摘    要:目的通过研究锰暴露对血液和唾液中PARK2 mRNA表达的影响,探寻锰暴露的早期生物标志。方法选择锰铁合金厂三年以上工龄的炉前工人作为锰接触组,以铁厂工人为对照组。原子吸收法测各组血液和唾液中锰浓度,反转录-聚合酶链反应(RT-PCR)法测各组血液和唾液中PARK2表达。结果锰接触组工人血液和唾液中锰浓度均高于对照组,差异均有统计学意义(〈0.01);与对照组相比,PARK2在血液和唾液中表达分别下降了54%和57%,差异均有统计学意义(〈0.05)。结论唾液口腔脱落细胞中PARK2基因的表达变化有作为锰暴露的早期生物标志的可能。

Abstract:Objective To explore the possibility whether PARK2 may be served as early biomarker of manganese exposure. Methods The Mn-Fe alloy factory workers with manganese exposure experience of three years or above were recruited as the trail group, and the iron factory workers were selected as the control group. Atom Absorption Spectrum (AAS) was applied to analyze the concentration of Mn in blood and saliva, and the expression of PARK2 was detected by real-time RT-PCR. Results The manganese concentrations in the trail group both in blood and saliva were significantly higher than those in the control group (P 〈0.01 ); By contrast with the control group, the expression of PARK2 decreased both in blood (54%) and saliva (57%) (P 〈0.05 ). Conclusion The expression of PARK2 in buccal cells might serves as an early biomarker of manganese exposure.
