Abstract:Objective To establish a high-performance liquid chromatography(HPLC) method for the content determination of dehydroiisoeugenol in Myristicae Semen roasted with flour.Methods The chromatographic column was Waters C18column(250 mm×4.6 mm,5 μm),the mobile phase was methanol-water(72-28,V/V),the flow rate was 1.0 mL/min,the detection wavelength was 274 nm,the column temperature was 30℃,and the injection volume was 10 μL.Results The injection volume of dehydroiisoeugenol showed a good linear relationship with the peak area in the range of 0.132 0-0.792 0 μg(r=1.000 0,n=6),and the average recovery of dehydroiisoeugenol was 98.34% with an RSD of 1.05%(n=12).The RSDs of precision,stability and repeatability tests were less than 2%.Conclusion The method is simple,accurate,reproducible and stable,which can be used for the content determination of dehydroiisoeugenol in Myristicae Semen roasted with flour.
