been the result of decreased sterol response element binding protein-1 and -2 expressions, and increased adenosine triphosphate-binding cassette transporter-1 and adiponectin receptor expression. The observed increases in renal lipid levels and serum and renal transforming growth factor-β in OVX rats may be associated with the increased expression of extracellular matrix proteins, such as fibronectin, and the decreased activity of metalloproteinase-2, an extracellular matrix–degrading enzyme. Ovariectomy also induced oxidative stress by the reduction of antioxidative enzymes, whereas genistein reversed these detrimental ovariectomy-induced effects.ConclusionGenistein may help to maintain normal kidney function through the alleviation of many ovariectomy-induced risk factors for renal damage, including an increased insulin resistance index, renal oxidative stress, lipid accumulation, and extracellular matrix protein expression.  相似文献

This study aimed to investigate the effects of low-exposure Cd on normal and osteoporotic bone. For this purpose, 12-week-old Sprague-Dawley female rats were assigned randomly to a control group, a Cd group, and an ovariectomy (OVX)+Cd group. OVX+Cd rats underwent bilateral ovariectomy via ventral incision. Twelve weeks after ovariectomy, cadmium chloride (CdCl(2)) was given to rats (Cd and OVX+Cd groups) as intraperitoneal (ip) injection of 0.5mg/kg three times a week for 18 weeks and distilled water was given to control group via ip route for 18 weeks. Bone mineral density (BMD) was measured at mid-diaphysis femoral region by dual-energy X-ray absorbsiometry. Cross-sectional area of the femoral shaft was evaluated by computerized tomography. Biomechanical measurements were performed at the mid-diaphysis of the left femur. Collagen fibers were evaluated at light microscopic level. BMD, cortical thickness, cortical area, and femur length were not changed in Cd and OVX+Cd groups in comparision to controls. In the OVX+Cd group, strength, displacement, energy, stress, strain, and toughness were significantly lower than those of the control group. The Cd concentration of bone was significantly increased in the OVX+Cd group compared to that in the control group. Collagen fiber intensity was decreased in all groups except control group. The results of the present study indicate that the administration of low-dose Cd does not affect normal bone biomechanical parameters, but it has a significant effect on osteoporotic bone.  相似文献

用测定骨元素含量的方法,观察运动和尼尔雌醇(CCE3)联合应用对去卵巢大鼠骨元素代谢的影响。将健康4月龄雌性SD大鼠40只,用抽签法随机分成5组:正常对照组;假手术组;去卵巢组;去卵巢+CCE3组;去卵巢+CCE3+运动组。去卵巢+CCE3组和去卵巢+CCE3+运动组大鼠于去卵巢手术后第2 d开始给予CCE3灌胃,0.5 mg/(kg.BW),1次/w,持续11周。去卵巢+CCE3+运动组于去卵巢术后第7 d开始运动训练,每周5 d,每天连续匀速跑45 min,16 m/min,跑道倾角0°,持续10周。结果表明,去卵巢组大鼠骨Ca、S、Mg、Zn、Co、Mn等元素含量均明显低于假去卵巢组,骨P含量明显高于假去卵巢组;去卵巢+CCE3组大鼠骨Ca、S、Mg、Zn、Co、Mn等元素含量虽然较去卵巢组明显增加,但仍显著低于假去卵巢组;运动与CCE3联合应用后,去卵巢大鼠骨Ca、S、Mg、Zn、Co、Mn等元素含量均显著回升,骨P含量显著回降,并且基本恢复到对照组水平。提示运动可加强CCE3对抗由于去卵巢引起的大鼠骨元素代谢紊乱。  相似文献

This study demonstrates that feeding difructose anhydride III (DFAIII) improves bone strength and femoral mineral concentrations in a rat model of oestrogen deficiency. We showed the relationship between Ca, Mg and P absorption and bone characteristics in rats. Two groups of female Sprague-Dawley rats (6 weeks old) underwent bilateral ovariectomy (ovariectomized rats, OVX rats) or bilateral laparotomy (sham rats). At 10 weeks old, OVX and sham rats were divided into three subgroups and fed a control, 1.5 % DFAIII or 3 % DFAIII diet for 8 weeks, respectively. Ca but not Mg absorption rates were lowered by ovariectomy; however, ingestion of the 1.5 % and 3 % DFAIII diets similarly restored the reduced Ca absorption in OVX rats at 4 and 8 weeks after feeding of the test diets. DFAIII increased Mg absorption dose-dependently in sham and OVX rats. The bone strength, femoral Ca and Mg concentrations, and distal bone mineral density in the 3 % DFAIII group were higher than those in the control group in OVX rats. The absorption rates of Ca and Mg were significantly correlated with femoral Ca and Mg concentrations and strength, which suggests that increasing both Ca and Mg absorption improves bone characteristics in OVX rats. There were no differences in any of the variables in the femur between the 1.5 % and 3 % DFAIII groups in OVX rats. In conclusion, feeding of a low dose of DFAIII increased intestinal Ca and Mg absorption, and the promotive effect of DFAIII persisted for over 8 weeks. This effect was associated with prevention of ovariectomy-induced osteopenia.  相似文献
