摘    要:目的建立日本血吸虫机械断尾尾蚴制备的童虫的体外培养,并用于相关的遗传操作。方法采用常规1 640培养基和添加激素等成分,建立日本血吸虫机械童虫的体外培养,并采用电穿孔技术将外源的小RNA分子导入到日本血吸虫机械童虫。结果成功体外连续培养了14d的日本血吸虫机械童虫,并建立了电穿孔技术将小RNA分子导入到日本血吸虫机械童虫的方法。结论建立的日本血吸虫机械童虫体外培养方法和转染小RNA分子的技术为血吸虫基因功能研究奠定了基础。

Study on culture for genetic manipulation of schistosomula of Schistosoma japonicum

Abstract:Objective To establish in vitro cultivation of Schistosoma japonicum schistosomula transformed mechanically from cercariae,and its application in genetic manipulation.Methods The schistosomula were mechanically transformed from cercariae and cultured using RPMI 1640 medium supplemented with hormone,etc.The transfection of small RNAs to schistosomula of S.japonicum was carried out by electroporation.Results The schistosomula were successfully maintained for 14 days in vitro.Methods for transfection of small RNAs were established for genetic manipulation in S.japonicum.Conclusion The methods established for in vitro culture of schistosomules and technique for transfection of small RNAs will provide a basis for study on the gene function of schistosome.
