木犀草素对早期糖尿病大鼠心肌组织中醛糖还原酶、NOS系统及Na+-K+-ATPase的影响

摘    要:目的探讨木犀草素(luteolin)对糖尿病(diabetes mellitus,DM)大鼠心肌病(diabetic cardiomyopathy,DC)防治作用的机制。方法应用链脲佐菌素(streptozotocin,STZ)左腹腔注射诱发糖尿病,以木犀草素灌胃治疗,连续用药2个月。大鼠分为正常对照组、糖尿病模型组和木犀草素治疗组。分别于30d和60d测定各组大鼠心肌匀浆中的NO含量、NOS活性、Na^+-K^+-ATPase及醛糖还原酶(aldose reductase,AR)含量。结果DM大鼠心肌匀浆中的NO、NOS水平及Na^+-K^+-ATPase较正常组显著降低(P〈0.05),而AR较正常组显著升高(P〈0.05)。经木犀草素治疗后心肌匀浆中NO、NOS水平及Na^+-K^+-ATPase明显增加,AR降低。结论木犀草素能逆转心肌组织中AR、NOS及Na^+-K^+-ATPase活性,对治疗早期DM大鼠心肌病有一定的效果。

Abstract:Objective To investigate the protective effects of luteolin on cardiomyopathy of early diabetes rats.Methods SD rats were divided into normal group,diabetes mellitus(DM)group and luteolin group.Model rats were injected into intraperitoneally with streptozotocin(STZ).The levels of NO,aldose reductase(AR),Na -K -ATPase,and NOS in cardiac muscles of all rats were detected at 30 d and 60 d.Results Compared with normal group,the levels of NO,NOS and Na -K -ATPase in cardiac muscles decreased significantly in DM group(P<0.05),but the level of AR increased significantly(P<0.05).Compared with DM group,the level of AR in cardiac muscles of DM rats in luteolin group was lower,but the levels of NOS and Na -K -ATPase were higher.Conclusion Luteolin may play a role in cardiomyopathy of early DM rats by reversing the activities of AR,NOS and Na -K -ATPase.
