摘    要:目的探讨胰岛素样生长因子-1(IGF-1)对人晶状体上皮细胞增殖的影响。方法对永生化人晶状体上皮细胞系B-3(HLEB-3)进行培养及传代,应用不同浓度的IGF-1(5、10、20、30、40、50、60μg.L-1)作用于HLEB-3细胞,在不同时间点采用四甲基偶氮唑盐比色(MTT)法检测IGF-1对HLEB-3细胞增殖的影响。采用蛋白质印迹(West-ern blotting)方法检测晶状体上皮α-肌动蛋白(α-actin)的表达。结果 IGF-1在5μg.L-1时就对HLEB-3细胞有明显的促增殖作用(P<0.05),且随着IGF-1浓度的升高,促增殖作用增加,即促增殖效应具有浓度依赖性;同时也可上调α-actin蛋白的表达,与对照组比较,各浓度组IGF-1均可显著促进α-actin蛋白表达上调(P<0.01),且随着IGF-1浓度的增加,α-actin蛋白含量逐渐升高。结论 IGF-1具有促进人晶状体上皮细胞增殖的作用,其作用强度呈浓度依赖性。

Abstract:Objective To investigate the effect of insulin-like growth factor-1(IGF-1) on proliferation of human lens epithelial cells(LECs) in vitro.Methods Immortalized human lens epithelial B-3(HLEB-3)were cultured and passaged in this experiment.IGF-1 was added into the medium to obtain the terminal concentration at 5,10,20,30,40,50,60 μg·L-1 respectively.And HLEB-3 was exposed to different concentrations for different time points.Proliferation of HLEB-3 was detected with methyl thiazolyl tetrazolium(MTT) assay.The expression of α-actin in LECs was examined with Western blotting mothods.Results IGF-1 induced proliferation of HLEB-3 in 5 μg·L-1(P<0.05),and proliferation of HLEB-3 was concentration-dependent.HLEB-3 treated by IGF-1 showed an increase in α-actin expression at protein levels.Proliferation of HLEB-3 line and the expression of α-actin protein in HLEB-3 was considerably increased with IGF-1 in experimental group compared with control group(P<0.01),and the expression of α-actin protein increased with the IGF-1 concentration increasing.Conclusion IGF-1 can enhance proliferation of human LECs in vitro and play its role at concentration-dependent manner.
