人参总皂甙对K562细胞增殖抑制与诱导分化的实验研究

摘    要:采用细胞体外培养,流式细胞术、形态学观察、组化与免疫细胞化学等方法,研究人参总皂甙(TSPG)对K_652细胞增殖与分化的影响。结果表明:TSPG对K_562细胞增殖具有明显的抑制作用,并能阻止K652细胞由G0/G1期向G2/M+S期过渡:经200μg/ml的TSPG诱导后,K562细胞形态趋向成熟分化;联苯胺染色、糖原染色和HIR2.CDw41阳性表达细胞数显著提高,且阳性强度增加。推测TSPG是能抑制白血病细胞增殖并诱导其分化的天然诱导剂,但对其机理有待进一步探讨。

Abstract:By using cell culture ex.vivo, flow cytometry (FCM), morphological observation, cytochemistry and immunocytochemistry, the effects of Total Saponins of Panax Ginseng (TSPG) on proliferation and differentiation of K562 cells were studied. The results indicated that TBPG. could markedly inhibit the proliferation of K562 cells , block K562 cells entering the active proliferative phase (G2/M S), and promote K562 cells to differentiate toward the mature blood cell. Our experiment also showed that after induced by TSPG, the ratio of positive K562 cells of expressing HIR2 CDw4t, glycogen, benzidine stain is obviously higher than that of control,Combining with our previous reasearcb works, we infer that TSPG may be one of the natural inducers which can not only enhance normal hematopoiesis, but also inhibit proliferation of such cancer cells as leukemia cells, and induce differentiation of these cells. The mechanism should be continually studied.
