肾透明细胞癌组织IGF-1和IGF-1R的表达及其临床意义

摘    要:目的:探讨胰岛素样生长因子(IGF)-1及IGF-1受体(IGF—1R)在肾透明细胞癌组织的表达及其临床意义。方法:应用实时荧光定量PCR法检测IGF-1及IGF-1R基因在肾透明细胞癌组织和相应癌旁组织中的表达水平,并分析其与临床病理指标的关系。结果:IGF-1mRNA和IGF-1R mRNA在肾透明细胞癌组织中的阳性表达率均为100%,在相应癌旁组织中均为80%。肾透明细胞癌组织中IGF-1mRNA和IGF-1R mRNA的相对表达强度分别为0.42±0.05、0.38±0.02,对应癌旁组织中分别为0.05±0.01和0.06±0.01,两组比较差异有统计学意义(P〈0.05)。肾透明细胞癌患者的AJCC分期越高,IGF-1、IGF-1R水平越高(P〈0.05)。结论:肾透明细胞癌组织和相应癌旁组织普遍表达IGF-1和IGF-1R基因,其中肾透明细胞癌存在IGF-1和IGF-1R高表达;IGF-1和IGF-1R与肾透明细胞癌密切相关,可有助于预测患者病情进展。

Abstract:Objective : To investigate the expression of insulin like growth factor-1 ( IGF-1 ) and IGF-1 receptor (IGF-1R) in renal clear cell carcinoma tissues and their clinical significance. Methods: Expression of IGF-1 and IGF-1R in renal clear cell carcinoma and pericancerous tissues were examined by real-time fluorescent quantitative polymerase chain reaction (RT-qPCR). and relationship between the expression of IGF-1/IGF-1R and the indexes of clinical pathology in renal clear cell carcinoma was further explored. Results: The expression percentages of IGF-1 and IGF-1R were both 100% in renal clear cell carcinoma, which were instead of 80% in the corresponding pericancerous tissues. The relative expression levels of IGF-1 and IGF-1R were 0. 42 ±0. 05 and 0. 38 ± 0. 02 in renal clear cell carcinoma, while those in the corresponding pericancerous tissues were 0. 05 ± 0. 01 and 0.06 ± 0. 01, respectively. The higher stage of renal clear cell carcinoma, the more expressions of IGF-1 and IGF- 1R were found (P 〈0.05). Conclusions: The expression of IGF-1 and IGF-1R was frequently observed in renal clear cell carcinoma and its corresponding pericancerous tissues. The expression of IGF-1 and IGF-1R was increased in renal clear cell carcinoma tissues, which were closely related to the progression of renal clear cell carcinoma and may be helpful for prognosis prediction of renal clear cell carcinoma.
