六味地黄丸血中移行成分的分离及结构鉴定

摘    要:目的:在前期六味地黄丸血清药物化学初步研究的基础上分离并鉴定六味地黄丸血中移行成分。方法:以血清色谱指纹图为指导,采用大孔吸附树脂柱色谱、硅胶柱色谱及RP-C18柱制备色谱进行分离纯化,根据波谱数据进行结构鉴定。结果:对11种六味地黄丸血中移行成分进行分离和结构鉴定,共分离得到了8个化合物,分别鉴定为5-羟甲基-2-糠酸(A)、莫诺苷(B)、獐牙菜苷(C)、马钱子苷(D)、2-羟基-苯乙酮-4-葡萄糖醛酸苷(E)、4-甲氧基-5-磺酸基苯乙酮-2-葡萄糖醛酸苷(F)、2,4-二羟基-苯乙酮(G)、(H)2,5-二羟基-4-甲氧基苯乙酮。其中5-羟甲基-2-糠酸(A)为熟地、山茱萸和泽泻中的成分代谢形成;莫诺苷(B)、獐牙菜苷(C)、马钱子苷(D)为山茱萸所含成分直接入血而形成;2-羟基-苯乙酮-4-葡萄糖醛酸苷(E)、4-甲氧基-5-磺酸基苯乙酮-2-葡萄糖醛酸苷(F)、2,4-二羟基-苯乙酮(G)、(H)2,5-二羟基-4-甲氧基苯乙酮为丹皮酚的代谢产物。结论:将血清药物化学方法引入到化合物的分离制备领域中,成功地分离鉴定了六味地黄丸的体内直接作用物质。

Abstract:AIM: To isolate and identify constituents absorbed into blood after oral administration of Liuwei Dihuang Pill. METHODS: The isolation and purification of constituents were performed on macropole resin column, silica gel column and RP-C18 preparation column based on serum fingerprint profiles, and the structures of constituents were identified according to relative spectral data. RESULTS: Eight compounds were identified as 5-hydroxymethyl-2-furoic acid, morroniside, sweroside, loganin, 2-hydroxyacetophenone-4-O-glucopyranuronoside, 4-methoxyl-5-sulphoacetophenone-2-O-glucopyranuronoside, 2,4-dihydroxyacetophenone and 2,5-dihydroxy-4-methoxylacetophenone. CONCLUSION: Serum pharmacochemistry was introduced to determine the bioactive constituens of Chinese medicine, and the constituents absorbed into blood after oral administration of Liuwei Dihuang Pill were systematically isolated and identified.
