人骨肉瘤组织PTEN蛋白的表达及临床意义

摘    要:目的:探讨抑癌基因PTEN在人骨肉瘤及骨软骨瘤组织中的表达。方法:应用免疫组化(SP)法,检测36例骨肉瘤组织标本中PTEN蛋白的表达,并与15例骨软骨瘤进行对照研究。结果:36例骨肉瘤组织中,PTEN蛋白的阳性表达率为75%,骨软骨瘤组织中阳性率为93.33%,二组间差异无统计学意义。PTEN蛋白表达与骨肉瘤Enneking分期及组织学分型也无相关性。结论:PTEN蛋白通过PTEN/Akt信号传导途径抑制肿瘤细胞凋亡的作用未在骨肉瘤中表达出来,表明PTEN/Akt信号传导途径不能控制骨肉瘤细胞的增殖与凋亡。

Abstract:Objective To study the expression of human tumor supressor gene phosphatase and tensin homology deleted on chromosome Ten in osteosarcoma and osteochondroma group. Methods Expression of phosphatase and tensin homology deleted on chromosome Ten protein was detected by SP immunostaining technique in 36 cases of osteosarcoma, and compared with that in 15 cases of osteochondroma. Results In 36 cases of osteosarcoma, the positive rate of phosphatase and tensin homology deleted on chromosome Ten protein was 75%, and was 93. 33% in 15 cases of osteochondroma. Expression of phosphatase and tensin homology deleted on chromosome Ten protein was detected in no significantly greater proportion of osteosarcoma than osteochondroma. There was no significant difference between phosphatase and tensin homology deleted on chromosome Ten protein expression and pathological type, and between phosphatase and tensin homology deleted on chromosome Ten protein expression and Ennecking stage. Conclusion Phosphatase and tensin homology deleted on chromosome Ten protein doesn't inhibit tumor cell apoptosis and proliferation through phosphatase and tensin homology deleted on chromosome Ten /Akt signal transmissional way in osteosarcoma.
