胎儿内耳膜迷路蛋白的提取及电泳分析

摘    要:使用含0.1%十二烷基磺酸钠(SDS),5%二巯基乙醇(β-ME)和10%甘油的提取液制备 胎儿内耳膜迷路蛋白,并采用十二烷基磺酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)技术分析其蛋白 组成。结果:胎儿内耳膜迷路蛋白 Mr为 136 000、68 000、62 000、55 000、50 000、43 000、32 000、 30 000等。结果为进一步分析,纯化内耳特异性抗原,为临床采用免疫印迹法检测抗内耳组织自身 抗体提供了可能。

Abstract:This paper reports distinct extraction procedures that employed solutions contatining 0.1% SDS, 5% B-ME and 10% glycerol and had led to the preparation of antigens from the inner ear. Followed by Using SDS-PAGE technique to analyse the membranous labyrinth proteins of human foetus, some main bands were found in electrophoresis. The result showed that the membranous labyrinth protein in the inner ears of human foetuses contained Mr 130 000, 68 000, 43 000, 62 000, 55 000, 50 000, 32 000, and 30 000,and that it was possible to analyze and purify the specific antigens in the inner ears which may be used to detect the autoantibodies in the inner ear tissue by adopting Western Blot Assay.
