护卵汤对GnRHa超排卵大鼠卵巢形态的影响

摘    要:目的观察护卵汤对GnRHa超排卵大鼠卵巢形态的影响,为护卵汤辅治ART提供相关理论依据。方法模仿人类辅助生殖中的长周期,对大鼠进行GnRHa垂体降调节后再用HMG等药物超排卵(模型组),同时随机给予不同剂量的护卵汤辅助治疗(分别为护卵汤低、中、高剂量组),以自然周期未使用任何药物的大鼠为正常对照(正常组)。在HCG日称体质量并计算各大鼠卵巢指数,光镜观察排卵前卵泡并计算优质卵泡率,电镜观察卵母细胞的超微结构。结果护卵汤中剂量组的卵巢指数明显高于模型组,差异有统计学意义(P<0.01);模型组和护卵汤各剂量组的卵巢指数均高于正常组,差异有统计学意义(P<0.05或<0.01);模型组的优质卵泡率明显低于正常组和护卵汤中、高剂量组,差异有统计学意义(P<0.05或<0.01);模型组较正常组卵母细胞凋亡及胞浆水肿明显,而护卵汤中剂量组较模型组卵母细胞超微结构改善,胞内脂滴明显增多,细胞凋亡减少。结论 1、GnRHa超排卵对大鼠的卵泡发育及卵子质量可能有不利影响;2、护卵汤能改善GnRHa超排卵大鼠的卵泡发育及卵子质量。

Abstract:Objective To observe the effects of Huluantang on the ovarian morphology in rats under GnRHa controlled ovarian hyperstimulation(COH), and to provide related basis for Huluantang in assisted reproductive technology (ART). Methods Accroding to the human long period of COH, the modeling rats were established by pretreated with GnRHa for pituitary downregulation and then treated with HMG. Then, these rats were randomly divided into ART Huluan- tang high-dose group, median-dose group and low-dose group. At the same time, a number of rats without treatment were selected as the normal group. On the day of HCG administration, the weight of the rats were weighed, and the ovary indexes were calculated, the ovarian morphology and follic- ular were observed under light microscopy and such as, the rate of high quality follicular were calculated, the uhrastructure of oocyte were observed by electron microscopy. Results The ovary index of the Huluantang median-dose group was higher than that of the modeling group (P〈 0.01), and the ovary index of modeling group, Huluantang three dose groups were higher than that of the normal group (P〈0.05 or P〈0.01). The rate of high quality follicular of modeling group was lower than that of normal group and Huluantang high-dose group and median-dose group(P〈0.05 or P〈0.01). Compared with normal group, there were oocyte apoptosis and cytoplasmic edema in the modeling group. Compared with modeling group, the oocyte uhrastrueture was improved, intra- cellular fat droplets was significantly increased, cell apoptosis was reduced. Conclusion 1.The Gn- RHa controlled ovarian hyperstimulation may have some adverse effects on follicular development and quality of oocytes. 2. Huluantang can improve the follicular development and quality of oocytes in rats under GnRHa controlled ovarian hyperstimulation.
