苦参提取液诱导K562细胞分化的研究

摘    要:利用体外培养技术,以K562细胞为实验模型,通过K562细胞的生长曲线、细胞形态、乳酸脱氢酶活性、血红蛋白测定等,观察了苦参提取液对K562细胞的诱导分化作用。以MTT法结合细胞形态探讨苦参提取液作用K562细胞的有效分化浓度及有效作用时间,研究结果表明:12mg/ml浓度的苦参提取液作用K562细胞二天,从形态到代谢都发生了变化。LDH活性明显降低;血红蛋白生成;细胞增殖明显受到抑制。电镜形态提示,细胞向似红细胞系、粒细胞系及单核细胞系分化。

Abstract:Using culture assay in Vitro, K562 cell strain as anexperimental model, We observed the effect of sophoa extract on induced differentiation of K562 cell strain by cell growth curve, cell morphology, Latic dehydrogenase activity assay and hemoglobin determination. The effective concentration and time by MTT assay combined with cell morphology was studied. Results showed that K562 cell bad been acted on by sophoa extract 12mg/ml for two days. Its morphology and metabolism were changed. Their LDH activity lowered obviously; the Hb was produced by these cells; the proliferation of the cell was suppressed. Electron microscopy Showed: the cells differentiate toward the primitive cell with erythrocyte , granulocyte and monocyte lineages
