摘    要:目的:确证在DNA数据库构建过程中一样本D13S317基因座出现的稀有分型标准物外off-ladder(OL)等位基因。方法:AGCU17+1STR荧光检测试剂盒进行STR分型时,D13S317基因座检出OL等位基因;利用Chelex-100法提取血样DNA进行荧光PCR,应用聚丙烯酰胺凝胶电泳和ABI3130电泳检测并由上海生工进行测序。结果:OL等位基因只含有6个TATC]重复,不在D13S317基因座AllelicLadder范围之内,是一个新的等位基因。结论:新的等位基因的出现,对扩大DNA数据库的覆盖范围及全面性和提高基因座个体识别能力等具有重要的借鉴意义。

Abstract:Objective: To certify the rare off-ladder (OL) alleles found on the D13S317 locus during the DNA database construction. Methods: OL allele was screened by using AGCU 17 + 1 system kit, Chelex-100 was used to extract DNA from the blood sample for fluorescence polymerase chain reaction. The PCR products were detected by PAG vertical electrophores and ABI 3130 ge- netic analyzer, sequencing by Shanghai Sangon. Results: OL allele only contained six repeats of TATC ] without the range of D13S317 allelic ladder which meant it was a new allele. Conclusion: The discovery and typing of OL allele is great significant to ex- pand coverage and completeness of DNA database and improve the loci individual recognition ability.
