胃癌根治手术与血清、组织E-钙粘附素变化的关系

目的 通过检测胃癌患者血清E-钙粘附素(E-Cd)浓度和组织E-Cd的mRNA水平,探讨E-Cd与胃癌浸润和转移的关联性。方法 应用酶联免疫吸附法(ELISA)测定。39例胃癌患者血清可溶性E-Cd浓度;应用点杂交法和计算机灰度扫描分析组织的mRNA灰度均值。结果血清E-Cd浓度胃癌组明显高于对照组(P<0.01),浸润组或有淋巴结转移组明显高于膨胀组或无淋巴结转移组,切除瘤体后上述各组的血清 E-Cd浓度均明显下降。各组的组织 E-Cd mRNA均值变化与血清E-Cd浓度的一致,且两指标间具有明显的相关性。结论 血清、组织E-Cd可能与人胃癌的生长方式和淋巴结转移密切相关。

Relationship of serum and tissue E-Cadherin levels and radical operation of stomach carcinomas

Objective By detecting both serum E--Cadhein (E-Cd) concentration and tissue E--Cd mRNA level in 29 cases of gastric carcinoma to investigate the relationship between the activity of E--Cd and the aggessiveness and metastasis of human gastric cancer. Methods Fasting concentration of serum souble E--Cd was determined by ELISA, and gray values of E-Cd in the tissue was analyzed by dot blot and analyzing of computer scanning. Results The sem soluble E-Cd concentrations were significantly higher in patients with gastric cancers than in healthy subjects (P < 0. 01), and those in cancers with in- vasiveness or with metastatic lymph nodes than in ones with expansion or without melastatic lymph nodes (P < 0. 01--0. 05), respectively. After the removal of the tumor, the E-Cd level was dramatically decreased (P < 0. 01-0. 05) in all cancers. Changes of tissue E--Cd mRNA level in all groups were similar to those of the serum E-Cd, and a positive correlation was found (P < 0. 05) between the two parameters. Conclu- sion Serum and tissue E--Cd levels may be a key factor associated with the invasive and metastatic poten- tial of gastri carcinomas.
