摘    要:目的对星状病毒进行核酸快速诊断并对其部分核酸序列进行测序分析。方法采用RT-PCR、基因测序技术对标本中的核酸进行检测分析。结果 RT-PCR结果表明标本为星状病毒阳性,对PCR扩增产物测序得到了409bp长度的ORF2部分基因序列,经Blast比对发现其为Ⅰ型血清型,与北京分离的一株星状病毒序列(GenBank号FJ755403.1)最为相似,仅有一个C→T的核苷酸变异。但编码的氨基酸并无改变,均为甘氨酸(Gly)。结论应用PCR技术扩增星状病毒特定片段可用于病毒的快速检测,同时对扩增产物测序还可对病毒进行分型,并了解其基因变异情况。

Detection and analysis of nucleic acids of an astrovirus in Dongguan City in 2010

Abstract:Objective To determine the nenome sequence of astrovirus nucleic acid for rapid diagnosis of astrovirus infection in dainrrhea children. Methods The stool samples were collected from diarrhea children and the sequnce of astrovirus nucleic acids was determined by using RT-PCR method. Results The astrovirus was detected and the 409bp sequnce belonged to part of ORF2 gene was detected after amplification by RT-PCR and identified to be typeⅠastrovirus and similar to the virus isolated from Beijing(GenBank number:FJ775403.1) by Blast.There was only one nucleotide difference(C→T),without difference at amino acid level. Conclusions PCR technology can be used for rapid detection of astrovirus infection and genotyping of this virus.
