摘    要:目的建立扩增片段小于250bp的STR基因座D11S4951、D7S3062、GATA168F06、D10S1426的荧光标记复合扩增体系并进行法医学实用性研究。方法利用荧光标记引物和传统的复合扩增方法,建立一个新的、使用ABI-310毛细管电泳仪进行荧光标记复合PCR扩增的复合扩增体系,对该体系的灵敏度和准确度、基因座的种属特异性,组织同一性及可重复性进行研究,并使用该系统对西南地区人群进行遗传学特征调查。结果成功建立了由D11S4951、D7S3062、GATA168F06和D10S1426四个STR基因座组成的荧光标记复合扩增体系。本体系条件易于优化,成本低廉,效率较高,法医学应用性好。结论可以应用于个人识别和亲子鉴定,成为中国地区法医物证实践的良好补充。

Abstract:Objective The aim of this study was to establish and validate the forensic application of the fluorescent Multiplex STR system ofDIIS4951, D7S3062, GATA168F06 and D10Sl426 which shorter than 250bp. Methods By the way of traditional fluorescent multiplex methods using ABI-310 Genetic Analyzer, we set up a new fluorescent Multiplex STR sys- tem. We also validated the study of Forensic Science, which included sensitivity and accuracy, species specificity, tissular identity, repeatability and performance of typing forensic samples. And then using this new system we did the forensic ge- netics in the people of southwest China. Results The fluorescent multiplex system of D11 S4951, D7S3062, GATA168F06 and D 10S 1426 was successfully established. Moreover, it was easy to use, lower cost, higher performance and validation study of Forensic Science showed that this system could make successfully utilization of forensic cases with no especial mu- tation in the people of southwest China. Conclusion The multiplex system could be recommended to the forensic genetics practice such as personal identification and paternity test. And it also could be the favorable supplement of current forensic genetics practice.
