沙棘原花青素对大鼠应激性胃溃疡的预防作用

目的:探讨沙棘原花青素(SBPC)对因束缚-浸水应激引起的大鼠应激性胃溃疡的影响。方法:60只雄性Wistar大鼠随机分为6组:正常组,对照组,低、中、高SBPC剂量组(50,100,150 mg/kg diet)及雷尼替丁阳性对照组。采用束缚-浸水应激的方式造应激性胃溃疡模型,造模前灌胃不同剂量的SBPC或阳性药物。测定溃疡指数、大鼠胃液中胃酸含量和胃蛋白酶活性;大鼠的血浆中皮质醇(COR)、促肾上腺皮质激素(ACTH)及NO;免疫组化的方法测定胃粘膜组织中iNOS的表达;透射电镜观察大鼠胃粘膜的超微细胞结构。结果:高剂量组SBPC(150 mg/kg diet)的溃疡指数明显降低(P<0.01)。SBPC可以抑制胃酸的分泌(P<0.01),降低胃蛋白酶的活性(P<0.01),降低血浆中COR、ACTH、NO的水平(P<0.01),抑制粘膜中的iNOS的表达。结论:预服一定剂量的SBPC对应激性胃溃疡的发生有一定的预防作用。

EFFECTS OF SEA BUCKTHORN PROCYANIDINS ON GASTRIC ULCER IN STRESSED RATS

Objective: To study the effects of different sea buckthorn procyanidins(SBPC) on gastric ulcer in stressed rats.Method: Sixty Wistar male rats were assigned into 6 groups at random: normal(unstressed),control,low,medium,high-dose(50,100,150mg/kg diet) SBPC supplement and ranitidine hydrochloride 30mg/kg diet supplement.The rat model of acute gastric ulcer was induced by water immersion and restraint stress(WRS).Rats were orally administered with SBPC/ranitidine/water before stressed.Gastric lesion index,acidity and pepsin activity of gastric juice,cortisol,ACTH,NO in plasma and nitric oxide synthase(iNOS) expression in gastric mucosa were determined.The ultrastructural change of parietal cells was observed by transmission electron microscope.Results: Compared with the control group,the ulcer index,the level of [H ] and pepsin activity of gastric juice,the level of cortisol,ACTH,NO in plasma,and iNOS expression in gastric mucosa in high-dose SBPC group were significantly decreased.Conclusion:Sea buckthorn procyanidin has remarkable preventive effects on gastric ulcer induced by water immersion and restraint stress in rats.
