| 摘 要： | 目的:探讨人骨髓间充质干细胞(hMSCs)体外分离培养及鉴定。方法:成人骨髓取自手术中非血液系统疾病、非肿瘤及乙肝表面抗原阴性患者摘取的肋骨,采用贴壁筛选法进行体外扩增;用免疫组织化学法对培养的细胞进行鉴定。结果:体外培养的原代hMSCs9d融合,免疫组织化学染色CD44表达阳性。结论:贴壁筛选法在体外很容易分离培养和扩增hMSCs,可作为获得hMSCs的一种有效手段。

| Abstract: | Objective:To investigate the method of isolation,culture,and identification of human mesenchymal stem cells(hMSCs).Methods:Human bone marrow was obtained,and hMSCs were isolated and purified by using direct anchoring and identified by using immunohistochemical method.Results:Primary cultured hMSCs reached confluence within 9 days.hMSCs were positive for CD44.Conclusion:hMSCs can be isolated,cultured,and expanded easily by using direct anchoring,which can be used as an effective method for culturing hMSCs. |
