碱性成纤维细胞生长因子对脑缺血再灌注后功能恢复的影响

摘    要:目的:研究碱性成纤维细胞生长因子(bFGF) 对大鼠局部脑缺血再灌注后神经功能恢复的影响和机理。方法:应用线栓法制作Wistar大鼠大脑中动脉闭塞再灌注模型(MCAO),采用行为测试评分离缺血MCAO后的功能恢复情况。结果:脑缺血MCAO后1d大鼠患肢感觉运动功能降至最低点,随时间延长功能逐渐恢复,功能恢复曲线在术后约5d陡直上升,以后缓慢上升,至第27d仍未达到正常水平。脑室注射bFGF各时间点行为功能恢复水平高于自然恢复组。前肢试验的恢复高峰在术后第9d,前,后肢放置试验到27d与对照组比较差异无统计学意义。后肢试验与平衡木试验在4周末显示了第2个恢复高峰。结论:脑缺血MCAO后肢体功能有自然恢复的趋势,外源性bFGF能促使脑缺血MCAO后的肢体功能恢复。

Abstract:Objective: To observe the effect of intracisternally basic fibroblast growth factor(bFGF) on the funtional recovery after focal cerebral ischemia and reperfusion in rats and the mechanism.Methods: Wistar rafs were subjected to 120 min of middle cerebral artery occlusion(MCAO) to establish MCAO model. By using behavioral test, the functional recovery after cerebral ischemia and reperfusion was assessed.Results: One h after cerebral ischemia and reperfusion, the function of the sensorimotor in the affected extremity was increased to the lowest, then gradually recovered with the prolongation. The functional recovery curve was elevated right at 5 th day after operation, then slowly elevated, and didn't reach the normal levels at 27 th day. At various time points of BFGF intraventricular injection, the behavioral functional recovery was better than in the spontaneous recovery group. Conclusion: After cerebral ischemia/reperfusion, the posterior limb had a tendency of spontaneous recovery.Exogenous bFGF could promote the functional recovery of the extremity after cerebral ischemia and reperfusion.
