摘    要:为研究高于生理浓度的细胞外Ca2+对腮腺腺细胞分化和分泌功能的影响,以10倍和20倍生理浓度的Ca2+作用于培养中的腮腺腺细胞,应用免疫组织化学技术,观察细胞外Ca2+对细胞连接蛋白及与分化有关的蛋白表达的影响。结果显示,细胞粘接分子(CAM)、缝隙连接蛋白(Cx32)随细胞外Ca2+升高而表达升高;cjun蛋白随Ca2+浓度增高表达更广泛;而表皮生长因子受体(EGFγ),cfos及腮腺特异性分泌蛋白α淀粉酶、富含脯氨酸蛋白(PRP)表达无明显变化。研究结果提示,细胞外Ca2+在一定程度上可促进细胞分化,但对细胞分泌功能无明显影响。

Abstract:To study the role of calcium on parotid cell differentiation and secretion, high extracellular calcium was used to treat the parotid gland cells in culture. Immunohistochemical staining was applied to examine the expression of proteins involved in cell-cell interaction, cell differentiation and parotid gland secretary proteins. The results demonstrated that the expression of CAM and Cx32 increased and more cells had positive reaction to c-jun oncoprotein. No effect of calcium on the expression of EGF-gamma, c-fos, alpha-amylase and PRP was observed. The data indicate that extracellular calcium play an important role on cells differentiation. However, it does not affect the cell secretion.
