摘    要:目的本研究利用蛋白质组学的方法分析急性非淋巴细胞白血病(ANLL)与急性淋巴细胞白血病(ALL)细胞的蛋白质表达谱的差异,为了解两系别的生物学和临床特征的差别提供分子基础。方法利用二维凝胶电泳方法分别分离来源于25例ANLL和20例ALL患者细胞的蛋白质,用MALDI-TOF-MS生物质谱和电喷雾质谱(ESI-MS/MS)对ANLL和ALL的差异蛋白质进行鉴定分析。结果本研究获得ANLL和ALL的差异表达蛋白质谱,其中首次报道了髓系相关蛋白8和14不仅可以作为区别ANLL和ALL的分子标志,还可以区分ANLL细胞分化阶段,Op18、热休克蛋白27等在ANLL和ALL中差异表达的蛋白将为区分ANLL和ALL提供新的分子标志。结论这些差异蛋白将有助于了解ANLL和ALL间临床和生物学特征差异的分子机制。

Abstract:ObjectiveThe distinct proteins that contribute to acute non-lymphoid leukemia(ANLL) and acute lymphoid leukemia(ALL) were analyzed by proteomic technology in our study to lay the molecular basis for the different biological and clinical characteristics of each group. MethodsThe proteins of leukemic cells from 24 cases of ANLL and 20 cases of ALL characterized by FAB classification were separated by two-dimensional electrophoresis(2-DE) individually, and the differentially-expressed protein spots between the two groups were identified by both matrix assisted laser desorption/ionization-time of flight-mass spectrometry(MALDI-TOF-MS) and electronspray ionization(ESI)-MS/MS. ResultsThe distinct proteins of ANLL and ALL were successfully explored. Among them, myeloid related protein 8 and 14 were first reported to not only differentiate ANLL from ALL, but also mark ANLL differentiation. Other proteins, such as Op18, Heat shock 27 kDa protein 1 and other proteins differentially-expressed between ANLL and ALL, may play important roles in clinically distinguishing ANLL from ALL. ConclusionThese data have implications in the explanation of the different biological and clinical features between the ANLL and ALL.
