小鼠系统性白念珠菌感染脾γ-干扰素水平测定

摘    要:目的通过测定脾脏γ-干扰素含量在小鼠系统性白念珠菌感染不同时期水平,推测其在系统性白念珠菌感染过程中所起的作用。方法建立健康小鼠及环磷酰胺诱导的免疫抑制鼠系统性感染模型,用酶联免疫吸附试验(ELISA)测出脾脏γ-干扰素含量,用平皿系列稀释法检测肾内菌落形成单位(CFU),并记录平均生存时间。结果白念珠菌感染组小鼠脾脏γ-干扰素均在感染后7天达最高峰;其中亚致死量感染组(B组)γ-干扰素含量高于亚致死量白念珠菌感染及免疫抑制剂组(A组)(P<0.05),A组高于免疫抑制组(C组)(P<0.05);B组肾脏菌落计数明显低于A组(P<0.01),肾脏菌落计数与γ-干扰素水平呈明显负相关(P<0.01)。结论小鼠白念珠菌感染早期γ-干扰素是升高的,推测其有限制真菌生长、繁殖、增强机体抗白念珠菌作用。

Abstract:Objective To discuss the role of the Interferon-gamma(IFN-γ) in the spleen of experimental murine systemic candidiasis.Methods In the Cyclophosphamide-induced immuno-suppressed murine systemic candidiasis models and control healthy mice,two-site ELISA was applied to determine the levels of IFN-γ,coloney forming units(CFUs) of infected kindneys were determined with the plating dilution method and mean survival time(MST)of mice was also recorded.Results Comparing sublethal infection group( B group),MST of sublethal infection of immunosuppressed group (A group) was obviously reduced,in which the level of IFN-γ was lower than that in group B (P<0.05) and higher than that in immunosuppressed group( C group) (P<0.05),Meanwhile,CFUs of group B in infected kindneys were lower than that in group A (P<0.05) and the level of IFN-γ was related negative to CPUs.Conclusion The elevation of the level of IFN-γ in experimental murine systemic candidiasis suggests IFN-γ is associated with restraining fungus from growing and reproduction and enhances the ability against the candida albicans.
