何首乌不同提取物对雌二醇致肾阳虚小鼠的影响

摘    要:目的观察何首乌四种提取物对雌二醇致肾阳虚模型小鼠的影响,探讨其"甘温,入肝、肾经"药性的现代科学内涵。方法取ICR雄性小鼠84只,随机分为正常对照组,模型对照组,桂附地黄丸组,何首乌水提取物组、正丁醇提取物组、乙酸乙酯提取物组和石油醚提取物组,共7组,每组12只。采用腹腔注射苯甲酸雌二醇造成小鼠肾阳虚模型,对小鼠体征、肛温、抓力、游泳时间、自主活动次数、血清肌酐(Cr)及尿素(UR)、血液学指标、睾丸和精囊腺指数进行测定。结果何首乌四种提取物均能改善小鼠体征、抓力、自主活动次数、RBC、Hb、PLT、WBC、LY、NE;何首乌水提取物能提高肾阳虚小鼠游泳时间、MO,降低Cr水平;乙酸乙酯提取物能提高游泳时间、睾丸指数、MCV,降低Cr水平;正丁醇提取物能提高MCV、MO,降低UR水平;石油醚提取物能提高游泳时间、MCV,降低UR水平。结论何首乌的药效可作为改善肾阳虚证腰膝酸软、精神萎靡、阳虚水泛,以及肾精亏虚、肝气虚弱所致血液亏虚的基础,并作为其甘温、入肝、肾经药性现代科学内涵的组成部分。何首乌四种提取物药效相近,其中水提取物与乙酸乙酯提取物、正丁醇提取物与石油醚提取物的药效更为接近。

Abstract:Objective To observe the effects of four extracts from Radix Polygoni Multiflori on kidney-yang deficiency mice induced by estradiol,and to reveal the modern scientific connotation of its properties of sweet-flavor,warm-nature,and meridian tropism in liver and kidney. Methods Eighty-four healthy adult male ICR mice were divided into seven groups randomly:normal control group,model control group,positive control group (Guifu Dihuang Wan ),and four groups of Radix Polygoni Multiflori (water extract,n-butanol extract,acetoacetate extract and petroleum benzine extract),twelve mice in each group. Kidney-yang deficiency mice model was established with intraperitoneal injection of estradiol benzoate.The effects were observed by measuring physicalsigns,rectal temperature,grip strength,swimming time,locomotor activity,serum creatinine and urea levels,haematological indexes,testis index and seminal vesicle index. Results All of the four extracts from Radix Polygoni Multiflori could markedly improve the physical signs of kidney-yang deficiency mice,heighten grip strength and locomotor activity,increase RBC,Hb,PLT,WBC,LY,NE,in blood. The water extract could obviously prolong swimming time,increase MO in blood,and decrease the serum level of Cr. The acetoacetate extract could prolong swimming time,increase testis index and mean cell volume (MCV) in blood,decrease the serum level of Cr. The n-butanol extract could obviously heighten MCV,MO in blood,decrease the level of UR in serum. The petroleum benzine extract could prolong swimming time,enhance MCV in blood,decrease the serum level of UR. Conclusion The above pharmaceutical actions of Radix Polygoni Multiflori could be the basis of its therapeutic mechanism on relieving symptoms of kidney-yang deficiency such as soreness of the waist and knees,lack of energy,hydroretention with asthenic yang,and on blood deficiency induced by deficiency of kidney-essence and deficiency of liver-Qi. Meanwhile,they could be parts of the modern scientific connotation of the properties theories of sweet-flavor,warm-nature,and meridian tropism in liver and kidney. The four extracts from Radix Polygoni Multiflori have similar effects. Of them,the effect of water extract is similar to that of acetoacetate extract,while the effect of n-butanol extract is similar to that of petroleum benzine extract,which will offer some enlighten for searching the common effective substance groups of Radix Polygoni Multiflori,and provide some basis for clinical application and product development.
