河南汉族人群PentaD和PentaE基因座的遗传多态性

摘    要:目的调查分析PentaD和PentaE 2个5核苷酸短串联重复序列遗传标记在河南汉族人群中的遗传多态性分布。方法酚氯仿法提取河南汉族群体155名无关个体血样本基因组DNA,PentaD和PentaE用PowerPlexTM16试剂盒进行扩增,AB13130xl全自动遗传分析仪检测聚合酶链反应产物。结果 PentaD和PentaE 2个基因座的等位基因及基因型频率在河南汉族群体中分布均符合Hardy-Weinberg平衡定律(P>0.05)。PentaD基因座观察到9个等位基因和29个基因型;PentaE基因座观察到20个等位基因和82个基因型。PentaD、PentaE 2个基因座的杂合度、个体识别率、非父排除概率分别是0.845 2和0.916 1,0.943 0和0.984 0,0.685 0和0.828 0;累积非父排除概率和累积个体识别率分别是0.945 8和0.999 1。结论 PentaD和PentaE在河南汉族群体中具有较高的遗传多态性,在法医学个体识别中具有应用价值。

Abstract:Objective To investigate genetic polymorphisms distribution of two pentanucleotide motif of short tandem repeats including PentaD and PentaE loci in Chinese Han population of Henan province.Methods Genomic DNA of blood samples of 155 unrelated individuals were extracted by using phenol-choroform method.PentaD and PentaE were amplified with PowerPlexTM16 kit,and the polymerase chain reaction products were detected by AB13130xl genetic analyzer.Results The distribution of alleles and genotypes of PentaD and PentaE loci in Han population of Henan province was in Hardy-Weinberg equilibrium(P>0.05).Nine alleles and twenty-nine genotypes of locus PentaD were observed,and twenty alleles and eighty-two genotypes of locus PentaE were found.Heterozygosity,power of discrimination,power of exclusion of locus PentaD and PentaE were 0.845 2 and 0.916 1,0.943 0 and 0.984 0,0.685 0 and 0.828 0,respectively.Cumulative power of exclusion and cumulative power of discrimination were 0.945 8,0.999 1,respectively.Conclusion PentaD and PentaE loci in Han population of Henan province have high polymorphism,which has application value in forensic individual indentification.
