罗丹明6GDN对小鼠肝癌的细胞毒与治疗效应

摘    要:Hep A腹水肝癌小鼠腹腔注射亲癌荧光染料罗丹明6GDN(Rh 6GDN)2.0mg/kg后,肝癌细胞抑制现象出现缓慢而作用持久。癌细胞呈变性形态学改变。于接种肿瘤后第1、3、5d分别注射生理盐水0.4ml、Rh 6GDH 0.5mg/kg、2-脱氧葡萄糖(2-DG)1.0g/kg、Rh 6GDN 0.5mg/kg加2-DG 1.0g/kg和5-FU10.0mg/kg后,各组肝癌小鼠的平均生存天数分别为14.3、17.7、16.6、20.7(p<0.01)和21.6d(p<0.01)。上述资料表明Rh 6GDN有直接肝癌细胞毒效应,与2-DG合用于Hep A肝癌小鼠有明显疗效。

CYTOTOXIC AND THERAPEUTIC EFFECTS OF RHODAMINE 6GDN ON MOUSE HEPATOMA

Abstract:The cytotoxic and therapeutic effects of rhodamine 6GDN (Rh 6GDN), a fluorescent dye, on mouse Hep A hepatoma were studied 6, 18, 80, 42 and 90 hours after an i. p. injection at a dose of 2.0mg/kg body weight. The inhibitory rate of the Hep A hepatoma cells by Rh 6GDN was 9.1, 42.4, 47.8, 57.9 and 88.6% at different times respectively. When 3 repeated i. p. doses of normal saline 0.4ml, Rh 6GDN 0.5mg/kg, 2-deoxy-D-glucose (2-DG) 1.0g/kg, Rh 6GDN 0.5mg/kg plus 2-DG1.0g/kg and 5-FU 10 0mg/kg were given to different groups of mice 1, 3, end 5 days after ascites transplantation, their respective mean survival time was 14.3, 17.7, 16.6, 20.7 (p<0.01) and 21.6 days (p<0.01). The longest survival time was 16, 30, 26, 32 and 28 days. The results suggest that Rh 6DGN possesses a protracted direct cytotoxic effect against the hepatoma cells. When combined with 2-DG, Rh 6DGN is efficacious for mouse Hep A hepatoma.
