黑木耳多糖对ICR小鼠皮肤光老化的保护作用

摘    要:目的:探讨黑木耳多糖(Auricularia auricular polysaccharide,AAP)对ICR小鼠皮肤光老化的保护作用。方法:选取健康雌性ICR小鼠共50只,随机分成5组各10只:正常组(A)、模型对照组(B)、模型+AAP低剂量(50mg/kg.d)灌胃组(C)、模型+AAP中剂量(100mg/kg.d)灌胃组(D)、模型+AAP高剂量(200mg/kg.d)灌胃组(E)。长波紫外线(ultraviolet A,UVA)联合中波紫外线(ultravioletB,UVB)照射小鼠制备皮肤光老化模型。每周一、三、五照射,第1周每次照射时间1h,以后每周依次增加1h,自第5周起为5h,直至第14周照光结束。每次照光前各治疗组小鼠给予不同浓度的AAP灌胃。实验结束后取小鼠背部皮肤用测试盒测定超氧化物歧化酶(superoxide dismutase,SOD)及谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-PX)活力、丙二醛(malondial dehyde,MDA)含量;组织切片HE染色观察皮肤结构改变。结果:与正常组比较,模型对照组皮肤组织中SOD、GSH-PX活力显著降低(P<0.05),MDA含量显著增高(P<0.05);与模型组对照组比较,AAP各灌胃组SOD、GSH-PX活力显著升高(P<0.05),MDA含量显著降低(P<0.05)。模型对照组皮肤出现皮屑、褐黄、粗糙肥厚、深皱纹、缺乏弹性等光老化特征,组织切片呈现光老化改变。AAP各治疗组皮肤外观及组织学特点均较模型对照组明显改善。结论:AAP能有效提高小鼠抗氧化能力,对小鼠皮肤光老化有保护作用。

Abstract:Objective To study the protective effects of Auricularia auricular polysaccharide(AAP)on photoaged ICR mice skin.Methods Fifty healthy female ICR mice were divided into five groups(n = 10) randomly: normal group(A),model control group(B),at dose of 50 mg/kg.d group(C),at dose of 100 mg/kg.d group(D),at dose of 200 mg/kg.d group(E).To establish skin photoaging mice models by irradiated UVA and UVB three times weekly(Monday,Wednesday and Friday).In the first week the time of UV exposure was 1 hour each time,then increased by 1 hour per week until week 5,and maintained the time until the 14th week.Before every UV irradiation,the AAP were orally administered with different dose to treated mice groups respectively.At the end of the experiment,the superoxide dismutase(SOD) and glutathione peroxidase(GSH-PX) activities,malondialdehyde(MDA) content in the skin were detected by assay kit.The skin tissue was observed by HE staining histological sections.Results Compared with the normal group,the activities of SOD and GSH-PX were decreased significantly(P<0.05),and the content of the MDA was increased significantly(P<0.05) in the model control group.Compared with the model control group,the activities of SOD and GSH-PX in group C,D and E were increased significantly(P<0.05),and the content of MDA was reduced significantly(P<0.05) in every AAP treatment group.The model control group appeared photoaging features,such as scale,brown-yellow color,coarse,thickened,deep wrinkles and poor flexibility.While compared with the model control group,both the skin appearance and histological change of AAP treatment groups were improved significantly.Conclusion AAP has protective effect on the skin photoaging in ICR mice via increasing the capacity of anti-oxidation.
