摘    要:目的 探讨微小RNA-15b (microRNA-15b,miR-15b)对人肺癌耐药细胞A549/DDP凋亡的影响.方法 采用体外转染法将MicroRNA-15b瞬时转染到A549/DDP细胞后,应用Real-time PCR检测A549/DDP细胞中MicroRNA-15b的表达情况;流式细胞仪(FCM)检测细胞凋亡变化;并用Western印迹法(Western blot)检测细胞中Bcl-2表达.结果 转染后miR-15b组的miR-15b表达水平显著增加(P〈0.05);miR-15b组细胞凋亡率为:32.4%±5.1%,与Mock组(5.73%±1.2%)和miR-15b-Cont(6.24%±2.4%)比较,差异具有统计学意义(P〈0.05);miR-15b组的Bcl-2表达量较对照组明显增加.结论 miR-15b可能通过下调Bcl-2的表达从而诱导A549/DDP细胞的凋亡,这可能为肺癌耐药的治疗提供新靶点.

Abstract:Objective To investigate the effect of miRNA-15b on the apoptosis of drug-resistant cells of lung cancer A549/DDP. Methods MiR-15b in vitro was transiently transferred into A549/DDP by FuGENE-HD. The expression of MiR-15b in A549/DDP cells was detected by real-time fluorescence quantitative PCR, and the apoptosis of A549/DDP cells was analyzed by flow eytometry. The ex- pression of Bcl-2 protein was detected by Western blot. Results The expression of miR-15b in miR-15b-transfected A549/DDP cells in- creased significantly (P 〈0.05). The apoptosis of transfected cells was 32. 4% ~5. 1% , 5.73% ~ 1.2% and 6. 24% ~2.4% respec- tively in the miR-15b group, the Mock group and the miR-15b-Cont group, which showed significant differences (P 〈0. 05). The expres- sion of Bcl-2 increased more obviously in the miR-15b group than in the control group (P 〈 0. 05 ). Conclusion The miR-15b may in- duce the apoptosis of A549/DDP cells by down-regnlating the expression of Bcl-2, which provides a new target for the treatment of lung cancer.
