活性维生素D3对大鼠早期糖尿病肾病相关炎性因子的影响

摘    要:目的探讨活性维生素D3对2型糖尿病大鼠肾脏损伤的预防保护作用。方法雄性SD大鼠30只随机分为对照组、2型糖尿病组(T2DM组)、2型糖尿病活性维生素D3干预组(干预组)各10只。对照组饲以普通饲料,T2DM组及干预组饲以高脂高糖饲料喂养6周后,干预组大鼠给予活性维生素D3灌胃,T2DM组给予等量花生油灌胃,7周后检测3组24h尿蛋白水平,观察大鼠肾脏组织形态学和肾功能、血生化指标,采用免疫组织化学法检测3组肾脏转化生长因子-β1,CD68和单核细胞趋化蛋白-1表达情况。结果 T2DM组大鼠三酰甘油、血糖、24h尿蛋白、胆固醇高于对照组,尿素氮、体质量低于对照组(P<0.05);干预组24h尿蛋白、血糖、肾质量高于对照组,尿素氮低于对照组(P<0.05);干预组胆固醇、24h尿蛋白低于T2DM组,肾质量和体质量高于T2DM组(P<0.01);T2DM组单核细胞趋化蛋白-1、CD68和转化生长因子-β1阳性细胞数明显高于对照组及干预组(P<0.01)。结论活性维生素D3可通过抑制糖尿病大鼠肾脏转化生长因子-β1、CD68及单核细胞趋化蛋白-1等细胞因子表达,抑制巨噬细胞浸润,对糖尿病大鼠肾脏损伤有预防保护作用。

Influence of 1,25-dihydroxyvitamin D3 on the expression renal cytokines in rats with diabetic nephropathy

Abstract:Objective To explore the preventive protection effect of 1,25-dihydroxyvitamin D3 on the kidney function in rats with type 2 diabetes mellitus.Methods Thirty male SD rats were randomly divided into control group,type 2 diabetes mellitus(T2DM) group and intervention group(type 2 diabetes mellitus rats intervened with 1,25-dihydroxyvitamin D3) with 10 rats in each group.After high-glucose and high-lipid diet for 6 weeks,the intervention group was given 1,25-dihydroxyvitamin D3 lavage,and T2DM group was given peanut oil in the same dose as 1,25-dihydroxyvitamin D3 used in the intervention group.Control group was fed with normal diet.The morphology of kidney tissues was observed,and kidney function and 24-hour urinary protein quantitative index were detected after 7 weeks of treatment,while the expressions of transforming growth factor-beta 1,CD68 and monocyte chemoattractant protein-1 in kidney cortex were observed with immunohistochemistry in these three groups.Results The levels of triacylglycerol,plasma glucose,24-hour urinary protein and cholesterol were higher,and the body mass and urea nitrogen were lower in T2DM group than those in control group(P<0.05).The levels of 24-hour urinary protein,plasma glucose and kidney mass were higher in the intervention group than those in control group(P<0.05).The urea nitrogen was lower in the intervention group than that in the control group(P<0.05).The levels of cholesterol and 24-hour urinary protein were lower,and kidney mass and body mass were higher in the intervention group than those in T2DM group(P<0.01).The expression of transforming growth factor-beta 1,monocyte chemoattractant protein-1 and CD68 were higher in T2DM group than those in the other two groups(P<0.01).Conclusion 1,25-dihydroxyvitamin D3 can protect kidney of diabetic rats by inhibiting the expression levels of transforming growth factor-beta 1,monocyte chemoattractant protein-1 and CD68,and restraining macrophages infiltration.
