维拉帕米对缺血再灌注心肌细胞BCL-2基因蛋白表达的影响

目的：观察维拉帕米对缺血再灌注心肌细胞BCL－2基因蛋白的影响。从分子水平探讨维拉帕米对心肌缺血再灌注损伤发挥心肌保护作用的可能机制。方法：建立家兔心肌缺血再灌注模型，根据心肌缺血再灌注的不同时限对33只家兔进行分组，采用免疫组织化学法检测同一剂量维拉帕米（0．2mg/kg静脉注射）对缺血再灌注不同时限心肌细胞BCL－2基因蛋白的表达情况。结果：与假手术组和生理盐水组比较，维拉帕米能显著上调缺血再灌注心肌细胞BCL－2基因蛋白的表达（P＜0．001）。结论：维拉帕米对缺血再灌注心肌的保护机制可能是通过其促进缺血再灌注心肌细胞BCL－2基因蛋白的表达来实现的。

The influence of verapamil on the proteinexpression of bcl-2 gens of ischemia-Reperfusion myocardial cell

Objective To investigate the myocardial protection of verapamil on myocardial ischemia-reperfusion injuiy in rabbits and the molecular mechanism of its myocardial protection by observing the protein expression of bcl-2 genes in rabbits. Methods The model of myocardial ischemia-reperfusion in rabbits were established ,33 rabbits were divided into groups according to the different time of myocardial ischemia-reperfusion and the protein expression of bcl-2 gene in rabbits in groups pretreated by verapamil (0.2mg/kg , i.v) were observed and estimated by immunohistochemical method. Results Compared with the control groups verapamil could significantly increase the protein expression of bcl-2 genes in ischemia -reperfusion myocardial cell (P<0.001).Conclusion The ischemia-reperfusion myocardial cell protection of verapamil may be realized by increasing the protein expression of bcl-2 genes.
