SD大鼠皮肤组织教学石蜡切片的改进

目的:探讨一种改善皮肤组织教学切片质量的方法。方法:取大鼠的皮肤,用Bouin液固定、脱水﹑氯仿透明﹑浸蜡﹑包埋﹑切片﹑HE染色和封片,最后在光镜下观察。结果:新方法制作的皮肤组织切片结构完整,厚薄均匀,染色恰当,形态清晰无断裂破碎现象。结论:用Bouin固定液软化,降低组织硬度,氯仿透明不引起组织收缩变形及过度硬脆。这种改进的实验方法可切实提高皮肤组织石蜡切片效果,是可行的。

Improved method for making paraffin-embedded rat tissue sections

Objective: To discuss an improved method for making paraffin-embedded rat tissue sections.Methods:Rat skin sections were fixed in Bouin’s fluid,dehydrated,chloroform transparentized and then immersed in paraffin for HE staining and sealing.Results:Under electron microscopy,the paraffin-embedded rat tissue sections was even in sickness and color showing clear shapes and complete tissues.Conclusions:Bouin’s fluid can make the skin tissues soft,and transparent chloroform can prevent the tissues becoming deformed or brittle,thus this method can help to make good paraffin-embedded rat tissue sections.
