摘    要:目的建立HPLC测定大鼠血浆中钩藤碱浓度的方法。方法色谱柱为ZORBAX SB C18柱(150mm×4.6 mm,5 m)。以甲醇-水(80:20)(含0.0025 mol.L-1三乙胺,冰乙酸调pH至7.5)为流动相,检测波长为254 nm,流速为1.0 ml/min,柱温为30℃,进样量为20 l。结果钩藤碱与内源性杂质有较好分离度,钩藤碱在0.39~12.50g/ml浓度范围内呈良好线性关系;最低检测浓度为0.39 g/ml;回收率大于95%;日内和日间RSD均小于7%。结论此方法准确可靠,可用于测定钩藤碱的血药浓度。

Abstract:Objective To establish the method to determine the plasma level ofRhynchophylline (RHY) in rats by HPLC. Methods The plasma samples were chromatographed on ZORBAX SB C1s column (150mm×4.6 mm,5 m) with a mixture of methanol and water (80: 20) as the mobile phase including 0.0025 mol/L triethylamine through glacial acetic acid to adjust pH 7.5. The detector wavelength was 254 nm. The flow-rate was 1.0 ml/min and the column temperature was set at 30℃. The injection volume was 20 μl. Results The linearity was obtained within the range of RHY concentrations from 0.39 to 12.50 μg/ml. The minimal inspect level was 0.39 μg/ml. The average recovery was more than 95%. Both of the inter- and intraoday RSD were less than 7%. Conclusion This method is stable, simple, rapid and accurate and applicable for the determination of plasma RHY level.
